1Fast extraction of winter wheat planting area in Huang-Huai-Hai Plain using high-resolution satellite imagery on a cloud computing platform显示文摘To extract regional winter wheat planting area using higher-resolution satellite imagery still faces many challenges due to large data size and long processing time in traditional remote sensing classification.Google Earth Engine(GEE),a cloud computing analysis platform based on global geospatial analysis,provides a new opportunity for rapid analysis of remote sensing data.In this study,high-quality Landsat-8 imagery was used to extract the winter wheat planting area from the Huang-Huai-Hai Plain in China.The random forest algorithm was used to identify and map the winter wheat sown in 2019 and harvested in 2020,and Sentinel-2 imagery was used to verify the results.The spectral indices,texture,and terrain features of the image were derived,and their contribution to the classification accuracy of winter wheat was evaluated by scoring.Then the top nine features were selected to form an optimal feature subset.Comparing the set of thirty-four features and the optimized feature subset as the input variables of the random forest classifier,the results show that the accuracy difference between the two feature classification schemes is small,but the classification effect of all feature sets is slightly better than the optimal feature subset.The overall classification accuracy of sample plots verification was 86%-95%,the Kappa coefficient was between 0.7 and 0.85,and the percentage error of the total area was 5.42%.The research demonstrates a reliable method for mapping a wide range of winter wheat planting area,and provides a good prospect for exploring the precise mapping of other crops,which is of great significance to crop monitoring and agricultural development.Dongyan Zhang Mengru Zhang Fenfang Lin Zhenggao Pan Fei Jiang Liang He Hang Yang Ning Jin 2022International Journal of Agricultural and Biological Engineering2022,15,1:0
2Imatinib blocks tyrosine phosphorylation of Smad4 and restores TGF-β growth-suppressive signaling in BCR-ABL1-positive leukemia显示文摘Loss of TGF-β-mediated growth suppression is a major contributor to the development of cancers,best exemplified by loss-offunction mutations in genes encoding components of the TGF-βsignaling pathway in colorectal and pancreatic cancers.Alternatively,gain-of-function oncogene mutations can also disrupt antiproliferative TGF-βsignaling.However,the molecular mechanisms underlying oncogene-induced modulation of TGF-βsignaling have not been extensively investigated.Here,we show that the oncogenic BCR-ABL1 of chronic myelogenous leukemia(CML)and the cellular ABL1 tyrosine kinases phosphorylate and inactivate Smad4 to block antiproliferative TGF-βsignaling.Mechanistically,phosphorylation of Smad4 at Tyr195,Tyr301,and Tyr322 in the linker region interferes with its binding to the transcription co-activator p300/CBP,thereby blocking the ability of Smad4 to activate the expression of cyclin-dependent kinase(CDK)inhibitors and induce cell cycle arrest.In contrast,the inhibition of BCR-ABL1 kinase with Imatinib prevented Smad4 tyrosine phosphorylation and re-sensitized CML cells to TGF-β-induced antiproliferative and pro-apoptotic responses.Furthermore,expression of phosphorylation-site-mutated Y195F/Y301F/Y322F mutant of Smad4 in Smad4-null CML cells enhanced antiproliferative responses to TGF-β,whereas the phosphorylation-mimicking Y195E/Y301E/Y322E mutant interfered with TGF-βsignaling and enhanced the in vivo growth of CML cells.These findings demonstrate the direct role of BCR-ABL1 tyrosine kinase in suppressing TGF-βsignaling in CML and explain how Imatinib-targeted therapy restored beneficial TGF-βanti-growth responses.Lijing Wang Shuchen Gu Fenfang Chen Yi Yu Jin Cao Xinran Li Chun Gao Yanzhen Chen Shuchong Yuan Xia Liu Jun Qin Bin Zhao Pinglong Xu Tingbo Liang Hongyan Tong Xia Lin Xin-Hua Feng 2023Signal Transduction and Targeted Therapy2023,8,4:0
