| 1 | Nanoparticles for targeted delivery of therapeutics and small interfering RNAs in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC) is the 5th most common malignancy which is responsible for more than half million annual mortalities; also, it is the third leading cause of cancer related death. Unfavorablesystemic side-effects of chemotherapeutic agents and susceptibility to the degradation of small interfering RNAs(si RNAs), which can knock down a specific gene involved in the disease, have hampered their clinical application. So, it could be beneficial to develop an efficient carrier for the stabilization and specific delivery of drugs and si RNA to cells. Targeted nanoparticles have gained considerable attention as an efficient drug and gene delivery system, which is due to their capability in achieving the highest accumulation of cytotoxic agents in tumor tissue, modifiable drug pharmacokinetic- and bio-distribution, improved effectiveness of treatment, and limited sideeffects. Recent studies have shed more light on the advantages of novel drug loaded carrier systems vs free drugs. Most of the animal studies have reported improvement in treatment efficacy and survival rate using novel carrier systems. Targeted delivery may be achieved passively or actively. In passive targeting, no ligand as homing device is used, while targeting is achieved by incorporating the therapeutic agent into a macromolecule or nanoparticle that passively reaches the target organ. However, in active targeting, the therapeutic agent or carrier system is conjugated to a tissue or cell-specific receptor which is overexpressed in a special malignancy using a ligand called a homing device. This review covers a broad spectrum of targeted nanoparticles as therapeutic and nonviral si RNA delivery systems, which are developed for enhanced cellular uptake and targeted gene silencing in vitro and in vivo and their characteristics and opportunities for the clinical applications of drugs and therapeutic si RNA are discussed in this article. Asialoglycoprotein receptors, low-density lipoprotein, ganglioside GM1 cell surface ligand, epidermal growth factor receptor receptors, monoclonal antibodies, retinoic acid receptors, integrin receptors targeted by Arg-Gly-Asp peptide, folate, and transferrin receptors are the most widely studied cell surface receptors which are used for the site specific delivery of drugs and si RNA-based therapeutics in HCC and discussed in detail in this article. | Jaleh Varshosaz Maryam Farzan | 2015 | World Journal of Gastroenterology2015,21,42: | 10 |
| 2 | Australian tertiary care outcomes of entecavir monotherapy in treatment naive patients with chronic hepatitis B显示文摘AIM:To evaluate the long-term treatment outcomes of entecavir monotherapy in treatment naive patients in an Australian tertiary care setting. METHODS:A retrospective analysis of treatment naive patients receiving entecavir monotherapy through Westmead Hospital was performed.Patients were excluded if they had received previous treatment with another nucleoside or nucleotide analogue,were pregnant or less than 18 years old. RESULTS:Out of 336 patients,163 patients fulfilled the selection criteria.Range of follow up was 3-46 mo (mean 26 mo).134 patients(82.2%)had pre-treatment biopsies,with 26 patients(16.0%)demonstrating F3-4 fibrosis.In total,153 patients(93.9%)achieved at least Partial Virological Suppression(PVS),with 134 patients (82.2%)achieving complete virological suppression. The cumulative CVS and PVS rates at 36 mo were 82.1%and 96.4%,respectively.3 patients(1.8%)failed to achieve PVS,while 5 patients(3.0%)developed virological rebound.128 patients(78.5%)maintained CVS throughout follow up.Predictors of CVS included lower baseline DNA level(P=0.001),hepatitis B virus e antigen negative status(P=0.001)and increasing age at treatment(log rank 0.001).No significant adverse effects were reported necessitating cessation of entecavir. | 3 | Application of positron emission tomography/computed tomography in radiation treatment planning for head and neck cancers显示文摘18-fluorodeoxygluocose positron emission tomography/computed tomography(18FDG-PET/CT) provides significant information in multiple settings in the management of head and neck cancers(HNC). This article seeks to define the additional benefit of PET/CT as related to radiation treatment planning for squamous cell carcinomas(SCCs) of the head and neck through a review of relevant literature. By helping further define both primary and nodal volumes, radiation treatment planning can be improved using PET/CT. Special attention is paid to the independent benefit of PET/CT in targeting mucosal primaries as well as in detecting nodal metastases. The utility of PET/CT is also explored for treatment planning in the setting of SCC of unknown primary as PET/CT may help define a mucosal target volume by guiding biopsies for examination under anesthesia thus changing the treatment paradigm and limiting the extent of therapy. Implications of the use of PET/CT for proper target delineation in patients with artifact from dental procedures are discussed and the impact of dental artifact on CT-based PET attenuation correction is assessed. Finally, comment is made upon the role of PET/CT in the high-risk post-operative setting, particularly in the context of radiation dose escalation. | 4 | Application of fluorodeoxyglucose positron emission tomography in the management of head and neck cancers显示文摘The use of fluorodeoxyglucose positron emission to-mography(FDG PET) scan technology in the manage-ment of head and neck cancers continues to increase. We discuss the biology of FDG uptake in malignant lesions and also discuss the physics of PET imaging. The various parameters described to quantify FDG up-take in cancers including standardized uptake value, metabolic tumor volume and total lesion glycolysis are presented. PET scans have found a significant role in the diagnosis and staging of head and neck cancers. They are also being increasingly used in radiation ther-apy treatment planning. Many groups have also used PET derived values to serve as prognostic indicators of outcomes including loco-regional control and overall survival. FDG PET scans are also proving very useful in assessing the efficacy of treatment and management and follow-up of head and neck cancer patients. This review article focuses on the role of FDG-PET com-puted tomography scans in these areas for squamous cell carcinoma of the head and neck. | 13 | Effects of synthesized nanoparticles and Henna-Tragacanth solutions on oil/water interfacial tension:Nanofluids stability considerations显示文摘Rising global energy demand has encouraged engineers to create and design new methods to improve oil recovery from reservoirs.In this study,feasibility of using Henna extract as a natural surfactant and synthesized nanoparticles(Titanium dioxide(TiO2),Silicon dioxide(SiO2),Graphene and composite of TiO2-Graphene)for reduction of oil-water interfacial tension has been experimentally investigated.Nanoparticles were synthesized via sol-gel method and XRD,FESEM,EDAX and FTIR tests were conducted to confirm the authenticity of this synthesizing materials.Nano-surfactants were stabled with a natural water-based suspending surfactant called Tragacanth extract,which could be introduced as a practical substitute for industrial nanoparticles'stabilizers in oil industry.After CMC determination of Henna extract surfactant,the optimal concentration of Tragacanth extract surfactant,with the purpose of nano-surfactants’stabilization,was determined through particle size and zeta potential tests.Results of interfacial tension(IFT)measurements showed that the increase of Henna extract concentration from 0 wt%to 10 wt%reduced IFT between kerosene and water from 37.23 to 15.24 mN/m.Furthermore,adding 1 wt%of synthesized TiO2 nanoparticle to the Henna extract surfactant at its CMC value reduced IFT from 18.43 to 14.57 mN/m.As an impact of this significant reduction in IFT value,oil recovery factor could be improved drastically during EOR operations.Results proved that TiO2 nano-surfactant was as effective as industrial surfactants,which put human's and environment's health at risk and impose heavy economic strain on governments. | Pouya Tavakoli Seyed Reza Shadizadeh Farzan Hayati Moslem Fattahi | 2020 | Petroleum2020,6,3: | 1 |
