| 3 | Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. Such a result supports the idea that the present desert landform of the northern Ulan Buh Desert started to form since 2 ka, which was likely a response to the desertification caused by ruin of the Han Dynasty and the large-scale abandonment of farming of the Han nationality. | 6 | Promoting ring-opening efficiency for suppressing toxic intermediates during photocatalytic toluene degradation via surface oxygen vacancies显示文摘Aromatic ring-opening process is well recognized as the rate-determining step for catalytic toluene degradation. In photocatalytic toluene degradation, the toxic intermediates w让h harmful effects may be generated. To clarify the precise reaction mechanism and control the toxic intermediates generation, a closely combined in situ DRIFTS and DFT calculation is utilized to address these important issues. We construct the BiOCl w让h oxygen vacancies (OVs) and reveal the structure of OVs. The defect level caused by oxygen vacancies could promote the light adsorption and charge separation, which further boosts the activation of ring-opening species and enhances the generation process of free radicals. The reaction energy barriers of four possible ring-opening processes on defective BiOCl (OVBOC) are all declined in comparison with perfect BiOCl (BOC). The existence of oxygen vacancies could smooth the ratedetermining step so the ring-opening efficiency of photocatalytic toluene degradation is highly increased. Most importantly, the methyl species would be further oxidized and tend to open the benzene-ring at benzoic acid on BOC while the ring would be broken at the benzyl alcohol on OVBOC. These results indicate that the toluene degradation pathway is shortened via the surface OVs, which enables the production of radicals with high oxidation capability for the accelerated chain scission of the ring-opening intermediates. Finally, the efficiency of the key ring-opening process could be enormously improved and toxic intermediates are effectively restrained. | 9 | Anion intercalated layered-double-hydroxide structure for efficient photocatalytic NO remove显示文摘Due to the easily controllable interlayer anions, metal cation composition proportion and thickness, which is beneficial to modify surface chemical state and tune bandgap, layered double hydroxides(LDHs) have great promising potential for photocatalytic applications. In this study,we have successfully synthesized the ZnAl–LDH intercalated the single anion between ZnAl cationic interlayer without anionic impurities by using a facile calcining and reconstructing routes. The electron structure and surface chemical state of the prepared products have been investigated by combining the DFT calculation and experimental characterization methods. UV–vis DRS was used to certify the light absorption of the prepared products, and we performed the DFT calculation to demonstrate the density of state and activation of reactant. These results suggested that the ZnAl–LDH–CO3 possessed the more proper band structure and superior ability to activate NO and O2 for accelerating the photocatalytic NO oxidation activity. Moreover, the in situ DRIFTS with dynamically monitoring intermediates and products over the ZnAl–LDH–CO3 was adopted to declare the photocatalytic NO oxidized process during the photocatalytic reaction process. | 13 | Developing a microfluidic-based system to quantify cell capture efficiency显示文摘Micro-fabrication technology has substantial potential for identifying molecular markers expressed on the surfaces of tissue cells and viruses. It has been found in several conceptual prototypes that cells with such markers are able to be captured by their antibodies immobilized on microchannel substrates and unbound cells are flushed out by a driven flow. The feasibility and reliability of such a microflu- idic-based assay, however, remains to be further tested. In the current work, we developed a microflu- idic-based system consisting of a microfluidic chip, an image grabbing unit, data acquisition and analysis software, as well as a supporting base. Specific binding of CD59-expressed or BSA-coupled human red blood cells (RBCs) to anti-CD59 or anti-BSA antibody-immobilized chip surfaces was quan- tified by capture efficiency and by the fraction of bound cells. Impacts of respective flow rate, cell concentration, antibody concentration and site density were tested systematically. The measured data indicated that the assay was robust. The robustness was further confirmed by capture efficiencies measured from an independent ELISA-based cell binding assay. | 14 | Hypertrophic cardiomyopathy: Can the noninvasive diagnostic testing identify high risk patients?