2Tonoplast-associated calcium signaling regulates manganese homeostasis in Arabidopsis显示文摘Manganese(Mn)is an essential micronutrient in plants.However,excessive Mn absorption in acidic and waterlogged soils can lead to Mn toxicity.Despite their essential roles in Mn homeostasis,transcriptional and post-transcriptional modifications of Mn transporters remain poorly understood.Here,we demonstrated that high-Mn stress induces an obvious Ca^(2+) signature in Arabidopsis.We identified four calcium-dependent protein kinases,CPK4/5/6/11,that interact with the tonoplast-localized Mn and iron(Fe)transporter MTP8 in vitro and in vivo.The cpk4/5/6/11 quadruple mutant displayed a dramatic high-Mn-sensitive phenotype similar to that of the mtp8 mutant.CPKs phosphorylated the N-terminal domain of MTP8 primarily at the Ser31 and Ser32 residues.Transport assays combined with multiple physiological experiments on phospho-dead variant MTP8^(S31/32A)and phospho-mimetic variant MTP8^(S31/32D)plants under different Mn and Fe conditions suggested that Ser31 and Ser32 are crucial for MTP8 function.In addition,genetic analysis showed that CPKs functioned upstream of MTP8.In summary,we identified a tonoplast-associated calcium signaling cascade that orchestrates Mn homeostasis and links Mn toxicity,Ca^(2+) signaling,and Mn transporters.These findings provide new insight into Mn homeostasis mechanisms and Ca^(2+) signaling pathways in plants,providing potential targets for engineering heavy metal toxicity-tolerant plants.Zhenqian Zhang Dali Fu Zhihui Sun Chuanfeng Ju Cuicui Miao Zhangqing Wang Dixiang Xie Liang Ma Zhizhong Gong Cun Wang 2021Molecular Plant2021,14,5:2

8Calcium-dependent protein kinases CPK21 and CPK23 phosphorylate and activate the iron-regulated transporter IRT1 to regulate iron deficiency in Arabidopsis显示文摘Iron(Fe)is an essential micronutrient for all organisms.Fe availability in the soil is usually much lower than that required for plant growth,and Fe deficiencies seriously restrict crop growth and yield.Calcium(Ca2+)is a second messenger in all eukaryotes;however,it remains largely unknown how Ca2+regulates Fe deficiency.In this study,mutations in CPK21 and CPK23,which are two highly homologous calcium-dependent protein kinases,conferredimpaired growth and rootdevelopment under Fe-deficient conditions,whereas constitutively active CPK21 and CPK23 enhanced plant tolerance to Fe-deficient conditions.Furthermore,we found that CPK21 and CPK23 interacted with and phosphorylated the Fe transporter IRONREGULATED TRANSPORTER1(IRT1)at the Ser149 residue.Biochemical analyses and complementation of Fe transport in yeast and plants indicated that IRT1 Ser149 is critical for IRT1 transport activity.Taken together,these findings suggest that the CPK21/23-IRT1 signaling pathway is critical for Fe homeostasis in plants and provides targets for improving Fe-deficient environments and breeding crops resistant to Fe-deficient conditions.Zhangqing Wang Yanting Zhang Yisong Liu Dali Fu Zhang You Panpan Huang Huiling Gao Zhenqian Zhang Cun Wang 2023Science China(Life Sciences)2023,66,11:1

11Modulating Pd e_(g) orbital occupancy in Pd-Au metallic aerogels for efficient carbon dioxide reduction显示文摘The electronic structure of electrocatalysts plays a critical role in energy conversion,whereas for an efficient catalyst,it is challenging to modulate the orbitals.Herein,we present a new strategy to modulate the e_(g) orbital occupancy of Pd by constructing composition-controllable Pd-Au metallic aerogels(MAs),optimizing the d-band center of Pd to achieve excellent performance for electrochemical carbon dioxide reduction reaction(CO_(2)RR).Specifically,Pd_(1)Au_(2) MAs achieve almost 100% Faraday efficiency(FE) of CO in the range of-0.40 to-0.80 V vs.reversible hydrogen electrode(RHE),as well as the long-term stability,being one of the best Pd-based materials for CO_(2)RR.The X-ray photoelectron spectroscopy(XPS) results and density functional theory(DFT) calculations demonstrate that the introduction of Au modulates the Pd e_(g) orbital occupancy,which significantly weakens *CO adsorption on Pd,reduces the CO_(2)RR energy barrier and consequently improves the electrocatalytic activity and stability for long-term applications.Our work highlights a new strategy for designing efficient electrocatalysts for CO_(2)RR and beyond.Yao Chen Juan Wang Tingjie Mao Cun Chen Hanjun Li Honggang Huang Hui Fu Feili Lai Jiadong Chen Nan Zhang Tianxi Liu 2023Journal of Energy Chemistry2023,,12:0

