| 1 | Critical size limit of biodegradable nanopartides for enhanced lymph node trafficking and paracortex penetration显示文摘Lymph node (LN) targeti ng through interstitial drain age of nan oparticles (NPs) is an attractive strategy to stimulate a pote nt immune respo nse, as LNs are the primary site for lymphocyte priming by antigen presenting cells (APCs) and triggering of an adaptive immune response. NP size has been shown to influence the efficiency of LN-targeting and retention after subcutaneous injection. For clinical translation, biodegradable NPs are preferred as carrier for vaccine delivery. However, the selective 'size gateM for effective LN-drainage, particularly the kinetics of LN trafficking, is less well defined. This is partly due to the challenge in generating size-controlled NPs from biodegradable polymers in the sub-100-nm range. Here, we report the preparation of three sets of poly(lactic-co-glycolic)-b-poly(ethylene-glycol)(PLGA-b-PEG) NPs with number average diameters of 20-, 40-, and 100-nm and narrow size distributions using flash nanoprecipitation. Using NPs labeled with a near-infrared dye, we showed that 20-nm NPs drain rapidly across proximal and distal LNs following subcutaneous inoculation in mice and are retai ned in LNs more effectively than NPs with a nu mber average diameter of 40-nm. The drain age of 100-nm NPs was n egligible. Furthermore, the 20-nm NPs showed the highest degree of penetration around the paracortex region and had enhanced access to dendritic cells in the LNs. Together, these data confirmed that small, size-controlled PLGA-b-PEG NPs at the lower threshold of about 30-nm are most effective for LN trafficking, retention, and APC uptake after s.c. administration. This report could inform the design of LN-targeted NP carrier for the delivery of therapeutic or prophylactic vaccines. | Gregory P. Howard Garima Verma Xiyu Ke Winter M. | 2 | pharmacognostical study and establishment of quality parameters of aerial parts of Costus speciosus-a well known tropical folklore medicine显示文摘Objective:To evaluate the diagnostic pharmacognostical characters of Costus speciosus(aerial parts)along with their physico-chemical parameters and fluorosence analysis.Method:The pharmacognostical characters were determined in terms of macroscopy, microscopy, powder microscopy, leaf constant, fluorescence analysis and preliminary phytochemical investigation. Results:The findings of macroscopy revealed that leaves elliptic to oblong or oblong-lancoelate,thick,spirally arranged, with stem clasping sheaths up to 4 cm,flowers large, white, conelike terminal spikes, with bright red bracts.Transverse section of leaflet showed the presence of cuticularised epidermis with polygonal cells on adaxial surface and bluntly angled cells on abaxial surface of lamina, mesophyll cells differentiated in to single layered palisade cells on each surface and 2-3 layered spongy parenchyma, unicellular and uniseriate multicellular covering trichomes, paracytic stomata and vascular bundles surrounded by sclerenchymatous multicellular sheath.Preliminary phytochemical screening exhibited the presence of various phytochemical groups like alkaloids, glycosides, steroids, phenolic constituents.Further, the leaf constants, powder microscopy and fluorescence characteristics indicated outstanding results from this investigation Conclusions:Various pharmacognostical and physico-chemical parameters have pivotal roles in identification, authentication and establishment of quality parameters of the species. | Pradeep Singh Ratan Lal Khosa Shruti Srivastava Garima Mishra Keshri Kishor Jha Sourabh Srivastava Sangeeta Ramesh Kumar Verma Mohd Adil Tahseen | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,6: | 0 |
| 3 | Fuzzy Based Ant Colony Optimization Scheduling in Cloud Computing显示文摘Cloud computing is an Information Technology deployment model established on virtualization.Task scheduling states the set of rules for task allocations to an exact virtual machine in the cloud computing environment. However, task scheduling challenges such as optimal task scheduling performance solutions, are addressed in cloud computing.First, the cloud computing performance due to task scheduling is improved by proposing a Dynamic Weighted Round-Robin algorithm. This recommended DWRR algorithm improves the task scheduling performance by considering resource competencies, task priorities, and length.Second,a heuristic algorithm called Hybrid Particle Swarm Parallel Ant Colony Optimization is proposed to solve the task execution delay problem in DWRR based task scheduling. In the end,a fuzzy logic system is designed for HPSPACO that expands task scheduling in the cloud environment.A fuzzy method is proposed for the inertia weight update of the PSO and pheromone trails update of the PACO.Thus, the proposed Fuzzy Hybrid Particle Swarm Parallel Ant Colony Optimization on cloud computing achieves improved task scheduling by minimizing the execution and waiting time, system throughput, and maximizing resource utilization. | K.Rajakumari M.Vinoth Kumar Garima Verma S.Balu Dilip Kumar Sharma Sudhakar Sengan | 2022 | Computer Systems Science & Engineering2022,40,2: | 0 |
