| 3 | Changes of NF-kB,p53,Bcl-2 and caspase in apoptosis induced by JTE-522 in human gastric adenocarcinoma cell line AGS cells:role of reactive oxygen species显示文摘AIM: To identify whether JTE-522 can induce apoptosis inAGS cells and ROS also involved in the process, and toinvestigate the changes in NF-kB, p53, bcl-2 and caspase in the apoptosis process.METHODS: Cell culture, MTT, Electromicroscopy, agarosegel electrophoresis, lucigenin, Western blot andelectrophoretic mobility shift assay (EMSA) analysis wereemployed to investigate the effect of JTE-522 on cellproliferation and apoptosis in AGS cells and relatedmolecular mechanisms.RESULTS: JTE-522 inhibited the growth of AGS cells andinduced the apoptosis. Lucigenin assay showed the generationof ROS in cells under incubation with JTE-522. The inc eareasedROS generation might contribute to the induction of AGS cellsto apoptosis. EMSA and Westem blot revealed that NF-kBactivity was almost completely inhibited by preventing thedegradation of IkBα. Additionally, by using Western blot weconfirmed that the level of bcl-2 was decreased, whereas p53showed a great increase following JTE-522 treatment. | 5 | Changes in the level of serum liver enzymes after laparoscopic surgery显示文摘AIM: The purpose of this study was to investigate the effectof laparoscopic surgery on liver function in humans and thepossible mechanisms behind such effect.METHODS: Blood samples from 286 patients whounderwent laparoscopic cholecystectomy (LC) and 40patients who underwent open cholecystectomy (OC) weretested for liver function by measuring the level of serumalanine aminotrasferase (ALT) and aspartate aminotrasferase(AST) before and after the operations. The same tests werealso applied to 18 laparoscopic colorectal cancer resection(LCR) patients and 23 open colorectal cancer resection (OCR)patients to determine whether CO2 pneumoperitoneum couldalter the serum liver enzymes.RESULTS: The level of serum ALT and AST increasedsignificantly during the first 48 hours post operations in bothLC and LCR patients. However, no significant change of theserum liver enzymes was detected in both OC and OCRpatients. As a result, there was statistically significantdifference in change of both ALT and AST levels betweenLC and OC patients and LCR and OCR patients, respectively.By the 7th day post operation, the level of both enzymesreturned to normal values in LC, OC and OCR patients exceptLCR patients whose enzymes remained at a higher level.CONCLUSION: Transient elevation of hepatic transaminasesoccurred after laparoscopic surgery. The major causativefactor seemed to be the CO2 pneumoperitoneum. In mostof the laparoscopic surgery patients, the transient elevationof serum liver enzymes showed no apparent clinicalimplications. | 6 | Treatment outcomes for different subgroups of nasopharyngeal carcinoma patients treated with intensity-modulated radiation therapy显示文摘Although many studies have investigated intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC),sample sizes in the reported studies are usually small and different in outcomes in different T and N subgroups are seldom analyzed.Herein,we evaluated the outcomes of NPC patients treated with IMRT and further explored treatment strategy to improve such outcome.We collected clinical data of 865 NPC patients treated with IMRT alone or in combination with chemotherapy,and classified all cases into the following prognostic categories according to different TNM stages:early stage group (T1-2N0-1M0),advanced local disease group (T3-4N0-1M0),advanced nodal disease group (T1-2N2-3M0),and advanced locoregional disease group (T3-4N2-3M0).The 5-year overall survival (OS),local relapse-free survival (LRFS),and distant metastases-free survival (DMFS) were 83.0%,90.4%,and 84.0%,respectively.The early disease group had the lowest treatment failure rate,with a 5-year OS of 95.6%.The advanced local disease group and advanced nodal disease group had similar failure pattern and treatment outcomes as well as similar hazard ratios for death (4.230 and 4.625,respectively).The advanced locoregional disease group had the highest incidence of relapse and death,with a 5-year