13Ca^(2+)-dependent successive phosphorylation of vacuolar transporter MTP8 by CBL2/3-CIPK3/9/26 and CPK5 is critical for manganese homeostasis in Arabidopsis显示文摘Manganese(Mn)is an essential micronutrient for all living organisms.However,excess Mn supply that can occur in acid or waterlogged soils has toxic effects on plant physiology and development.Although a variety of Mn transporter families have been characterized,we have only a rudimentary understanding of how these transporters are regulated to uphold and adjust Mn homeostasis in plants.Here,we demonstrate that two calcineurin-B-like proteins,CBL2/3,and their interacting kinases,CIPK3/9/26,are key regulators of plant Mn homeostasis.Arabidopsis mutants lacking CBL2 and 3 or their interacting protein kinases CIPK3/9/26 exhibit remarkably high Mn tolerance.Intriguingly,CIPK3/9/26 interact with and phosphorylate the tonoplast-localized Mn and iron(Fe)transporter MTP8 primarily at Ser35,which is conserved among MTP8 proteins from various species.Mn transport complementation assays in yeast combined with multiple physiological assays indicate that CBL-CIPK-mediated phosphorylation of MTP8 negatively regulates its transport activity from the cytoplasm to the vacuole.Moreover,we show that sequential phosphorylation of MTP8,initially at Ser31/32 by the calcium-dependent protein kinase CPK5 and subsequently at Ser35 by CIPK26,provides an activation/deactivation fine-tuning mechanism for differential regulation of Mn transport.Collectively,our findings define a two-tiered calcium-controlled mechanism for dynamic regulation of Mn homeostasis under conditions of fluctuating Mn supply.Chuanfeng Ju Zhenqian Zhang Jinping Deng Cuicui Miao Zhangqing Wang Lukas Wallrad Laiba Javed Dali Fu Ting Zhang Jörg Kudla Zhizhong Gong Cun Wang 2022Molecular Plant2022,15,3:0
14d-d Orbital coupling induced by crystal-phase engineering assists acetonitrile electroreduction to ethylamine显示文摘The d-d orbital coupling induced by crystal-phase engineering can effectively adjust the electronic structure of electrocatalysts,thus showing significant catalytic performance,while it has been rarely explored in electrochemical acetonitrile reduction reaction(ARR)to date.Herein,we successfully realize the structural transformation of Pd Cu metallic aerogels(MAs)from face-centered cubic(FCC)to body-centered cubic(BCC)through annealing treatment.Specifically,the BCC Pd Cu MAs exhibit excellent ARR performance with high ethylamine selectivity of 90.91%,Faradaic efficiency of 88.60%,yield rate of 316.0 mmol h^(-1)g^(-1)_(Pd+Cu)and long-term stability for consecutive electrolysis within 20 h at-0.55 V vs.reversible hydrogen electrode,outperforming than those of FCC Pd Cu MAs.Under the membrane electrode assembly system,BCC Pd Cu MAs also demonstrate excellent ethylamine yield rate of 389.5 mmol h^(-1)g^(-1)_(Pd+Cu).Density functional theory calculation reveals that the d-d orbital coupling in BCC Pd Cu MAs results in an evident correlation effect for the interaction of Pd and Cu sites,which boosts up the Cu sites electronic activities to enhance ARR performance.Our work opens a new route to develop efficient ARR electrocatalysts from the perspective of crystalline structure transformation.Honggang Huang Yao Chen Hui Fu Cun Chen Hanjun Li Zhe Zhang Feili Lai Shuxing Bai Nan Zhang Tianxi Liu 2024Journal of Energy Chemistry2024,89,2:0
