| 1 | Detailed resume of RNA m^(6)A demethylases显示文摘N6-Methyladenosine(m^(6)A) is the most abundant internal modification in eukaryotic mRNA,playing critical role in various bioprocesses. Like other epigenetic modifications, m^(6)A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m^(6)A demethylases have been reported, fat mass and obesity-associated protein(FTO)and alkylation protein AlkB homolog 5(ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. | 3 | Photocatalytic decomposition of acrylonitrile with N–F codoped TiO_2/SiO_2 under simulant solar light irradiation显示文摘The solid acid catalyst, N–F codoped Ti O2/Si O2 composite oxide was prepared by a sol–gel method using NH4 F as nitrogen and fluorine source. The prepared materials were characterized by X-ray diffraction(XRD), scanning electron microscopy(SEM), X-ray photoelectron spectroscopy(XPS), UV–Visible diffuse reflectance spectroscopy(UV–Vis),ammonia adsorption and temperature-programmed desorption(NH3-TPD), in situ Fourier transform infrared spectroscopy(FT-IR) and N2 physical adsorption isotherm. The photocatalytic activity of the catalyst for acrylonitrile degradation was investigated under simulant solar irradiation. The results showed that strong Lewis and Brnsted acid sites appear on the surface of the sample after N–F doping. Systematic investigation showed that the highest photocatalytic activity for acrylonitrile degradation was obtained for samples calcined at 450°C with molar ratio(NH4F to Ti) of 0.8. The degradation ratio of 71.5% was achieved with the prepared catalyst after 6-min irradiation, demonstrating the effectiveness of photocatalytic degradation of acrylonitrile with N–F codoped Ti O2/Si O2 composite oxide. | 4 | Efficacy on rabbits with arrhythmia: needling acupoint of Neiguan(PC6) at shallow or deep depth, and retaining needles for 10, 20, or 30 min显示文摘OBJECTIVE:To compare the effects of Neiguan(PC6)acupuncture at different depths and retention time on arrhythmia duration,myocardial tissue morphology,mRNA expression level of L-type calcium channelα1C subunit and Ca^(2+)-Mg^(2+)-At Pase activity in tachirrhythmia model of rabbits.METHODS:The tachyarrhythmia model was made by intravenous injection of barium chloride into the ears of rabbits.A total of 56 healthy adult male New Zealand big-eared white rabbits,apply the random number table method,divided into normal control group(group A),model group(group B),shallow needling Neiguan(PC6)10 min group(group C),shallow needling Neiguan(PC6)20 min group(group D),shallow needling Neiguan(PC6)30 min group(group E),deep needling Neiguan(PC6)10 min group(group F),deep needling Neiguan(PC6)20 min group(group G),deep needling Neiguan(PC6)30 min group(group H),7 animals in each group.Electrocardiograms were used to collect the duration of arrhythmia;hematoxylin-eosin staining method was performed on myocardial tissue,RT-PCR tested the expression ofα1C subunit mRNA,and the activity of Ca^(2+)-Mg^(2+)-ATPase were quantified by phosphorus determination method.RESULTS:The duration of arrhythmia in each acupuncture treatment group was shortened to varying degrees.Compare to the model group,the tissue damage from barium chloride inducing was improved in the acupuncture group.Compared to the model group,except for group E,most treatment groups had varying degrees of improvement with significantly down-regulated L-type calcium channelα1C subunit mRNA expressions level and increased Ca^(2+)-Mg^(2+)-ATPase activity.CONCLUSIONS:The effect of acupuncture at Neiguan(PC6)with different depths and retention time can reduce the duration of arrhythmia induced by barium chloride relatively,improve the induced pathological changes,down regulate L-type calcium channelα1C subunit mRNA expressions level and increase Ca^(2+)-Mg^(2+)-ATPase activity.Both the shallow and deep tissues of Neiguan(PC6)may be involved in transmitting acupuncture information.There is an optimal induction period for shallow needling at Neiguan(PC6)to reach the best therapeutic effect. | GU Chunling CHENG Kai YANG Shouqin RAN Shaofeng TAI Hengyap GAO Xiaofeng PANG Dandan | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 1 |
| 5 | PHF6 functions as a tumor suppressor by recruiting methyltransferase SUV39H1 to nucleolar region and offers a novel therapeutic target for PHF6-muntant leukemia显示文摘Mutations in the plant homeodomain-like finger protein 6(PHF6)gene are strongly associated with acute myeloid(AML)and T-cell acute lymphoblastic leukemia(T-ALL).In this study,we demonstrated that PHF6 can bind to H3K9me3 and H3K27me1 on the nucleolar chromatin and recruit histone methyltransferase SUV39H1 to the rDNA locus.The deletion of PHF6 caused a decrease in the recruitment of SUV39H1 to rDNA gene loci,resulting in a reduction in the level of H3K9me3 and the promotion of rDNA transcription.The knockdown of either SUV39H1 or PHF6 significantly attenuated the effects of increase in H3K9me3 and suppressed the transcription of rDNA induced by the overexpression of the other interacting partner,thereby establishing an interdependent relationship between PHF6 and SUV39H1 in their control of rRNA transcription.The PHF6 clinical mutants significantly impaired the ability to bind and recruit SUV39H1 to the rDNA loci,resulting in an increase in rDNA transcription activity,the proliferation of in vitro leukemia cells,and the growth of in vivo mouse xenografts.Importantly,significantly elevated levels of pre-rRNA were observed in clinical AML patients who possessed a mutated version of PHF6.The specific rDNA transcription inhibitor CX5461 significantly reduced the resistance of U937 AML cells deficient in PHF6 to cytarabine,the drug that is most commonly used to treat AML.Collectively,we revealed a novel molecular mechanism by which PHF6 recruits methyltransferase SUV39H1 to the nucleolar region in leukemia and provided a potential therapeutic target for PHF6-mutant leukemia. | Hsiang-i Tsai Yanping Wu Rui Huang Dandan Su Yingyi Wu Xiaoyan Liu Linglu Wang Zhanxue Xu Yuxin Pang Chong Sun Chao He Fan Shu Haitao Zhu Dongqing Wang Fang Cheng Laiqiang Huang Hongbo Chen | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 1 |

