| 2 | Gene editing nuclease and its application in tilapia显示文摘Gene editing nucleases including zinc-finger nucleases(ZFNs), transcription activator like effector nucleases(TALENs) and clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated(Cas) system(CRISPR/Cas9) provide powerful tools that improve our ability to understand the physiological processes and their underlying mechanisms. To date, these approaches have already been widely used to generate knockout and knockin models in a large number of species. Fishes comprise nearly half of extant vertebrate species and provide excellent models for studying many aspects of biology. In this review, we present an overview of recent advances in the use of gene editing nucleases for studies of fish species. | 3 | The transcription factor Sox30 is involved in Nile tilapia spermatogenesis显示文摘Spermatogenesis is a complex process in which spermatogonial stem cells differentiate and develop into mature spermatozoa.The transcriptional regulatory network involved in fish spermatogenesis remains poorly understood.Here,we demonstrate in Nile tilapia that the Sox transcription factor family member Sox30 is specifically expressed in the testes and mainly localizes to spermatocytes and spermatids.CRISPR/Cas9-mediated sox30 mutation results in abnormal spermiogenesis,reduction of sperm motility,and male subfertility.Comparative transcriptome analysis shows that sox30 mutation alters the expression of genes involved in spermatogenesis.Further chromatin immunoprecipitation followed by high-throughput sequencing(Ch IP-seq),Ch IP-PCR,and luciferase reporter assays revealed that Sox30 positively regulates the transcription of ift140 and ptprb,two genes involved in spermiogenesis,by directly binding to their promoters.Our data,taken together,indicate that Sox30 plays an essential role in Nile tilapia spermatogenesis by directly regulating the transcription of the spermiogenesis-related genes ift140 and ptprb. | Ling Wei Yaohao Tang Xianhai Zeng Yueqin Li Song Zhang Li Deng Lingsong Wang Deshou Wang | 2022 | Journal of Genetics and Genomics2022,49,7: | 2 |

| 7 | Occurrence and risk assessment of triclosan in freshwater lakes in the middle Yangtze River basin (Wuhan, Central China)显示文摘Triclosan(TCS)is an endocrine disrupting chemical which is commonly used as a disinfectant in pharmaceuticals and personal care products(PPCP's).Since early 2020,the worldwide outbreak of COVID-19 has increased the use of PPCP's,so the occurrence and impact of TCS on freshwater lakes should be considered.However,little attention has been given to the effect of TCS on freshwater lakes in China.This study is the first attempt at a risk assessment focusing on the temporal and spatial occurrence of TCS in freshwater lakes in the middle Yangtze River basin.The surface water and sediments of Donghu Lake and Liangzi Lake(Wuhan,Central China)were collected from October 2020 to August 2021.The maximum concentrations of TCS were 466 ng/L and 239 ng/L in surface water,71 ng/g and 25 ng/g(dry weight)in sediments of Donghu Lake and Liangzi Lake,respectively.Significant temporal and spatial differences of TCS were observed within and between the lakes,with the highest concentrations measured in winter.Furthermore,higher concentrations of TCS were observed in areas that are more impacted by human activities.There was a significant positive correlation between TCS and nitrogen in the surface water.A risk assessment using the risk quotient(RQ)method showed that a potentially high risk(RQ>1)was found only in surface waters from Donghu Lake,and that the sediments posed a lower risk than the surface waters.These results provide timely data on the temporal and spatial occurrence of TCS in freshwater lakes in China following the outbreak of COVID-19 and demonstrate a possible high risk of exposure to TCS for aquatic biota. | Yuewei Wang Deshou Cun Zihan Zhang Dongwei Pu Xiaohui Li Wei Liang Tao Fang | 2022 | Water Biology and Security2022,1,4: | 1 |

