2Monitoring Intra-annual Spatiotemporal Changes in Urban Heat Islands in 1449 Cities in China Based on Remote Sensing显示文摘This study aimed to accurately study the intra-annual spatiotemporal variation in the surface urban heat island intensities(SUHIIs) in 1449 cities in China.First, China was divided into five environmental regions.Then, the SUHIIs were accurately calculated based on the modified definitions of the city extents and their corresponding nearby rural areas.Finally, we explored the spatiotemporal variation of the mean, maximum, and minimum values, and ranges of SUHIIs from several aspects.The results showed that larger annual mean daytime SUHIIs occurred in hot-humid South China and cold-humid northeastern China, and the smallest occurred in arid and semiarid west China.The seasonal order of the SUHIIs was summer > spring > autumn > winter in all the temperate regions except west China.The SUHIIs were obviously larger during the rainy season than the dry season in the tropical region.Nevertheless, significant differences were not observed between the two seasons within the rainy or dry periods.During the daytime, the maximum SUHIIs mostly occurred in summer in each region, while the minimum occurred in winter.A few cold island phenomena existed during the nighttime.The maximum SUHIIs were generally significantly positively correlated with the minimum SUHIIs during the daytime, nighttime and all-day in all environmental regions throughout the year and the four seasons.Moreover, significant correlation scarcely existed between the daytime and nighttime ranges of the SUHIIs.In addition, the daytime SUHIIs were also insignificantly correlated with the nighttime SUHIIs in half of the cases.LI Yuanzheng WANG Lan ZHANG Liping LIU Min ZHAO Guosong 2019Chinese Geographical Science2019,29,6:3
3Evaluation on monthly sea surface wind speed of four reanalysis data sets over the China seas after 1988显示文摘This study investigates the long-term changes of monthly sea surface wind speeds over the China seas from 1988 to 2015. The 10-meter wind speeds products from four major global reanalysis datasets with high resolution are used: Cross-Calibrated Multi-Platform data set(CCMP), NCEP climate forecast system reanalysis data set(CFSR),ERA-interim reanalysis data set(ERA-int) and Japanese 55-year reanalysis data set(JRA55). The monthly sea surface wind speeds of four major reanalysis data sets have been investigated through comparisons with the longterm and homogeneous observation wind speeds data recorded at ten stations. The results reveal that(1) the wind speeds bias of CCMP, CFSR, ERA-int and JRA55 are 0.91 m/s, 1.22 m/s, 0.62 m/s and 0.22 m/s, respectively.The wind speeds RMSE of CCMP, CFSR, ERA-int and JRA55 are 1.38 m/s, 1.59 m/s, 1.01 m/s and 0.96 m/s,respectively;(2) JRA55 and ERA-int provides a realistic representation of monthly wind speeds, while CCMP and CFSR tend to overestimate observed wind speeds. 5Definition of water exchange zone between the Bohai Sea and Yellow Sea and the effect of winter gale on it显示文摘The marine dynamic environment of the Bohai Sea and the Yellow Sea in the winter of 2006 is simulated by the Regional Ocean Modelling System(ROMS) marine numerical model. Using the simulated temperature and salinity, the water exchange zone between the Bohai Sea and Yellow Sea is defined through the Spectral Mixture Model(SMM). The influence of winter gales on the water exchange is also discussed. It is found that the Yellow Sea water masses in winter are distributed in a 'tongue' shape in the Bohai Strait region, the water exchange zone presents a zonal distribution along the margin of the 'tongue', with a tendency of running from northwest to southeast, and the water exchange is intensified at the tip of the 'tongue'. Besides, the coastal area in the northernmost Yellow Sea does not participate in the water exchange between the Bohai Sea and Yellow Sea. The result shows that the winter gale events play a role in enhancing the water exchange. It is specifically shown by the facts: the Yellow Sea warm current is enhanced to intrude the Bohai Sea by the gale process; the water exchange zone extends into the Bohai Sea; the water exchange belt in the southern part becomes wider; the mixture zone of river runoff with the Bohai Sea water upon its entry is enlarged and shifts northwards. 7Causes of seasonal sea level anomalies in the coastal region of the East China Sea显示文摘Based on the analysis of sea level, air temperature, sea surface temperature(SST), air pressure and wind data during 1980–2013, the causes of seasonal sea level anomalies in the coastal region of the East China Sea(ECS) are investigated. The research results show:(1) sea level along the coastal region of the ECS takes on strong seasonal variation. The annual range is 30–45 cm, larger in the north than in the south. From north to south, the phase of sea level changes from 140° to 231°, with a difference of nearly 3 months.