1The effect of exposure to normbaric hypoxia on the body weight in rats显示文摘为了确定大鼠的体重是否在标准气压缺氧环境下会减轻,本实验将12只5周龄Wis-tar大鼠移高至海拔2260m并使之持续暴露于含10%O2的低氧小低压舱内(相当于在海拔5000m)20日。在基线每5日测一次体重。由于标准气压缺氧环境可影响血循环功能,所以在大鼠移入高原前后20天,分别对其肺动脉压和血红蛋白浓度进行了测定。基线处体重为(222.4±20.7)g,而与基线处相比。大鼠体重在移高5、10、15和20天后明显下降,分别下降了(4.88±SD)%、(7.67土SD)%、(11.05±SD)%和(11.9±SD)%,差异显著(P<0.05)。基线处大鼠的平均肺动脉压为(21.3±2.5)mmHg,低氧暴露20天后肺动脉压明显增高[(45.3±5.8)mmHg,P<0.001]。血红蛋白浓度在基线处的平均值为(14.5±3.2)g/L,而在低氧暴露20天后明显增加[(22.6±5.4)g/L,P<0.001]。该结果显示,模拟高海拔可使大鼠体重明显下降,这与先前报道的人体试验结果相似。结果也进一步说明,标准气压下持续的低氧环境对肺动脉压和血红蛋白浓度可产生实质性影响。Yi Lina Keishi Kubo Ge Rili Benjamin Levine Tony Babb 2005青海医学院学报2005,26,3:6
2Effect of aqueous extract of Sanweitanxiang powder on calcium homeostasis protein expression in ischemic-reperfusion injury rat heart显示文摘OBJECTIVE: To investigate the underlying mechanism of reduced myocardial ischemia-reperfusion (I/R) injury in rats using the traditionalTibetan medicine Sanweitanxiang powder (SWTX). METHODS: Rats were randomly divided into six groups (n=10) as follows: (a) propranolol dinitrate control group, given propranolol dinitrate 0.02 g/kg for 10 days before I/R, (b) SWTX with a high dose group, given SWTX 1.5 g/kg for 10 days before I/R, (c) SWTX with a medium dose group, given SWTX 1.25 g/kg for 10 days before I/R, (d) sham group (Sham), in which the rat heart was exposed by pericardiotomy but without I/R, (e) SWTX with a low dose group, given SWTX 1.0 g/kg for 10 days before I/R, and (f) I/R injury group. Rats were intragastrically pretreated with propranolol dinitrate orSWTX. After that, the operation to cause ischemia and reperfusion was conducted.The histopathologic changes of rat hearts were observed by hematoxylin and eosin staining and transmission electron microscopy. Ca2+ homeostasis protein expression was determined by western blot. RESULTS: After SWTX pretreatment, the development of ultrastructural pathological changes from IR injury was attenuated. A decrease in the expression of B-cell lymphoma 2 associated X protein, and an increase in the expression of B-cell lymphoma 2 were observed. An increased activation of extracellular signal regulated kinases were found. Compared with the sham group, the expression of sarcoplasmic reticulum calcium-ATPase, phospholamban, and calsequestrin were all up-regulated after pretreatment with SWTX. 5The Difference of the Pulmonary-Artery Pressure of the Migrantsfrom Difference Elevation at the 4636~4907 m Altitude显示文摘Study Objective: To compare and analyze the changes of the pulmonary-artery pressure of the migrants coming from different elevation in the hypoxic environment of 4636~4907 m extreme altitude. To explore the susceptibility to hypoxic pulmonary-artery hypertension (PH) in the subjects from different altitude and profession.Methods: By using Color Doppler Ultrasonography (CDU), measuring the pulmonary-artery pressure of 207 healthy men, who had continuously being lived and worked at the extreme altitude for more than six months, and then were divided into three groups according to their profession and the altitude of original living place. Results: There was a significant difference in the outcomes of pulmonary-artery pressure from the 3 groups. Conclusions: Altitude of original living place, labor intensity are some of factors that impact the pulmonary-artery pressure of the people who exposure to a hypoxic environment. The pulmonary-artery pressure of person without strenuously physical work experience was more sensitive to hypoxic surroundings than that of labor workers. It was not always the fact at an extreme altitude that the moderate altitude mountaineers were superior to other migrants from a lower altitude or plain. The higher PH was found in the groups of the moderate altitude mountaineers and labor workers. 