| 16 | Removal of arsenic by pilot-scale vertical flow constructed wetland显示文摘Four pilot-scale Vertical Flow Constructed Wetlands(VFCWs)filled with gravel and planted with Phragmites australis were operated for seven months in the field to study the efficiency of arsenic removal in contaminated wastewater.The average arsenic removal efficiency by the VFCWs was 52.0%±20.2%,52.9%±21.3%,and 40.3%±19.4%at the theoretical concentrations of 50µg/L(CW50),100µg/L(CW100),and 500µg/L(CW500)arsenic in the wastewater,respectively.The results also showed no significant differences in the removal efficiency for conventional contaminants(nitrogen,phosphorus,or chemical oxygen demand)between wastewater treatments that did or did not contain arsenic(P>0.05),except for phosphorus in CW500.The highest average monthly removal rate of arsenic occurred in August(55.9%–74.5%)and the lowest in November(7.8%–15.5%).The arsenic removal efficiency of each VFCW was positively correlated with temperature(P<0.05).Arsenic accumulated in both substrates and plants,with greater accumulation associated with increased arsenic concentrations in the influent.The maximum accumulated arsenic concentrations in the substrates and plants at the end of the experiment were 4.47 mg/kg and 281.9 mg/kg,respectively,both present in CW500.The translocation factor(TF)of arsenic in the reeds was less than 1,with most of the arsenic accumulating in the roots.The arsenic mass balance indicated that substrate accumulation contributed most to arsenic removal(19.9%–30.4%),with lower levels in plants(3.8%–9.5%).In summary,VFCWs are effective for the treatment of arsenic-containing wastewater. | Yaocheng Fan Tiancui Li Deshou Cun Haibing Tang Yanran Dai Feihua Wang Wei Liang | 2021 | Frontiers of Environmental Science & Engineering2021,15,4: | 0 |
| 17 | Dibutyl phthalate adsorption characteristics using three common substrates in aqueous solutions显示文摘In recent years,the presence and adverse impacts of phthalic acid esters m aquatic environments have gained increasing attention.This work investigated the adsorption behavior of a typical phthalic acid ester,dibutyl phthalate(DBP),onto steel slag,gravel,and shell sand(substrates commonly used in constructed wetlands).The influence of dissolved organic matter(DOM)on DBP adsorption was investigated using humic acid as a proxy for DOM.The results demonstrated that the adsorption of DBP to three substrates reached equilibrium within 96 h,and the adsorption kinetics were well fitted by a pseudo-second-order model.The DBP adsorption isotherms were best fitted by the Langmuir adsorption model.The DBP adsorption capacity decreased in the order of steel slag>gravel>sheII sand,with values of 656 mg/kg,598 mg/kg,and 6.62 mg/kg at 25°C,respectively.DBP adsorbed to the surface of all substrates in a monolayer via an endothermic process.The DBP adsorption capacities of steel slag and gravel decreased as the DOM content increased.The DBP adsorption mechanisms to steel slag and gravel mainly involved the surface coordination of DBP with-OH or-COOH groups and electrostatic interactions.The results of this work suggest that steel slag and gravel may be ideal substrates for use in constructed wetlands to treat wastewater polluted with DBP. | Tiancui Li Yaocheng Fan Deshou Cun Yanran Dai Wei Liang | 2020 | Frontiers of Environmental Science & Engineering2020,14,2: | 0 |
| 18 | Cortisol safeguards oogenesis by promoting follicular cell survival显示文摘The role of glucocorticoids in oogenesis remains to be elucidated. cyp11c1 encodes the key enzyme involved in the synthesis of cortisol, the major glucocorticoid in teleosts. In our previous study, we mutated cyp11c1 in tilapia and analyzed its role in spermatogenesis. In this study, we analyzed its role in oogenesis. cyp11c1^(+/-)XX tilapia showed normal ovarian morphology but poor egg quality, as indicated by the mortality of embryos before 3 d post fertilization, which could be partially rescued by the supplement of exogenous cortisol to the mother fish. Transcriptome analyses revealed reduced expression of maternal genes in the eggs of the cyp11c1^(+/-)XX fish. The cyp11c1^(-/-)females showed impaired vitellogenesis and arrested oogenesis due to significantly decreased serum cortisol. Further analyses revealed decreased serum E2 level and expression of amh, an important regulator of follicular cell development, and increased follicular cell apoptosis in the ovaries of cyp11c1^(-/-)XX fish, which could be rescued by supplement of either exogenous cortisol or E2. Luciferase assays revealed a direct regulation of cortisol and E2 on amh transcription via GRs or ESRs.
