5Analysis of Disequilibrium Hybridization in Hybrid and Backcross Progenies of Saccharum officinarum × Erianthus arundinaceus显示文摘Erianthus arundinaceus is an important, closely related genus of Saccharum officinarum L. It is therefore important to understand how the chromosomes are transmitted when it hybridizes with sugarcane. The hybrids and backcross progenies of S. officinarum and E. arundinaceus and their parents were used for Karyotype analysis and to study the law of chromosome transmission. The results showed that the somatic chromosome number of both of the E. arundinaceus Hainan92-105 and Hainan92-77 were 2n = 60 = 60sm, belonging to type 1A, and the BC1 YC01-21 was 2n = 104 = 100m + 4sm, belonging to type 2C. The other six tested clones belonged to type 2B. The both F1s YC96-66 and YC96-40 that originated from Badila (2n = 80 = 70m + 10sm) with E. Arundinaceus were 2n = 70 = 68m + 2sm, which suggests an n + n transmission. The cross between YC96-66 (female parent) and CP84-1198 (male parent, 2n = 120 = 114m + 6sm) also followed the same genetic law and the somatic chromosome number of their progeny, YC01-3 (2n = 105 = 95m + 10sm). The cross derived from YC96-40 (female) and CP84-1198 (male), YC01-21 had 2n = 104 = 100m + 4sm chromosomes, following the same genetic law of n + n. However, YC01-36 had 2n = 132 = 130m + 2sm chromosomes, which suggests a 2n + n chromosome transmission. It can be inferred that the inheritance of chromosomes was very complex in the BC1. The difference in chromosome number between clones was as high as 28. This could be explained by the 2n + n transmission of chromosomes. 7Effects of neuregulin-1 on autonomic nervous system remodeling post-myocardial infarction in a rat model显示文摘Sympathetic nerve and vagus nerve remodeling play an important part in cardiac function post-myocardial infarction(MI). Increasing evidence indicates that neuregulin-1(NRG-1) improves cardiac function following heart failure. Since its impact on cardiac function and neural remodeling post-MI is poorly understood, we aimed to investigate the role of NRG-1 in autonomic nervous system remodeling post-MI. Forty-five Sprague-Dawley rats were equally randomized into three groups: sham(with the left anterior descending coronary artery exposed but without ligation), MI(left anterior descending coronary artery ligation), and MI plus NRG-1(left anterior descending coronary artery ligation followed by intraperitoneal injection of NRG-1(10 μg/kg, once daily for 7 days)). At 4 weeks after MI, echocardiography was used to detect the rat cardiac function by measuring the left ventricular end-systolic inner diameter, left ventricular diastolic diameter, left ventricular end-systolic volume, left ventricular end-diastolic volume, left ventricular ejection fraction, and left ventricular fractional shortening. m RNA and protein expression levels of tyrosine hydroxylase, growth associated protein-43(neuronal specific protein), nerve growth factor, choline acetyltransferase(vagus nerve marker), and vesicular acetylcholine transporter(cardiac vagal nerve fiber marker) in ischemic myocardia were detected by real-time PCR and western blot assay to assess autonomous nervous remodeling. After MI, the rat cardiac function deteriorated significantly, and it was significantly improved after NRG-1 injection. Compared with the MI group, m RNA and protein levels of tyrosine hydroxylase and growth associated protein-43, as well as choline acetyltransferase m RNA level significantly decreased in the MI plus NRG-1 group, while m RNA and protein levels of nerve growth factor and vesicular acetylcholine transporters, as well as choline acetyltransferase protein level slightly decreased. 14Comparison of outcomes after human leukocyte antigen-matched and haploidentical hematopoietic stem-cell transplantation for multiple myeloma显示文摘Background:Allogeneic stem-cell transplantation (SCT) is a well-established immunotherapeutic strategy for multiple myeloma (MM) with a potent and often sustained graft-vs.-myeloma effect.This multicenter investigation aimed to analyze the complications and survival of haploidentical SCT in patients with MM,and compare the main outcomes with matched-related donors (MRDs).Methods:Haploidentical and MRD SCT was identified from a cohort of 97 patients with MM who received a myeloablative transplantation in 13 hospitals from May 2001 to December 2017.A matched-pair analysis was designed.For each haplo recipient,the recipients were randomly selected from the MRD group and were matched according to the following criteria:year of the hematopoietic SCT (±2 years),disease status at transplantation,and the length of follow-up.ults:Seventy cases received MRD and 27 received haploidendcal transplantation.The two groups showed no significant ifferences regarding age,gender,cytogenetic risk,and diagnostic stage.The cumulative incidences of non-relapse mortality (NRM) at1 and 3 years based on donor type were 20.5%(95% confidence interval [CI],10.90-30.10%) and 24.2%(95% CI,13.81-34.59%) for the MRD group and 16.80%(95% CI,1.71-31.89%) and 28.70%(95% CI,8.71-48.69%) for the haplo group,respectively.Cumulative incidence of NRM did not differ significantly between the two groups (x2 =0.031,P =0.861).The cumulative incidences of progression-free survival (PFS) and 1 year and 3 years by type of donors were 59.8 %(95 % CI,48.24-71.36 %) and 45.4 %(95 % CI,33.44-57.36%),and 65.6%(95% CI,47.18-84.02%) and 26.8%(95% CI,7.59-46.01%) for MRD and haploidentical donor,respectively.Cumulative incidence of PFS did not differ significantly between the two groups (x2 =0.182,P =0.670).In multivariate analyses,no statistically significant differences were observed between haploidentical and MRD for relapse,NRM,PFS,and overall survival.There were no statistically differences on main outcomes after haploidentical and MRD.Conclusion:Haploidentical SCT could be performed safely and feasibly for patients with MM in need.Yao Chen Wei-Jun Fu Lan-Ping Xu Han-Yun Ren Yong-Rong Lai Dai-Hong Liu Lin Liu Zi-Min Sun Yuan-Bin Wu Xin Wang Ling-Hui Xia Ming Jiang Tong-Lin Hu Ding-Ming Wan Xiao-Jun Huang 2019Chinese Medical Journal2019,,15:4