DMFS and OS of 62.3% and 62.2%,respectively,and a hazard ratio for death of 10.402.Comparing with IMRT alone,IMRT in combination with chemotherapy provided no significant benefit to locoregionally advanced NPC.Our results suggest that the decision of treatment strategy for NPC patients should consider combinations of T and N stages,and that IMRT alone for early stage NPC patients can produce satisfactory results.However,for advanced local,nodal,and locoregional disease groups,a combination of chemotherapy and radiotherapy is recommended. | Sheng-Fa Su 1,2,3,Fei Han 1,2,Chong Zhao 1,2,Ying Huang 1,2,Chun-Yan Chen 1,2,Wei-Wei Xiao 1,2,Jia-Xin Li 1,2 and Tai-Xiang Lu 1,2 1 State Key Laboratory of Oncology in South China,Guangzhou,Guangdong 510060,P.R.China 2 Department of Radiation Oncology,Sun Yat-sen University Cancer Center,Guangzhou,Guangdong 510060,P.R.China 3 Department of Oncology,Guiyang Medical College Hospital,Tumor Hospital of Guizhou,Guiyang,Guizhou 550003,P.R.China. | 2011 | Chinese Journal of Cancer2011,30,8: | 40 |
| 7 | Percutaneous cryoablation in combination with ethanol injection for unresectable hepatocellular carcinoma显示文摘AIM: To evaluate the effectiveness and safety of percutaneous hepatic cryoablation in combination with percutaneous ethanol injection (PEI) in patients with unresectable hepaocellular carcinoma (HCC).METHODS: A total of 105 masses in 65 HCC patients underwent percutaneous hepatic cryoablation. The cryoablation was performed with the Cryocare system (Endocare, Irvine, CA, USA) using argon gas as a cryogen.Two freeze-thaw cycles were performed, each reaching a temperature of -180 ℃ at the tip of the probe. PEI was given in 36 patients with tumor masses larger than 6 cm in diameter 1-2 weeks after cryoablation and then once per week for 4 to 6 sessions. The efficacy was evaluated with survival, change of tumor size and alpha-fetoprotein (AFP) levels. RESULTS: During a follow-up duration of 14 months in average with a range of 5 to 21 months, 33 patients (50.8 %)were free of tumors, 22 patients (33.8 %) alive with tumor recurrence: two had bone metastases, three were found to have lung metastases, and the remaining 17 recurrences occurred in the liver, of whom only 3 developed a cryosite recurrence. Among the 41 patients who were followed up for more than one year, 32(78 %) were alive despite of tumor recurrence. Seven patients (10.8 %) died due to disease recurrence. Three patients (4.6 %) died due to some noncancer-related causes. Among the 43 patients who had a CT scan available for review, 38 (88.4 %) had a shrinkage of tumor mass. Among the 22 patients who received biopsies of cryoablated tumor mass, all biopsies except one, showed only dead or scar tissues. Of the patients who had an increased AFP preablatively, 91.3 % had a decrease of AFP to normal or nearly normal levels during postablative 3-6 months. Complications of cryoablation included liver capsular cracking in one patient,transient thrombocytopenia in 4 patients and asymptomatic right-sided pleural effusions in 2 patients. | 8 | Galangin induces apoptosis of hepatocellular carcinoma cells via the mitochondrial pathway显示文摘AIM:To investigate the mechanism by which galangin,a polyphenolic compound derived from medicinal herbs,induces apoptosis of hepatocellular carcinoma(HCC) cells.METHODS:The 3-(4,5-Dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay was used to measure cell viability.Apoptosis was evaluated by in situ uptake of propidium iodide and Hoechst 33258 and was then detected by fluorescence microscopy.Protein expressions were detected by Western blotting.To confirm the apoptotic pathway mediated by galangin,cells were transfected by bcl-2 gene to overexpress Bcl-2 or siRNA to down-regulate Bcl-2 expression.RESULTS:Galangin(46.25-370.0 μmol/L) exerted an anti-proliferative effect,induced apoptosis,and decreased mitochondrial membrane potential in a dose and time-dependent manner.Treatment with galangin induced apoptosis by translocating the pro-apoptotic protein Bax to the mitochondria,which released apoptosis-inducing factor and cytochrome c into the cytosol.Overexpression of Bcl-2 attenuated galangin-induced HepG2 cell apoptosis,while decreasing Bcl-2 expression enhanced galangin-induced cell apoptosis.CONCLUSION:Our data suggests that galangin mediates apoptosis through a mitochondrial pathway,and may be a potential chemotherapeutic drug for the treatment of HCC. | Hai-Tao Zhang Liu-Bo Lan Da-Hua Fan Kai-Dan Zhu Xiao-Yi Chen Min Wen Hui-Ming Liu Guangdong Province China Hui Luo | 2010 | World Journal of Gastroenterology2010,16,27: | 32 |

