1Identification of ferroptosis as a novel mechanism for antitumor activity of natural product derivative a2 in gastric cancer显示文摘Ferroptosis is a type of cell death accompanied by iron-dependent lipid peroxidation,thus stimulating ferroptosis may be a potential strategy for treating gastric cancer,therapeutic agents against which are urgently required.Jiyuan oridonin A(JDA) is a natural compound isolated from Jiyuan Rabdosia rubescens with anti-tumor activity,unclear anti-tumor mechanisms and limited water solubility hamper its clinical application.Here,we showed a2,a new JDA derivative,inhibited the growth of gastric cancer cells.Subsequently,we discovered for the first time that a2 induced ferroptosis.Importantly,compound a2 decreased GPX4 expression and overexpressing GPX4 antagonized the anti-proliferative activity of a2.Furthermore,we demonstrated that a2 caused ferrous iron accumulation through the autophagy pathway,prevention of which rescued a2 induced ferrous iron elevation and cell growth inhibition.Moreover,a2 exhibited more potent anti-cancer activity than 5-fluorouracil in gastric canc er cell line-derived xenograft mice models.Patient-derived tumor xenograft models from different patients displayed varied sensitivity to a2,and GPX4 downregulation indicated the sensitivity of tumors to a2.Finally,a2 exhibited well pharmacokinetic characteristic s.Overall,our data suggest that inducing ferroptosis is the major mechanism mediating anti-tumor activity of a2,and a2 will hopefully serve as a promising compound for gastric cancer treatment.Ying Liu Zan Song Yajie Liu Xubin Ma Wang Wang Yu Ke Yichao Xu Dequan Yu Hongmin Liu 2021Acta Pharmaceutica Sinica B2021,11,6:10
2Helioscopianoids A–Q,bioactive jatrophane diterpenoid esters from Euphorbia helioscopia显示文摘The EtOH extracts of the whole plants of Euphorbia helioscopia afforded 17 new jatrophane diterpenoid esters, helioscopianoids A–Q(1–17), along with eight known compounds(18–25). Their structures were elucidated by extensive spectroscopic methods and Mo_2(OAc)_4-induced ECD analysis,and the structures of compounds 1, 2, and 7 were confirmed by X-ray crystallography. Compounds 1–17 were evaluated for inhibitory effects on P-glycoprotein(P-gp) in an adriamycin(ADM)-resistant human breast adenocarcinoma cell line(MCF-7/ADR) and neuroprotective effects against serum deprivationinduced and rotenone-induced PC12 cell damage. Compounds 8 and 16 increased the accumulation of ADM in MCF-7/ADR cells by approximately 3-fold at a concentration of 20 μmol/L. 4AN EFFICIENT APPROACH TO COMMENT SPAM IDENTIFICATION显示文摘This paper proposes a novel approach to comment spam identification based on content analysis. Three main features including the number of links, content repetitiveness, and text similarity are used for comment spam identification. In practice, content repetitiveness is determined by the length and frequency of the longest common substring. Furthermore, text similarity is calculated using vector space model. The precisions of preliminary experiments on comment spam identification conducted on Chinese and English are as high as 93% and 82% respectively. The results show the validity and language independency of this approach. Compared with conventional spam filtering approaches, our method requires no training, no rule sets and no link relationships. 6AN EFFICIENT APPROACH TO IMPORTANT BLOGGERS DISCOVERY显示文摘Popularity of blogs and the amount of information in the blogosphere increase so fast that it is difficult for Internet users to search the information they care about. Compared with conventional webs,links in the blogosphere are more abundant and conversations between bloggers are more fre-quent. This paper proposes a method of ranking bloggers based on link analysis,which can exemplify the characteristics of blogs,and reduce the influence of link spamming. This method can also bring convenience to users to read blogs,and it can supply a new methodology for information retrieval in the blogosphere. To ensure the reliability of the ranking results,some evaluation indicators of the im-portant bloggers are proposed,and the grading results of bloggers using the proposed method is compared with that using other indicators. 7A unified optimal planner for autonomous parking vehicle显示文摘In order to reduce the controlling difficulty caused by trajectory meandering and improve the adaptability to parking into regular lots,a versatile optimal planner(OP)is proposed.Taking advantage of the low speed specificity of parking vehicle,the OP algorithm was modeled the planning problem as a convex optimization problem.Collision-free constraints were formalized into the shortest distance between convex sets by describing obstacles and autonomous vehicle as affine set.Since employing Lagrange dual function and combining KKT conditions,the collision-free constraints translated into convex functions.Taking the national standard into account,5 kinds of regular parking scenario,which contain 0°,30°,45°,60°and 90°parking lots,were designed to verify the OP algorithm.The results illustrate that it is benefit from the continuous and smooth trajectory generated by the OP method to track,keep vehicle's stability and improve ride comfort,compared with A*and hybrid A*algorithms.Moreover,the OP method has strong generality since it can ensure the success rate no less than 82%when parking planning is carried out at the start node of 369 different locations.Both of evaluation criteria,as the pear error and RMSE in x direction,y axis and Euclidean distance d,and heading deviation 6,are stable and feasible in real tests,which illustrates that the OP planner can satisfy the requirements of regular parking scenarios.Dequan ZENG Zhuoping YU Lu XIONG Peizhi ZHANG Zhiqiang FU 2019Control Theory and Technology2019,17,4:0
8New compounds from the roots of Coriaria nepalensis显示文摘One new sesquiterpene lactone, corialactone E(1), one new neolignan, coriarianeolignan A(2), together with three known apocarotenoids(3-5) and one known neolignan(6) have been isolated from a CHCl_3 extract of the roots of Coriaria nepalensis. The structures including absolute configurations of 1-6 were elucidated through extensive NMR, HR-ESIMS, and CD data analysis. Structurally, compound 1 possessed novel variations in the structure, including the newly formed ether ring of C-3/0/C-9 and the lactone ring connecting C-13 and C-5. Compound 5 showed cytotoxic activity against SKOV3(human ovarian cancer)cells with IC_(50) values of 4.67 μmol/L. 9AUGER STUDY OF CARBON LAYER ON ON IMPLANTED STEEL SURFACE显示文摘The carbon layers on implanted steel surface have been studied by means of Auger spectra. Itis shown that the thickness of the carbon layer is proportional to the dose of implanted ions.By comparison with the results of friction and wear tests, the friction coefficient is smallerthan 0.20 at the first part of the friction coefficient curve. 10Study on the Practical and Ideal Mature Age of Pinus densiflora显示文摘This study was conducted on the analytic tree and got the fitting empirical equation of tree growth, in which the tree growth rate was used as the variable and time as the independent variable. The arithmetical operation to the function got the mature age of tree growth, and the practical and ideal mature ages of Pinus densiflora were 58 and 123.5 a, respectively.