15Rapid identification of Dendrobium officinale using Loop-Mediated Isothermal Amplification (LAMP) method显示文摘Dendrobium officinale is not only an ornamental plant, but also a valuable medicinal herb that is widely used in traditional Chinese medicine. However, distinguishing D. officinale from other Dendrobium species is usually a difficult task. In this study,we developed a rapid identification protocol for D. officinale using the loop-mediated isothermal amplification(LAMP) method. A set of primers were specifically designed to detect a modified internal transcribed spacer region of D. officinale at 65 ℃ within 40 min after adding SYBR~? Green I, which was used for the detection of D. officinale. Unlike commonly used adulterants, reaction mixtures containing D. officinale DNA changed from orange to green, and this color change was easily observed with the naked eye. Thus, this methodology can be used to accurately differentiate D. officinale from other Dendrobium species, is quick as all D. officinale samples were amplified within 40 min, and specific as samples of the adulterants were not amplified. The specificity of this LAMP-based method was confirmed by testing 17 samples of D. officinale and 32 adulterant samples from other Dendrobium species. 16Discovering small molecules as Wnt inhibitors that promote heart regeneration and injury repair显示文摘There are intense interests in discovering pro regenerative medicine leads that can promote cardiac differentiation and regeneration,as well as repair damaged heart tissues.We have combined zebrafish embryo-based screens with cardiomyogenesis assays to discover selective small molecules that modulate heart development and regeneration with minimal adverse effects.Two related compounds with novel structures,named as Cardiomogen 1 and 2(CDMG1 and CDMG2),were identified for their capacity to promote myocardial hyperplasia through expansion of the cardiac progenitor cell population.We find that Cardiomogen acts as a Wnt inhibitor by targeting p-catenin and reducing Tcf/Lef-mediated transcription in cultured cells.CDMG treatment of amputated zebrafish hearts reduces nuclear p-catenin in injured heart tissue,increases cardiomyocyte(CM)proliferation,and expedites wound healing,thus accelerating cardiac muscle regeneration.Importantly,Cardiomogen can alleviate the functional deterioration of mammalian hearts after myocardial infarction.Injured hearts exposed to CDMG1 display increased newly formed CMs and reduced fibrotic scar tissue,which are in part attributable to the^-catenin reduction.Our findings indicate Cardiomogen as a Wnt inhibitor in enhancing injury-induced CM proliferation and heart regeneration,highlighting the values of embryo-based small molecule screens in discovery of effective and safe medicine leads.Shuying Xie Wenbin Fu Guangju Yu Xueli Hu Kaa Seng Lai Xiangwen Peng Yating Zhou Xuejiao Zhu Ptamen Christov Leah Sawyer Terri T.Ni Gary A.Sulikowski Zhongzhou Yang Ethan Lee Chunyu Zeng Wei E.Wang Tao P.Zhong 2020Journal of Molecular Cell Biology2020,12,1:4

19Cgr1,a ripe rot resistance QTL in Vitis amurensis‘Shuang Hong’grapevine显示文摘Ripe rot is a serious grapevine disease in Vitis L.and Muscadinia(Planch.)Small.However,resistance to this disease has been reported in some oriental Vitis species.To identify resistance-related Quantitative Trait Loci(QTLs)from the Chinese grape species V.amurensis,an F1 population of V.vinifera‘Cabernet Sauvignon’×V.amurensis‘Shuang Hong’was used to map the ripe rot resistance loci expected in‘Shuang Hong’grape.A total of 7598 single nucleotide polymorphisms(SNPs)between the parents were identified in our previous study,and 934 SNPs were selected for genetic map construction.These SNPs are distributed across the 19 chromosomes covering a total of 1665.31 cM in length,with an average of 1.81 cM between markers.Ripe rot resistance phenotypes among the hybrids were evaluated in vitro using excised leaves for three consecutive years from 2016 to 2018;a continuous variation was found among the F1 hybrids,and the Pearson correlation coefficients of the phenotypes scored in all three years were significant at the 0.01 level.Notably,the first QTL reported for resistance to grape ripe rot disease,named Cgr1,was identified on chromosome 14 of‘Shuang Hong’grapevine.Cgr1 could explain up to 19.90%of the phenotypic variance.In addition,a SNP named‘np19345’was identified as a molecular marker closely linked to the peak of Cgr1 and has the potential to be developed as a marker for the Cgr1 resistance haplotype.Peining Fu Quanyou Tian Gongti Lai Rongfang Li Shiren Song Jiang Lu 2019Horticulture Research2019,6,1:3