| 10 | Mechanism of the Spring Persistent Rains over southeastern China显示文摘The Spring Persistent Rains (SPR) in the areas to the south of middle and lower reaches of the Yangtze River or over southeastern China (SEC) is a unique synoptic and climatic phenomenon in East Asia. This study reveals a possible mechanism responsible for the climatic cause of SPR formation through climatic mean data analysis and sensitive numerical model experiments. SEC is located at the down-stream of the southwesterly velocity center (SWVC) which lies on the southeastern flank of the Tibetan Plateau (TP). As a result, there are strong southwesterly wind velocity convergence and moisture con-vergence over SEC. This is the immediate climatic cause of SPR formation. In spring, the seasonal evolution of the southwesterly velocity consists with the surface sensible heating over southeastern TP, indicating that the formation of SPR is related to not only the southwesterly wind of mechanical de-flected flow of TP, but also the southwesterly wind of thermal-forced cyclonic low circulation. Sensitive numerical experiments demonstrate that, without TP, both SWVC and the SPR rain belt will disappear. The southwesterly wind velocity increases almost linearly with the amount of the total diabatic heating with TP rising. Therefore, SWVC is the result of the mechanical forcing and thermal forcing of TP. | 12 | (-)-Epigallocatechin-3-gallate inhibits growth of gastric cancer by reducing VEGF production and angiogenesis显示文摘AIM: To investigate the effect of (-)-epigallocatechin- 3-gallate (EGCG) on growth of gastric cancer and its possible mechanism. METHODS: Heterotopic tumors were induced by subcutaneously injection of SGC-7901 cells in nude mice. Tumor growth was measured by calipers in two dimensions. Tumor angiogenesis was determined with tumor microvessel density (MVD) by immunohistology. Vascular endothelial growth factor (VEGF) protein level and activation of signal transducer and activator of transcription 3 (Stat3) were examined by Western blotting. VEGF mRNA expression was determined by RT-PCR and VEGF release in tumor culture medium by ELISA. VEGF-induced cell proliferation was studied by MTT assay, cell migration by gelatin modified Boyden chamber (Transwell) and in vitro angiogenesis by endothelial tube formation in Matrigel. RESULTS: Intraperitoneal injection of EGCG inhibited the growth of gastric cancer by 60.4%. MVD in tumor tissues treated with EGCG was markedly reduced. EGCG treatment reduced VEGF protein level in vitro and in vivo. Secretion and mRNA expression of VEGF in tumor cells were also suppressed by EGCG in a dose-dependent manner. This inhibitory effect was associated with reduced activation of Stat3, but EGCG treatment did not change the total Stat3 expression. EGCG also inhibited VEGF-induced endothelial cell proliferation, migration and tube formation. | 14 | Relationship between expression of Smac and Survivin and apoptosis of primary hepatocellular carcinoma显示文摘BACKGROUND: T he second mitochondria-derived activator of caspase/direct inhibitor of apoptosis-binding protein with low pI (Smac/DIABLO) was recently identified as a protein that is released from mitochondria in response to apoptotic stimuli and promotes apoptosis by antagonizing inhibitor of apoptosis proteins. Furthermore, Smac/DIABLO plays an important regulatory role in the sensitization of cancer cells to both immune- and drug-induced apoptosis. However, little is known about the clinical significance of Smac/DIABLO in various cancers including hepatocellular carcinoma (HCC). This study was undertaken to investigate the expression of Smac and Survivin and their relationship with the apoptosis in primary HCC. METHODS: