1Varietal identification of tea(Camellia sinensis)using nanofluidic array of single nucleotide polymorphism(SNP)markers显示文摘Apart from water,tea is the world’s most widely consumed beverage.Tea is produced in more than 50 countries with an annual production of approximately 4.7 million tons.The market segment for specialty tea has been expanding rapidly owing to increased demand,resulting in higher revenues and profits for tea growers and the industry.Accurate varietal identification is critically important to ensure traceability and authentication of premium tea products,which in turn contribute to on-farm conservation of tea genetic diversity.Using a set of single nucleotide polymorphism(SNP)markers developed from the expressed sequence tag(EST)database of Camilla senensis,we genotyped deoxyribonucleic acid(DNA)samples extracted from a diverse group of tea varieties,including both fresh and processed commercial loose-leaf teas.The validation led to the designation of 60 SNPs that unambiguously identified all 40 tested tea varieties with high statistical rigor(p<0.0001).Varietal authenticity and genetic relationships among the analyzed cultivars were further characterized by ordination and Bayesian clustering analysis.These SNP markers,in combination with a high-throughput genotyping protocol,effectively established and verified specific DNA fingerprints for all tested tea varieties.This method provides a powerful tool for variety authentication and quality control for the tea industry.It is also highly useful for the management of tea genetic resources and breeding,where accurate and efficient genotype identification is essential.Wan-Ping Fang Lyndel W Meinhardt Hua-Wei Tan Lin Zhou Sue Mischke Dapeng Zhang 2014Horticulture Research2014,1,1:13
2Exogenous abscisic acid significantly affects proteome in teaplant (Camellia sinensis) exposed to drought stress显示文摘Tea[Camellia sinensis(L.)O.Kuntze]is an important economic crop,and drought is the most important abiotic stress affecting yield and quality.Abscisic acid(ABA)is an important phytohormone responsible for activating drought resistance.Increased understanding of ABA effects on tea plant under drought stress is essential to develop drought-tolerant tea genotypes,along with crop management practices that can mitigate drought stress.The objective of the present investigation is evaluation of effects of exogenous ABA on the leaf proteome in tea plant exposed to drought stress.Leaf protein patterns of tea plants under simulated drought stress[(polyethylene glycol(PEG)-treated]and exogenous ABA treatment were analyzed in a time-course experiment using two-dimensional electrophoresis(2-DE),followed by matrix-assisted laser desorption/ionization time-of-flight(MALDI-TOF)mass spectrometry(MS).Among the 72 protein spots identified by MALDI-TOF MS,16 proteins were downregulated and two were upregulated by exogenous ABA.The upregulated proteins have roles in glycolysis and photosystem II stabilization.Twenty-one protein spots were responsive to drought stress and most participate in carbohydrate and nitrogen metabolism,control of reactive oxygen species(ROS),defense,signaling or nucleic acid metabolism.The combined treatments of exogenous ABA and drought showed upregulation of 10 protein spots at 12 h and upregulation of 11 proteins at 72 h after initiation of drought stress.The results support the importance of the role that ABA plays in the tea plant during drought stress,by improving protein transport,carbon metabolism and expression of resistance proteins.Lin Zhou Hui Xu Sue Mischke Lyndel W Meinhardt Dapeng Zhang Xujun Zhu Xinghui Li Wanping Fang 2014Horticulture Research2014,1,1:12

4Developing single nucleotide polymorphism(SNP)markers from transcriptome sequences for identification of longan(Dimocarpus longan)germplasm显示文摘Longan(Dimocarpus longan Lour.)is an important tropical fruit tree crop.Accurate varietal identification is essential for germplasm management and breeding.Using longan transcriptome sequences from public databases,we developed single nucleotide polymorphism(SNP)markers;validated 60 SNPs in 50 longan germplasm accessions,including cultivated varieties and wild germplasm;and designated 25 SNP markers that unambiguously identified all tested longan varieties with high statistical rigor(P<0.0001).Multiple trees from the same clone were verified and off-type trees were identified.Diversity analysis revealed genetic relationships among analyzed accessions.Cultivated varieties differed significantly from wild populations(Fst=0.300;P<0.001),demonstrating untapped genetic diversity for germplasm conservation and utilization.Within cultivated varieties,apparent differences between varieties from China and those from Thailand and Hawaii indicated geographic patterns of genetic differentiation.These SNP markers provide a powerful tool to manage longan genetic resources and breeding,with accurate and efficient genotype identification.Boyi Wang Hua-Wei Tan Wanping Fang Lyndel W Meinhardt Sue Mischke Tracie Matsumoto Dapeng Zhang 2015Horticulture Research2015,2,1:6