6Study of Treating HAPE with HBO in Situ at the Altitude of 4636m显示文摘Study Objective: To evaluate the treatment effect of HAPE with HBO in situ at an extreme altitude of 4636 m. To investigate the relationship between pulmonary hypertension (PH) and HAPE. To emphasize the importance of the treatment in situ.Methods: The 32 patients from the plateau of 4636~5130 m (the barometric pressure: 57.41~53.28 kPa/431.6~400.6 mmHg(1 mmHg=133.3224 Pa), the partial pressure of oxygen: 12.0~11.6 kPa/90.0~87.0 mmHg) were treated in situ of 4636 m altitude with HBO. Before and after the treatment, the clinical symptoms / signs, radiographic evidence, SaO2, and MPAP were compared.Results: The clinical symptoms/signs, and all the targets of the patients were improved dramatically (p<0.001).Conclusions: Treating HAPE with HBO is the most effective method among various therapies in situ at an extreme altitude. The pulmonary -artery pressure was significantly decreased. All cases showed improved immediately. Among the subjects, 11 cases (34.3%) were cured at once. The therapy made it possible for patients to get further treatment at a lower altitude area with prospective good results. We suggested that treating HAPE with HBO should be the first choice in situ. There must be PH in the patients of HAPE, but the reverse is not the true. The PH is a mechanism of normal compensation of the body exposing in the hypoxic environment. 7Clinical Analysis of GPT Alteration of Constructors for Qinghai-Tibetan Railroad at Different Altitude Areas显示文摘Objective: To observe the effect of hypoxia on the liver function of constructors at high altitude. Method: Select 221 constructors aged 23~42 as subjects, who had worked for 3~6 months at 4500 m. Collect venous blood from fasting subjects and determine GPT by new Lai’s Method from “The Nationwide Clinical Inspection Operating Rules'. Results: The study indicated that the GPT of 45 subjects among Group 1 were abnormal, with the proportion of 20.3%. The peak of GPT appeared when subjects exposed to high altitude of 4500m for 1 month, and the mean value was 83.76±9.23.The GPT of workers were higher than that of administers at different altitude (p<0.05). And the GPT of subjects at 4500 m were higher than those at 3500 m (p<0.05). We can find a degrading tendency with the increasing of working time (p<0.05). There was significantly statistic difference between two groups. Conclusions: The study confirmed that hypoxia could cause the change of liver function mildly and moderately. And there was a positive correlation between the degree of hypoxia and liver function. But, the change began to become a decreasing trend when people went on working longer. We also found that the change of liver structure and function at high altitude correlated with several factors else, including labor, individual difference, methods and sensitivity to the test. 8Effects of hypoxia on mRNA expression of housekeeping genes in rat brain tissue and primary cultured neural cells显示文摘Internal standards are critical for quantitative RNA analyses.Housekeeping genes are often used as internal standards with the assumption that their express-ion levels remain relatively constant in different experi-mental conditions.In this study,four commonly used housekeeping genes,Glyceraldehyde-3-phosphate dehy-drogenase(GAPDH),β-actin,28S rRNA and 18S rRNA were selected to test whether this assumption is tenable under hypoxic conditions.We tested the RNA expression level of these four genes in different hypoxic conditions.Rats subjected to acute hypoxia for 2 hours were used for tissue detection.Primary cultured neural stem cells from E13 fetal rats were treated with 3%O_(2) or 10%O_(2) for 24 hours for in vitro experiments.In both experiments,expression levels of 28S rRNA and 18S rRNA were constant,independent of hypoxia types.However,expression levels of GAPDH and b-actin were all changed in all kinds of hypoxic conditions.In particu-lar,the mRNA expression level of GAPDH was increased by 43.4%under 3%O_(2) hypoxic conditions.These results suggest that 18S rRNA and 28S rRNA are reliable internal controls for comparative analyses of transcrip-tion under hypoxia.GAPDH appears particularly unfa-vorable for this purpose in hypoxic conditions.Yingzhong YANG Wenhong FAN Lingling ZHU Tong ZHAO Lan MA Yan WU Rili GE Ming FAN 2008Frontiers of Medicine2008,2,3:0