显示文摘Hypertrophic cardiomyopathy(HCM) is the most common cause of sudden cardiac death(SCD) in the young, particularly among athletes. Identifying high risk individuals is very important for SCD prevention. The purpose of this review is to stress that noninvasive diagnostic testing is important for risk assessment. Extreme left ventricular hypertrophy and documented ventricular tachycardia and fibrillation increase the risk of SCD. Fragmented QRS and T wave inversion in multiple leads are more common in high risk patients. Cardiac magnetic resonance imaging provides complete visualization of the left ventricular chamber, allowing precise localization of the distribution of hypertrophy and measurement of wall thickness and cardiac mass. Moreover, with late gadolinium enhancement, patchy myocardial fibrosis within the area of hypertrophy can be detected, which is also helpful in risk stratification. DC. leaves(BB oil)on wound healing in mice.METHODS: Undiluted BB oil and its diluted solutions with olive oil to 1/5 and 1/10 to yield BB oil1/5and BB oil1/10 were applied to the wounded skin before wound healing conditions were assessed by healing rate, histopathology, and contents of collagen, hydroxyproline, and Neuropeptide Substance P(SP). All above results were compared with the efficacies of the control, pure olive oil, basic fibroblast growth factor(BFGF), and cream of Jing Wan Hong(JWH).RESULTS: BB oil1/5and BB oil1/10 improved wound contraction and closure. Histopathology study further confirmed a desirable histological organization of wound tissues. BB oil1/5and BB oil1/10 reduced the number of inflammatory cells, increased wound-healing rates, and significantly increased the hydroxyproline content. Both BB oil1/5and BB oil1/10 improved formation of collagen, and reduced the frequency of fibroblasts. Moreover, BB oil1/5and BB oil1/10 markedly promoted SP expression. However, undiluted BB oil may induce skin thickening and hardening, inhibite collagen synthesis and delay complete skin wound healing.CONCLUSION: The BB oil1/5and BB oil1/10 promoted capillary regeneration, blood circulation, collagen deposition, granular tissue formation, epithelial deposition, and wound contraction. | 17 | Timely renal replacement therapy linked to better outcome in patients with sepsis-associated acute kidney injury显示文摘Background:Recent studies suggest that acute kidney injury(AKI)can be treated with renal replacement therapy(RRT).However,its benefits to patients with sepsis-associated AKI(SA-AKI),which is linked to high mortality and morbidity rates,remain under debate.The aim of this study was to compare the outcomes of different RRT strategies for patients with SA-AKI.Methods:This retrospective study evaluated patients who were admitted to the hospital with sepsis and devel-oped SA-AKI during hospitalization from 1st January 2014 to 31st January 2019.Mortality,renal recovery,and systemic organ function at 90 days following admission were compared between the RRT group(RG)and non-RRT group(NRG),as well as the early-RRT group(EG)and delayed-RRT group(DG).The groups were defined according to the time from admission to RRT initiation(criterion 1,EG1 and DG1)and Kidney Disease Improving Global Outcomes(KDIGO)classification(criterion 2,EG2 and DG2).Categorical and continuous variables were compared using the chi-squared test or Fisher’s exact test and Student’s t-test or Wilcoxon test.Kaplan-Meier curves were constructed to determine the unadjusted survival rates for the different subgroups.Results:A total of 116 patients were included in this study;of those,38 received RRT and 46 expired within 90 days.Among different strategies of RRT,there were no significant differences found in 90-day mortality(RG vs.NRG:��2=0.610,P=0.435;EG1 vs.DG1:��2=0.835,P=0.360;EG2 vs.DG2:��2=0.022,P=0.899)and renal recovery.However,the values of change in sequential organ failure assessment(ΔSOFA)max-min of patients in the EG and RG were significantly higher than those recorded in the NRG(ΔSOFA RG=7.0,ΔSOFA NRG=3.60,ΔSOFA EG1=9.00,ΔSOFA EG2=6.30;P<0.050).Also,the 90-day renal recovery in the EG was better than that noted in the DG with criterion 1(87.5%vs.38.5%,respectively,��2=10.425,P=0.032),suggesting that RRT(especially timely RRT)may be beneficial to the restoration of systemic organ function in patients with SA-AKI.Conclusion:RRT did not reduce the 90-day mortality among patients with SA-AKI.However,timely RRT may benefit the restoration of systemic organ function,thereby improving the quality of life of patients. | Yiwen Fan Liang Chen Shaowei Jiang Yingying Huang Yuxin Leng Chengjin Gao | 2022 | Journal of Intensive Medicine2022,2,3: | 2 |
