| 1 | Progress in the participation of Ca^(2+)-calmodulin in heat shock signal transduction显示文摘A novel heat shock(HS) signal transduction pathway in plants for the participation of Ca2+-calmodulin(CaM) in HS signal trans-duction was identified.HS induces a rapid increase in intracellular free calcium ion levels([Ca2+]i),and the involvement of phospholipase Cinositol 1,4,5-trisphosphate is one of the factors leading to elevation in [Ca2+]i induced by HS.HS also increases the expression of the CaM gene and the accumulation of the CaM protein.The CaM isoform,AtCaM3,in Arabidopsis is a key member in the HS signal trans-duction pathway.AtCaM3 regulates the activity of CaM-binding protein kinase(AtCBK3) or protein phosphatase(AtPP7),promoting the activation of the HS transcription factor,AtHSFA1a,by phosphorylation/dephosphorylation and the expression of heat shock pro-tein genes,then improving heat tolerance in plants. | Rengang Zhou Bing Li Hongtao Liu Daye Sun | 2009 | Progress in Natural Science:Materials International2009,19,10: | 10 |
| 2 | Extracellular calmodulin accelerates rbcS-GUS expression in suspension-cultured transgenic tobacco cell显示文摘The role of extracellular calmodulin in regulating expression of rbcS in darkness was examined. A suspension-cultured cell line was generated from the transgenic rbcS-GUS tobacco. It was demonstrated that purified calmodulin added to the media enhanced rbcS-GUS expression. The time course of expression of rbcS-GUS and that of the secretion of calmodulin in the suspended transgenic tobacco cells in darkness were very similar. Both showed initial increase followed by decline with maximum calmodulin secretion preceding maximum GUS expression. The addition of membrane-impermeable calmodulin antagonist W7-agarose inhibited the expression of rbcS-GUS in darkness, but this inhibitory effect was completely reversed by adding exogenous purified calmodulin. | 3 | Effects of extracellular calmodulin on pollen germination and tube growth显示文摘The effects of calmodulin (CaM) antagonist W7_agarose, anti_CaM serum and exogenous purified CaM on pollen germination and tube growth of Forsythia suspensa were studied. The pollen germination and tube growth were inhibited or completely stopped by CaM antagonist W7_agarose. The addition of exogenous purified CaM stimulated pollen germination and tube growth, whereas the same amount of bovine serum albumin (BSA) had no effect. The inhibitory effects caused by W7_agarose and anti_CaM serum could be reversed completely by the addition of exogenous purified CaM. The tube growth of germinated pollen was also inhibited or completely stopped by W7_agarose. But recent evidence verified that peptide primary messengers are also involved in plant defense responses, the recognition between pollen and stigma and keep the balance between cell proliferation and differentiations in shoot apical meristems. Those results suggest that plants may actually make wide use of peptide primary messengers, both in embryonic development and late life when they rally their cells to defend against pathogens and insect pests. | 15 | Molecular cloning of a full-length cDNA for ECBP21 from Angelica dahurica显示文摘ECBP21 is an extracellular calmodulin-binding protein which was first detected and purified from extracellular extracts of suspension-cultured cells of Angelica da-hurica. The purified protein was electroblotted onto PVDF membrane and the amino acid sequences from 1 to 20 were determined. Using degenerate oligonucleotides of the sequence, a full-length cDNA coding for ECBP21 was isolated by a combination of RT-PCR and 5’-RACE cloning. The cDNA contains 947 nucleotides and codes for a precursor protein of 216 amino acids. The N-terminal 1-25 amino acid sequence is a predicted signal peptide and the other 26-216 amino acid sequence is a mature peptide. The 26-45 amino acid sequence shows identity with the N-terminal amino acid sequence of purified ECBP21 from Angelica dahurica. The fragment of encoding the mature protein was cloned into pET-28b(+) and transformed into E. colt BL21(DE3). A protein with relative molecular mass 21 ku was expressed in E. coli. | 16 | Effect of Flotation Reagents on the Cake Moisture of Copper Concentrate显示文摘The effect of reagents used in separating chalcopyrite from pyrite on the cake moisture of the copper concentrate at Daye Iron Mine Mineral Processing Plant was investigated. The results showed that the dosage of lime used for depressing pyrite was the main factor that increased the filter cake moisture of copper concentrate. With increasing the dosage of lime, the cake moisture of copper concentrate increased sharply. The cause was concluded to be the addition of lime to the pulp, which resulted in the formation of floc and a high pH value. The collector Z-200#, used for collecting chalcopyrite, had, as well, an adverse effect on the cake moisture of copper concentrate, but its effect was inferior in respect to that of lime. The cake moisture of copper concentrate can be decreased by changing the method with which lime is added and the pH value of pulp is regulated. The experiment results showed that the sulfuric acid was the best regulator. When the clarified liquor of lime was used as a depressant and the pH value of the pulp was regulated to 6.5€*7.0 by adding sulfuric acid, the cake moisture of copper concentrate was reduced from 15.49% to 13.13%. The examination of chalcopyrite surface by using ESCA (Electron Spectroscopy for Chemical Analysis) showed that calcium sulfate and iron hydroxide had formed on the surface of chalcopyrite when lime was added to the pulp. The formation of calcium sulfate and iron hydroxide on its surface increased the hydrophilicity of chalcopyrite so that its cake moisture increased.