(2) Monthly mean sea level(MSL)anomalies often occur from August to next February along the coast region of the ECS. The number of sea level anomalies is at most from January to February and from August to October, showing a growing trend in recent years.(3) Anomalous wind field is an important factor to affect the sea level variation in the coastal region of the ECS. Monthly MSL anomaly is closely related to wind field anomaly and air pressure field anomaly. Wind-driven current is essentially consistent with sea surface height. In August 2012, the sea surface heights at the coastal stations driven by wind field have contributed 50%–80% of MSL anomalies.(4) The annual variations for sea level,SST and air temperature along the coastal region of the ECS are mainly caused by solar radiation with a period of12 months. But the correlation coefficients of sea level anomalies with SST anomalies and air temperature anomalies are all less than 0.1.(5) Seasonal sea level variations contain the long-term trends and all kinds of periodic changes. Sea level oscillations vary in different seasons in the coastal region of the ECS. In winter and spring, the oscillation of 4–7 a related to El Ni?o is stronger and its amplitude exceeds 2 cm. In summer and autumn, the oscillations of 2–3 a and quasi 9 a are most significant, and their amplitudes also exceed 2 cm. The height of sea level is lifted up when the different oscillations superposed. 10Overexpression of the diguanylate cyclase CdgD blocks developmental transitions and antibiotic biosynthesis in Streptomyces coelicolor显示文摘Cyclic dimeric GMP(c-di-GMP)has emerged as the nucleotide second messenger regulating both development and antibiotic production in high-GC,Gram-positive streptomycetes.Here,a diguanylate cyclase(DGC),CdgD,encoded by SCO5345 from the model strain Streptomyces coelicolor,was functionally identified and characterized to be involved in c-di-GMP synthesis through genetic and biochemical analysis.cdgD overexpression resulted in significantly reduced production of actinorhodin and undecylprodigiosin,as well as completely blocked sporulation or aerial mycelium formation on two different solid media.In the cdgD-overexpression strain,intracellular c-di-GMP levels were 13-27-fold higher than those in the wild-type strain.In vitro enzymatic assay demonstrated that CdgD acts as a DGC,which could efficiently catalyze the synthesis of c-di-GMP from two GTP molecules.Heterologous overproduction of cdgD in two industrial Streptomyces strains could similarly impair developmental transitions as well as antibiotic biosynthesis.Collectively,our results combined with previously reported data clearly demonstrated that c-di-GMP-mediated signalling pathway plays a central and universal role in the life cycle as well as secondary metabolism in streptomycetes.Xiaocao Liu Guosong Zheng Gang Wang Weihong Jiang Lei Li Yinhua Lu 2019Science China(Life Sciences)2019,62,11:2
11Multiplex genome editing using a dCas9-cytidine deaminase fusion in Streptomyces显示文摘CRISPR/Cas-mediated genome editing has greatly facilitated the study of gene function in Streptomyces. However, it could not be efficiently employed in streptomycetes with low homologous recombination(HR) ability. Here, a deaminase-assisted base editor d Cas9-CDA-ULstr was developed in Streptomyces, which comprises the nuclease-deficient Cas9(dCas9), the cytidine deaminase from Petromyzon marinus(PmCDA1), the uracil DNA glycosylase inhibitor(UGI) and the protein degradation tag(LVA tag). Using d Cas9-CDA-ULstr , we achieved single-, double-and triple-point mutations(cytosine-to-thymine substitutions)at target sites in Streptomyces coelicolor with efficiency up to 100%, 60% and 20%, respectively. This base editor was also demonstrated to be highly efficient for base editing in the industrial strain, Streptomyces rapamycinicus, which produces the immunosuppressive agent rapamycin. Compared with base editors derived from the cytidine deaminase rAPOBEC1, the PmCDA1-assisted base editor dCas9-CDA-ULstr could edit cytosines preceded by guanosines with high efficiency, which is a great advantage for editing Streptomyces genomes(with high GC content). Collectively, the base editor dCas9-CDA-ULstr could be employed for efficient multiplex genome editing in Streptomyces. 