1Synthesis of selective PAK4 inhibitors for lung metastasis of lung cancer and melanoma cells显示文摘The p21 activated kinase 4(PAK4) is serine/threonine protein kinase that is critical for cancer progression.Guided by X-ray crystallography and structure-based optimization,we report a novel subseries of C-3-substituted 6-ethynyl-1 H-indole derivatives that display high potential and specificity towards group Ⅱ PAKs.Among these inhibitors,compound 55 exhibited excellent inhibitory activity and kinase selectivity,displayed superior anti-migratory and anti-invasive properties against the lung cancer cell line A549 and the melanoma cell line B16.Compound 55 exhibited potent in vivo antitumor metastatic efficacy,with over 80% and 90% inhibition of lung metastasis in A549 or B16-BL6 lung metastasis models,respectively.Further mechanistic studies demonstrated that compound 55 mitigated TGF-β1-induced epithelial-mesenchymal transition(EMT).Peilu Song Fan Zhao Dahong Li Jiqiang Qu Miao Yao Yuan Su Hanxun Wang Miaomiao Zhou Yujie Wang Yinli Gao Feng Li Dongmei Zhao Fengjiao Zhang Yu Rao Mingyu Xia Haitao Li Jian Wang Maosheng Cheng 2022Acta Pharmaceutica Sinica B2022,12,6:2

3Tum or Necrosis Factor Expression in ArterialWallsof Diabetic Rats显示文摘Immunohistochemistry was used to detect tumor necrosis factor (TNF α) expression in arterial wall of diabetic rats. It was found that endothelial cells were swollen and markedly proliferative in these vessels and accordingly TNF α showed strong positive immunohistochemical reaction in endothelial cells or extracellular intimal matrix of such vessels, which might be caused by the expression and release of TNF α from monocytes and arterial wall cells stimulated by AGEs. 4Glacier changes in the Qaidam Basin from 1977 to 2018显示文摘Based on Landsat MSS/TM/OLI remote sensing images,glaciers vector data in the Qaidam Basin were extracted for 1977,2002,and 2018,and their spatial-temporal variations were analyzed.Results show that there were 2,050 glaciers covering an area of 1,693.54±40.96 km2 and having an ice volume of 108.65±2.43 km^3 in the Qaidam Basin in 2018.Glaciers with areas<1.0 km2 accounted for the largest number,while glaciers with areas of 1.0−5.0 km2 accounted for the most glacierized area.In the past 50 years,the number of glaciers decreased by 177,and the glacier area and volume reduced by 338.08 km2(−8.12 km^2/a)and 19.92 km3(−0.48 km^3/a),respectively.Retreat altitudes of glaciers were concentrated at 4,900−5,600 m,4,700−5,200 m,and 5,000−5,600 m and reduced areas accounted for 95.53%,77.80%,and 69.19%in the Kunlun,Qilian,and Altun mountains,respectively.The area of north-oriented glaciers decreased the most(−125.43 km^2),but the west-and east-oriented glaciers retreated at the fastest rate(i.e.,−27.11%and−27.10%).All glaciers showed a decreasing trend in sub-regions of the Qaidam Basin from 1977 to 2018.The decreasing trend was accelerated gradually from northwest to southeast in the northern part of the basin,while glacier change was the smallest in the middle section and gradually accelerated towards both ends of the basin's southern part.The temperature had continued to rise,and the precipitation had increased slowly in the Qaidam Basin during the past 50 years.The continuous rise in air temperature was the main reason for the retreat of glaciers.SuGang Zhou XiaoJun Yao Yuan Zhang DaHong Zhang Juan Liu HongYu Duan 2020Research in Cold and Arid Regions2020,12,6:1

8Targeting the Warburg effect:A revisited perspective from molecular mechanisms to traditional and innovative therapeutic strategies in cancer显示文摘Cancer reprogramming is an important facilitator of cancer development and survival,with tumor cells exhibiting a preference for aerobic glycolysis beyond oxidative phosphorylation,even under sufficient oxygen supply condition.This metabolic alteration,known as the Warburg effect,serves as a significant indicator of malignant tumor transformation.The Warburg effect primarily impacts cancer occurrence by influencing the aerobic glycolysis pathway in cancer cells.Key enzymes involved in this process include glucose transporters(GLUTs),HKs,PFKs,LDHs,and PKM2.Moreover,the expression of transcriptional regulatory factors and proteins,such as FOXM1,p53,NF-κB,HIF1a,and c-Myc,can also influence cancer progression.Furthermore,lncRNAs,miRNAs,and circular RNAs play a vital role in directly regulating the Warburg effect.Additionally,gene mutations,tumor microenvironment remodeling,and immune system interactions are closely associated with the Warburg effect.Notably,the development of drugs targeting the Warburg effect has exhibited promising potential in tumor treatment.This comprehensive review presents novel directions and approaches for the early diagnosis and treatment of cancer patients by conducting in-depth research and summarizing the bright prospects of targeting the Warburg effect in cancer.Minru Liao Dahong Yao Lifeng Wu Chaodan Luo Zhiwen Wang Jin Zhang Bo Liu 2024Acta Pharmaceutica Sinica B2024,14,3:0
9Twelve-week study of moxa smoke: occupational exposure in female rats显示文摘OBJECTIVE: To assess the toxicity of moxa smoke in rats.METHODS: Forty-eight female Wister rats were randomly divided into 4 groups(n = 12/group) to simulate moxa smoke exposure in Chinese medicine clinics(CMCs): the control group, and three moxa-smoke exposed groups of PM10 mass concentrations 3-5, 7-9 and 27-30 mg/m^3, respectively.These concentrations were 1 ×, 2-3 ×, and 7-9 ×fold the concentrations found in CMCs. Exposures continued for 12 weeks(200 min/d, 5 d/week).RESULTS: No deaths were noted. After the exposure, the body weights, ratios of organ weight to body weight, urinary parameters, hematological parameters, clinical chemistry parameters and microscopic examinations revealed no obvious toxicity.CONCLUSION: Moxa smoke did not induce toxic effects in female rats in the study. 10β-Carboline alkaloids from the roots of Peganum harmala L.显示文摘This study reports the isolation of four newβ-carboline alkaloids(1−4)and six previously identified alkaloids(5−10)from the roots of Peganum harmala L.Among these compounds,1 and 2 were characterized as rareβ-carboline-quinazoline dimers exhibiting axial chirality.Compound 3 possessed a unique 6/5/6/7 tetracyclic ring system with an azepine ring,and compound 4 was a novel annomontineβ-carboline.The structures of these compounds were elucidated by spectroscopic data and quantum mechanical calculations.The biosynthetic pathways of 1−3 were proposed.Additionally,the cytotoxicity of some isolates against four cancer cell lines(HL-60,A549,MDA-MB-231,and DU145)was evaluated.Notably,compound 4 exhibited significant cytotoxicity against HL-60,A549,and DU145 cells with IC_(50) values of 12.39,12.80,and 30.65μmol·L^(−1),respectively.Furthermore,compound 2 demonstrated selective cytotoxicity against HL-60 cells with an IC_(50) value of 17.32μmol·L^(−1).LI Shengge ZHANG Qin WANG Yuetong LIN Bin LI Dahong HUA Huiming HU Xu 2024Chinese Journal of Natural Medicines2024,22,2:0
