1Efficacy and mechanism of methyl salicylate in the enhancement of skin delivery of herbal medicines显示文摘Objective: To elucidate the molecular mechanism(s) by which methyl salicylate enhances the skin delivery of herbal ingredients with diverse lipophilicity.Methods: The toxicity of methyl salicylate on skin cell lines was evaluated using the MTT assay. The Franz diffusion cell method was used to measure the permeability enhancing activities of methyl salicylate for five herbal ingredients with a range of lipophilicities. The interaction between methyl salicylate and the stratum corneum(SC) was observed by using an infrared spectroscopy technique. Moreover, the solubilities and SC-vehicle partition coefficient were determined to monitor the impact of methyl salicylate on the drug thermodynamic activities and partition into the SC layer, respectively.Results: Compared with azone(1-dodecylazacycloheptan-2-one), methyl salicylate showed lower toxicity to skin cells in terms of the IC50 values. The in vitro skin permeation studies showed that methyl salicylate could greatly improve the cumulative amounts or steady state flux of the selected model drugs with the exception of osthole, which indicated that methyl salicylate was prone to promote the skin delivery of hydrophilic drugs. The Fourier transform infrared spectroscopy studies revealed that methyl salicylate mainly interacted with SC lipids, leading to the disruption of the orderly arrangement of the SC.In addition, methyl salicylate had no obvious effect on the drug thermodynamic activity and partition into the SC.Conclusion: Methyl salicylate could effectively promote the skin delivery of relatively hydrophilic ingredients in externally used traditional Chinese medicines(TCM) without obvious cytotoxicity.Jingyan Wang Xinyu Zhao Jiajia Chen Xin Li Doudou Hao Tingting Li Kai Mei Yi Lan Qing Wu 2021Journal of Traditional Chinese Medical Sciences2021,8,4:0
2Urine biomarkers discovery by metabolomics and machine learning for Parkinson’s disease diagnoses显示文摘Parkinson’s disease(PD)is a complex neurological disorder that typically worsens with age.A wide range of pathologies makes PD a very heterogeneous condition,and there are currently no reliable diagnostic tests for this disease.The application of metabolomics to the study of PD has the potential to identify disease biomarkers through the systematic evaluation of metabolites.In this study,urine metabolic profiles of 215 urine samples from 104 PD patients and 111 healthy individuals were assessed based on liquid chromatography-mass spectrometry.The urine metabolic profile was first evaluated with partial leastsquares discriminant analysis,and then we integrated the metabolomic data with ensemble machine learning techniques using the voting strategy to achieve better predictive performance.A combination of 8-metabolite predictive panel performed well with an accuracy of over 90.7%.Compared to control subjects,PD patients had higher levels of 3-methoxytyramine,N-acetyl-l-tyrosine,orotic acid,uric acid,vanillic acid,and xanthine,and lower levels of 3,3-dimethylglutaric acid and imidazolelactic acid in their urine.The multi-metabolite prediction model developed in this study can serve as an initial point for future clinical studies.Xiaoxiao Wang Xinran Hao Jie Yan Ji Xu Dandan Hu Fenfen Ji Ting Zeng Fuyue Wang Bolun Wang Jiacheng Fang Jing Ji Hemi Luan Yanjun Hong Yanhao Zhang Jinyao Chen Min Li Zhu Yang Doudou Zhang Wenlan Liu Xiaodong Cai Zongwei Cai 2023Chinese Chemical Letters2023,34,10:0