13Chromosome-level genomes of seeded and seedless date plum based on third-generation DNA sequencing and Hi-C analysis显示文摘Diospyros lotus L.(Date plum)is an important tree species that produces fruit with a high nutritional value.An accurate chromosomal assembly of a species facilitates research on chromosomal evolution and functional gene mapping.In this study,we assembled the first chromosome-level genomes of seeded and seedless D.lotus using Illumina short reads,PacBio long reads,and Hi-C technology.The assembled genomes comprising 15 chromosomes were 617.66 and 647.31 Mb in size,with a scaffold N50 of 40.72 and 42.67 Mb for the seedless and seeded D.lotus,respectively.A BUSCO analysis revealed that the seedless and seeded D.lotus genomes were 91.53% and 91.60%complete,respectively.Additionally,20,689(95.4%)and 22,844(98.5%)protein-coding genes in the seedless and seeded D.lotus genomes were annotated,respectively.Comparisons of the chromosomes between genomes revealed inversions and translocations on chromosome 8 and inversions on chromosome 11.We identified 490 and 424 gene families that expanded in the seedless and seeded D.lotus,respectively.The enriched pathways among these gene families included the estrogen signaling pathway,the MAPK signaling pathway,and biosynthetic pathways for flavonoids,monoterpenoids,and glucosinolates.Moreover,we constructed the first Diospyros genome database(http://gffzzc8eabe692799476bhnncocnwnp90n6kv0.ffgz.tsg.suse.edu.cn).On the basis of our data,we developed the first high-quality annotated D.lotus reference genomes,which will be useful for genomic studies on persimmon and for clarifying the molecular mechanisms underlying important traits.Comparisons between the seeded and seedless D.lotus genomes may also elucidate the molecular basis of seedlessness.Weitao Mao Guoxin Yao Shangde Wang Lei Zhou Guosong Chen Ningguang Dong Guanglong Hu 2021Forestry Research2021,1,1:1
14Statistical characteristics and mechanisms of mesoscale eddies in the North Indian Ocean显示文摘The statistical characteristics and mechanisms of mesoscale eddies in the North Indian Ocean are investigated by adopting multi-sensor satellite data from 1993 to 2019.In the Arabian Sea(AS),seasonal variation of eddy characteristics is remarkable,while the intraseasonal variability caused by planetary waves is crucial in the Bay of Bengal(BOB).Seasonal variation of the eddy kinetic energy(EKE)is distinct along the west boundary of AS,especially in the Somali Current region.In the BOB,larger EKE occurs at the northwest basin from March to May,to the east of Sri Lanka from June to September,and along the east coast of India from November to December.The wind stress work(WW)is further studied to figure out the direct influence of wind forcing on EKE.The WW exerts positive effects on EKE along the west boundary of AS and in the south of India/Sri Lanka during the two monsoon seasons.Besides,the WW also has impact on EKE along the east coast of India in November and December.Eventually,we investigate the characteristics and the driving mechanisms of long lifespan eddies.In the AS,long lifespan anti-cyclonic eddies(AEs)mainly generate in the Socotra,the West Indian Coastal Current and the East Arabian Current regions,while cyclonic eddies(CEs)are concentrated in the northwest region.In the BOB,long lifespan AEs mostly form near the west of Myanmar,while CEs are accumulated at the north and northwest basin.The instabilities caused by Rossby waves,coastal Kelvin waves,seasonal currents,together with wind stress forcing exert enormous efforts on the generation and evolution of these eddies.Chunjian Sun Xidong Wang Anmin Zhang Lianxin Zhang Caixia Shao Guosong Wang 2022Acta Oceanologica Sinica2022,41,5:1
15A harlequin ichthyosis pig model with a novel ABCA12 mutation can be rescued by acitretin treatment显示文摘Harlequin ichthyosis (HI) is a severe genetic skin disorder and caused by mutation in the ATP-binding cassette A12 (ABCA12) gene. The retinoid administration has dramatically improved long-term survival of HI, but improvements are still needed. However, the ABCA12 null mice failed to respond to retinoid treatment, which impedes the development of novel cure strategies for HI. Here we generated an ethylnitrosourea mutagenic HI pig model (named Z9), which carries a novel deep intronic mutation IVS49-727 A>G in the ABCA12 gene, resulting in abnormal mRNA splicing and truncated protein production. Z9 pigs exhibit significant clinical symptom as human patients with HI. Most importantly, systemic retinoid treatment significantly prolonged the life span of the mutant pigs via improving epidermal maturation, decreasing epidermal apoptosis, and triggering the expression of ABCA6.