9胰十二指肠切除术后胰胃吻合术vs胰空肠吻合术:一项随机对照试验的Meta分析显示文摘【背景】胰十二指肠切除术后剩余胰腺的最佳重建方式尚有争议。本研究旨在探讨胰胃吻合术和胰空肠吻合术2种较常用的重建方法围手术期的效果。【数据来源】从各文献库(MEDLINE/Pub Med,EMBASE,Web of Science,Cochrane图书馆)搜集对比胰胃吻合术和胰空肠吻合术的随机对照试验研究。最终纳入8项研究,其中607例行胰胃吻合术,604例行胰空肠吻合术。胰胃吻合术后患者胰瘘和腹内液体收集量明显低于胰空肠吻合术,差异有统计学意义,而两组间在延缓胃排空、胆漏、出血、再手术、伤口感染、总发病率、死亡率和住院时间方面比较差异均无统计学意义。【结论】本Meta分析显示,胰胃吻合术降低了术后胰瘘的发生率和严重程度,且腹内液减少。胰胃吻合术应该为更理想的胰腺重建术式。丁文珠 姚亚楠 汪谦 Que W Fang H Yan B 2015中华普通外科学文献(电子版)2015,9,5:3

11Association of types of diabetes and insulin dependency on birth outcomes显示文摘BACKGROUND Diabetes rates among pregnant women in the United States have been increasing and are associated with adverse pregnancy outcomes.AIM To investigate differences in birth outcomes(preterm birth,macrosomia,and neonatal death)by diabetes status.METHODS Cross-sectional design,using linked Missouri birth and death certificates(singleton births only),2010 to 2012(n=204057).Exposure was diabetes non-diabetic,pre-pregnancy diabetes-insulin dependent(PD-I),pre-pregnancy diabetes-non-insulin dependent(PD-NI),gestational diabetes-insulin dependent(GD-I),and gestational diabetes-non-insulin dependent(GD-NI).Outcomes included preterm birth,macrosomia,and infant mortality.Confounders included demographic characteristics,adequacy of prenatal care,body mass index,smoking,hypertension,and previous preterm birth.Bivariate and multivariate logistic regression assessed differences in outcomes by diabetes status.RESULTS Women with PD-I,PD-NI,and GD-I remained at a significantly increased odds for preterm birth(aOR 2.87,aOR 1.77,and aOR 1.73,respectively)and having a very large baby[macrosomia](aOR 3.01,aOR 2.12,and aOR 1.96,respectively);in reference to non-diabetic women.Women with GDNI were at a significantly increased risk for macrosomia(aOR1.53),decreased risk for their baby to die before their first birthday(aOR 0.41)and no difference in risk for preterm birth in reference to non-diabetic women.CONCLUSION Diabetes is associated with the poor birth outcomes.Clinical management of diabetes during pregnancy and healthy lifestyle behaviors before pregnancy can reduce the risk for diabetes and poor birth outcomes.Pamela K Xaverius Steven W Howard Deborah Kiel Jerry E Thurman Ethan Wankum Catherine Carter Clairy Fang Romi Carriere 2022World Journal of Clinical Cases2022,10,7:2

15Risk stratification in symptomatic intracranial atherosclerotic disease with conventional vascular risk factors and cerebral haemodynamics显示文摘Background and purpose Symptomatic intracranial atherosclerotic stenosis(sICAS)is associated with a considerable risk of recurrent stroke despite contemporarily optimal medical treatment.Severity of luminal stenosis in sICAS and its haemodynamic significance quantified with computational fluid dynamics(CFD)models were associated with the risk of stroke recurrence.We aimed to develop and compare stroke risk prediction nomograms in sICAS,based on vascular risk factors and these metrics.Methods Patients with 50%-99%sICAS confirmed in CT angiography(CTA)were enrolled.Conventional vascular risk factors were collected.Severity of luminal stenosis in sICAS was dichotomised as moderate(50%-69%)and severe(70%-99%).Translesional pressure ratio(PR)and wall shear stress ratio(WSSR)were quantified via CTA-based CFD modelling;the haemodynamic status of sICAS was classified as normal(normal PR&WSSR),intermediate(otherwise)and abnormal(abnormal PR&WSSR).All patients received guideline-recommended medical treatment.We developed and compared performance of nomograms composed of these variables and independent predictors identified in multivariate logistic regression,in predicting the primary outcome,recurrent ischaemic stroke in the same territory(SIT)within 1 year.Results Among 245 sICAS patients,20(8.2%)had SIT.The D2H2A nomogram,incorporating diabetes,dyslipidaemia,haemodynamic status of sICAS,hypertension and age≥50 years,showed good calibration(P for Hosmer-Lemeshow test=0.560)and discrimination(C-statistic 0.73,95%CI 0.60 to 0.85).It also had better performance in risk reclassification and provided larger net benefits in decision curve analysis,compared with nomograms composed of conventional vascular risk factors only,and plus the severity of luminal stenosis in sICAS.Sensitivity analysis in patients with anterior-circulation sICAS showed similar results.Conclusions The D2H2A nomogram,incorporating conventional vascular risk factors and the haemodynamic significance of sICAS as assessed in CFD models,could be a useful tool to stratify sICAS patients for the risk of recurrent stroke under contemporarily optimal medical treatment.Xuan Tian Hui Fang Linfang Lan Hing Lung Ip Jill Abrigo Haipeng Liu Lina Zheng Florence S Y Fan Sze Ho Ma Bonaventure Ip Bo Song Yuming Xu Jingwei Li Bing Zhang Yun Xu Yannie O Y Soo Vincent Mok Ka Sing Wong Thomas W Leung Xinyi Leng 2023Stroke & Vascular Neurology2023,8,1:2
