1Avenanthramides attenuate inflammation and atrophy in muscle cells显示文摘Background: Chronic inflammation is an important etiologic mechanism for muscle atrophy. Oat-derived phytochemical avenanthramides(AVAs) have been shown to suppress inflammatory responses in human clinical studies and in several cell lines in vitro, but their role in skeletal muscle is unclear. The aim of this study was to investigate whether AVA treatment can prevent tumor necrosis factor(TNF)-a-induced muscle fiber atrophy in C2 C12 cells.Methods: We treated 70% confluent cells for 24 h with AVA. Then, TNF-a was added to cell-cultured medium. Subsequently, cells were harvested at different time points. The cells were examined using various biochemical techniques for measuring protein, messenger RNA levels,nuclear binding activity, and viability. Fluorescence microscope was used for analysis of the myotube morphology.Results: Cells treated with TNF-a significantly increased nuclear factor k B activation, indicated by a marked decrease of Ik B(p < 0.05) and a6.6-fold increase in p65-DNA binding(p < 0.01); however, 30 mmol of AVA-A,-B, and-C treatment reduced the binding by 33%, 18%, and19%(p < 0.01), respectively, compared with cells treated with TNF-a without AVA. The interleukin-6 level increased by 2.5 fold(p < 0.01)with TNF-a, but decreased by 24%, 32%, and 28%(p < 0.01), respectively, with AVA-A,-B, and-C. The interleukin-1 b level also showed a47% increase with TNF-a(p < 0.01), whereas this increment was abolished in all AVA-treated cells. Reactive oxygen species production was1.3-fold higher in the TNF-a-treated group(p < 0.01) but not in the TNF-a + AVAs groups. Messenger RNA levels of muscle-specific E3 ubiquitin ligase atrogin-1 increased 23% in TNF-a vs. control(p < 0.05) but was decreased by 46%, 34%, and 53%(p < 0.01), respectively, with treatment of AVA-A,-B, and-C. Moreover, TNF-a treatment increased the muscle RING finger 1 messenger RNA level by 76%(p < 0.01);this change was abolished by AVAs. Cells treated with TNF-a demonstrated a reduced proliferation compared with control cells(p < 0.01), but this effect was not seen in TNF-a + AVAs cells. The diameter of the C2 C12 myotube decreased by 28%(p < 0.01) with TNF-a, whereas it showed no change when AVAs were included in the cell media.Conclusion: These results indicated that AVAs can reduce proinflammatory cytokine and reactive oxygen species production and ameliorate TNF-a-induced myotube atrophy in muscle cells.Dongwook Yeo Chounghun Kang Tianou Zhang Li Li Ji 2019Journal of Sport and Health Science2019,8,2:3
2Stiefel–Whitney classes and topological phases in band theory显示文摘We review the recent progress in the study of topological phases in systems with space–time inversion symmetry IST.ISTis an anti-unitary symmetry which is local in momentum space and satisfies I2 ST= 1 such as PT in two dimensions(2 D)and three dimensions(3 D) without spin–orbit coupling and C2 T in 2 D with or without spin–orbit coupling, where P, T, C2 indicate the inversion, time-reversal, and two-fold rotation symmetries, respectively. Under IST, the Hamiltonian and the periodic part of the Bloch wave function can be constrained to be real-valued, which makes the Berry curvature and the Chern number vanish. In this class of systems, gapped band structures of real wave functions can be topologically distinguished by the Stiefel–Whitney numbers instead. The first and second Stiefel–Whitney numbers w1 and w2, respectively,are the corresponding invariants in 1 D and 2 D, which are equivalent to the quantized Berry phase and the Z2 monopole charge, respectively. We first describe the topological phases characterized by the first Stiefel–Whitney number, including1 D topological insulators with quantized charge polarization, 2 D Dirac semimetals, and 3 D nodal line semimetals. Next we review how the second Stiefel–Whitney class characterizes the 3 D nodal line semimetals carrying a Z2 monopole charge.In particular, we explain how the second Stiefel–Whitney number w2, the Z2 monopole charge, and the linking number between nodal lines are related. 3Advanced metal–organic frameworks for aqueous sodium-ion rechargeable batteries显示文摘Inexpensive and abundant sodium resources make energy storage systems using sodium chemistry promising replacements for typical lithium-ion rechargeable batteries(LIBs).Fortuitously,aqueous sodium-ion rechargeable batteries(ASIBs),which operate in aqueous electrolytes,are cheaper,safer,and more ionically conductive than batteries that operate in conventional organic electrolytes;furthermore,they are suitable for grid-scale energy storage applications.As electrode materials for storing Na~+ ions in ASIBs,a variety of multifunctional metal-organic frameworks(MOFs) have demonstrated great potential in terms of having porous 3 D crystal structures,compatibility with aqueous solutions,long cycle lives(≥1000 cycles),and ease of synthesis.The present review describes MOF-derived technologies for the successful application of MOFs to ASIBs and suggests future challenges in this area of research based on the current understanding.Dongkyu Choi Seonguk Lim Dongwook Han 2021Journal of Energy Chemistry2021,30,2:2
4Inhibition of BET selectively eliminates undifferentiated pluripotent stem cells显示文摘Embryonic stem cells(ESCs) maintain their cellular identity through the systematic regulation of master transcription factors and chromatin remodeling complexes. Recent work has shown that the unusually large-scale enhancers—namely super-enhancers(SEs), on which BRD4, a member of the bromodomain and extraterminal domain(BET) family is highly enriched—could regulate pluripotency-related transcription factors. Moreover, inhibition of BRD4 binding on SEs has been shown to induce the differentiation of ESCs. However, the underlying mechanism of BRD4 inhibition-mediated stem cell differentiation remains elusive. Here we show that both mouse and human ESCs lose their capacity for self-renewal upon treatment with JQ1, a selective inhibitor of BET family including BRD4, with rapid suppression of pluripotency-associated genes.