| 9 | Effect of electroacupuncture on inflammatory signal expression in local tissues of rats with chronic pelvic pain syndrome based on purinergic 2X7 receptor/NOD-like receptor pyrin domaincontaining 3 signal pathway显示文摘OBJECTIVES: To study the expression of inflammatory signal in local prostate tissue of chronic pelvic pain syndrome(CPPS) rats by electroacupuncture(EA) of Guanyuan(CV4), Zhongji(CV3), Huiyang(BL35) and Sanyinjiao(SP6), and to explore the possible mechanism of anti-inflammatory and analgesic effects of EA. METHODS : A total of 36 Sprague-Dawley male rats were randomly divided into three groups: control, model and EA(n=12 rats/group). The CPPS model was made by injection of CFA into ventral lobes of the prostate(0.1 m L). Electric acupuncture apparatus was applied to stimulate Guanyuan(CV4), Zhongji(CV3), bilateral Huiyang(BL35) and Sanyinjiao(SP6) acupoints in EA group. The general condition of rats was observed and the prostate index(PI) was calculated. The thermal pain threshold was collected after each therapeutic course. Histopathological changes of the prostate tissue were examined by hematoxylin-eosin staining method. The expression levels of tumor necrosis factor α(TNF-α), interleukin-1β(IL-1β) and prostaglandin E2(PGE2) in prostatic homogenates were measured by enzyme linked immunosorbent assay(ELISA). Moreover, the expression levels of purinergic 2X7 receptor(P2X7R), NOD-like receptor pyrin domain-containing 3(NLRP3), caspase-1 and interleukin-18(IL-18) m RNA were quantified by quantitative real-time polymerase chain reaction. RESULTS: Compared with control group, the PI of rats increased, and the thermal pain threshold decreased significantly in model group. The morphological structure of prostate tissues of rats in model group was severely damaged with a large number of inflammatory cells infiltration. Additionally, the levels of TNF-α, IL-1β and PGE2 were higher, and the expressions of P2X7R, NLRP3, caspase-1 and IL-18 m RNA were higher than those in control group. After EA treatment, the PI was significantly decreased, the thermal pain threshold was significantly increased, and the tissue damage was significantly improved. The expressions of inflammatory cytokines were lower in EA group, and expression of P2X7R/NLRP3 pathway was down-regulated. CONCLUSION: The effect of EA at Guanyuan(CV4), Zhongji(CV3), Huiyang(BL35) and Sanyinjiao(SP6) can improve inflammation and pain symptoms of CPPS rats induced by Complete Freund’s adjuvant(CFA). | 11 | Topography guiding the accelerated and persistently directional cell migration induced by vaccinia virus显示文摘The rapid transmission of vaccinia virus(VACV)in vivo is thought to be closely related to the cell migration induced by it.Cell migration involved in dynamic changes of cell-substrate adhesion and actin cytoskeleton organization,which can influence by the micro/nano-scale topographic structures that cells are naturally exposed to via contact guidance.However,migration behaviors of VACV-infected cells exposed to topographic cues are still unknown.Herein,we designed an open chip with microgrooved poly(dimethyl siloxane)(PDMS)substrate to explore the topography roles in VACV-induced cell migration.Differed from the random cell migration observed in traditional scratch assay on planar substrate,VACV-infected cells had a tendency to persistently migrate along the axis parallel to microgroove with increased velocity.Moreover,infected cells exhibited a dominant elongated protrusion aligned to the micro-grating axis compare to the shorter lamella extended in any direction on smooth substrate.Interestingly,the Golgi complex preferred to relocate behind the nucleus confined within the micro-grating axis in majority of infected migratory cells.The directional polarization of cells embodied in protrusion formation and Golgi reorientation was responsible for the directionally persistent migration behaviors induced by VACV on microgrooved substrate.Infected cells response to substrate topography,causing the actin-filled stretched protrusion containing numerous virions and accelerated movement is likely to facilitate direct and rapid spread of VACV.This work opens a window for us to understand the migration behaviors of infected cells in vivo,and also provides a cue for revealing the relationship between virus-induced cell migration and virus rapid spread. | Cheng Wang Ji Wang Dandan Fu Qiong Yan Daiwen Pang Zhiling Zhang | 2020 | Chinese Chemical Letters2020,31,1: | 0 |
| 12 | Parallelism and Optimization of Numerical Ocean Forecasting Model显示文摘According to the characteristics of Chinese marginal seas, the Marginal Sea Model of China(MSMC) has been developed independently in China. Because the model requires long simulation time, as a routine forecasting model, the parallelism of MSMC becomes necessary to be introduced to improve the performance of it. However, some methods used in MSMC, such as Successive Over Relaxation(SOR) algorithm, are not suitable for parallelism. In this paper, methods are developedto solve the parallel problem of the SOR algorithm following the steps as below. First, based on a 3D computing grid system, an automatic data partition method is implemented to dynamically divide the computing grid according to computing resources. Next, based on the characteristics of the numerical forecasting model, a parallel method is designed to solve the parallel problem of the SOR algorithm. Lastly, a communication optimization method is provided to avoid the cost of communication. In the communication optimization method, the non-blocking communication of Message Passing Interface(MPI) is used to implement the parallelism of MSMC with complex physical equations, and the process of communication is overlapped with the computations for improving the performance of parallel MSMC.