15Preoperative serum cortisone levels are associated with cognition in preschool‑aged children with tetralogy of Fallot after corrective surgery:new evidence from human populations and mice显示文摘Background Tetralogy of Fallot(TOF)is the most common cyanotic congenital heart disease.Children with TOF would be confronted with neurological impairment across their lifetime.Our study aimed to identify the risk factors for cerebral morphology changes and cognition in postoperative preschool-aged children with TOF.Methods We used mass spectrometry(MS)technology to assess the levels of serum metabolites,Wechsler preschool and primary scale of intelligence-Fourth edition(WPPSI-Ⅳ)index scores to evaluate neurodevelopmental levels and multimodal magnetic resonance imaging(MRI)to detect cortical morphological changes.Results Multiple linear regression showed that preoperative levels of serum cortisone were positively correlated with the gyrification index of the left inferior parietal gyrus in children with TOF and negatively related to their lower visual spaces index and nonverbal index.Meanwhile,preoperative SpO_(2) was negatively correlated with levels of serum cortisone after adjusting for all covariates.Furthermore,after intervening levels of cortisone in chronic hypoxic model mice,total brain volumes were reduced at both postnatal(P)11.5 and P30 days.Conclusions Our results suggest that preoperative serum cortisone levels could be used as a biomarker of neurodevelopmental impairment in children with TOF.Our study findings emphasized that preoperative levels of cortisone could influence cerebral development and cognition abilities in children with TOF.Si-Yu Ma Yu-Ting Liu Yue-Shuang Cun Qiang Wang Ming-Cui Fu Ke-De Wu Xin-Yu Cai Shu-Ting Cheng·Nishant Patel Min Da Liang Hu Zhuoga Deqin Xue-Jun Kang Ming Yang Xu-Ming Mo 2024World Journal of Pediatrics2024,20,2:0

17Genotyping of strains of Yersinia pestis from Marmota baibacina-Spermophilus undulates Plague Focus of Tianshan Mountains in China显示文摘The airn of this study is to carry out genotyping of 61 Y.pestis strains isolated from Marmota baibacina-Spermophilus undulates Plague Focus of Tianshan Mountains in China. Primer pairs targeting the 22 DFRs were designed for detecting the genotypes of 61 strains. As a result, 61 strains of Y.pestis were divided into four genotypes 1, 2, 3, and 4. Genotypes 1, 2 and 3 were found from west part in Northern Tianshan Mountains Plague Focus, but type 1 was only from Nileke county. The type of strains from Aheqi was different from those of Atushi counties in Southern Tianshan Mountains and similar to strains in Northern Tianshan Mountains Plague Focus. The type 4 distributed over Atushi county, which was identical with that of strains from Marmota caudae Plague Focus of the Pamiri Plateau. It is concluded that geonotyping is identical with ecotyping made by Shuli Ji. 19Effects of Nb Addition on Charpy Impact Properties of TiVTa Refractory High‑Entropy Alloy显示文摘In the present study,the effects of Nb addition on Charpy impact properties of TiVTa refractory high-entropy alloy with high strength-ductility trade-off were systematically studied by using the instrumented Charpy impact testing machine.The experimental results showed that the impact toughness was remarkably improved by Nb addition in TiVTa to form TiVTaNb alloy.The crack initiation energy and propagation energy of TiVTaNb were 67.3%and 24.9%higher than that of TiVTa,indicating that Nb addition simultaneously reinforced the resistance to crack initiation and propagation.The impact fracture of TiVTaNb exhibited larger bending degree of shear lips,deeper dimples and more secondary cracks which effectively dissipated more impact energy.The deformation mechanism of TiVTa alloy was dominated by dislocation activities.While in TiVTaNb,the deformation mechanism was synergized by dislocation activities and deformation twinning,which were the main contributors for the improved impact properties and the stronger crack resistance of TiVTaNb alloy under impact loading.Xue Yin Yan‑Kun Dou Xin‑Fu He Ke Jin Cheng‑Long Wang Ya‑Guang Dong Cun‑Yong Wang Yun‑Fei Xue Wen Yang 2023Acta Metallurgica Sinica(English Letters)2023,36,3:0