The expression of Smac and Survivin proteins was evaluated by immunohistochemistry. The mRNA expression of Smac and Survivin was evaluated by reverse transcription-polymerase chain reaction (RT-PCR) in HCC tissues of 50 patients, para-carcinoma tissues of 20 patients, and normal liver tissues of 15 patients. RESULTS: Smac mRNA was detected by RT-PCR in HCC tissues of 21 (42.0%) of the 50 patients, para-carcinoma tissues of 19 (95.0%) of the 20 patients, and normal liver tissues of 15 (100%) of the 15 patients. Survivin mRNA was found in HCC tissues of 46 of the 50 patients, para- carcinoma tissues of 2 of the 20 patients, and normal liver tissues of 0 of 15 patients. Immunohistochemistry revealed Smac protein in HCC tissues of 20 patients (40.0%), in para-carcinoma tissues of 18 patients (90.0%), and normal liver tissues of 15 patients (100.0%). The expression of Smac was significantly different in HCC tissues and non- HCC tissues. Survivin protein was found in HCC tissuesin 45 patients, para-carcinoma tissues in 2 patients, and normal liver tissues in none of the patients. The expression of Survivin was significantly different in HCC tissues and non-HCC tissues. | 18 | Effect of a cancer vaccine prepared by fusions of hepatocarcinoma cells with dendritic cells显示文摘AIM To prepare a cancer vaccine (H22-DC) expressing high levels of costimulatory molecules based on fusions of hepatocarcinoma cells ( H22 ) with dendritic cells (DC) of mice and to analyze the biological characteristics and induction of specific CTL activity of H22-DC.``METHODS DCs were isolated from murine spleen by metrizamide density gradient centrifugation, purified based on its characteristics of semi-adhesion to culture plates and FcR , and were cultured in the medium containing GM. CSF and IL-4. A large number of DC were harvested. DCs were then fused with H22 cells by PEG and the fusion cells were marked with CDllc MicroBeads. The H22-DC was sorted with Mimi MACS sorter. The techniques of cell culture, immunocytochemistry and light microscopy were also used to test the characteristice of growth and morphology of H22-DC in vitro. As the immunogen, H22-DC was inoculated subcutaneously into the right armpit of BALB/C mice, and their tumorigenicity in vive was observed. MTT was used to test the CTL activity of murine spleen in vitro.``RESULTS DC cells isolated and generated were CD1 lc +cells with irregular shape, and highly expressed CD80,CD86 and CD54 molecules. H22 cells were CDllc cells with sphericel shape and bigger volume, and did not express CD80, CD86 and CD54 molecules. H22-DC was CDllc+ cells with bigger volume, being spherical, flat or irregular in shape, and highly expressed CD80, CD86 and CD54 molecules, too. H22-DC was able to divide and proliferate in vitro, but its activity of proliferation was significantly decreased as compared with H22 cells and its growth curve was flatter than H22 cells. After subcutaneous inoculation over 60 days, 1-12_2-DC showed no tumorigenecity in mice, which was significantly different from control groups (P< 0.01 ) . The spleen CTL activity against H22 cells in mice implanted with fresh H22-DC was significantly higher than control groups (P< 0.0l).``CONCLUSION H22-DC could significantly stimulate the specific CTL activity of murine spleen, which suggests that the fusion cells have already obtained the function of antigen presenting of parental DC and could present H22specific antigen which has not been identified yet, and H22-DC could induce antitumor immune response; although simply mixed H22 cells with DC could stimulate the specific CTL activity which could inhibit the growth of tumor in some degree, it could not prevent the generation of tumor. | 19 | Relationship between Egr-1 gene expression and apoptosis in esophageal carcinoma and precancerous lesions显示文摘AIM: To study the expression of early growth response gene1 (Egr-1 gene) and Bcl-X/L protein and its relationship with the cell apoptosis in human esophageal carcinoma(EC) and precancerous lesions.METHODS: In situ hybridization(ISH), immunohistochemistry (IHC) and TUNEL method were used respectively to detect Egr-1mRNA, Egr-1 protein, apoptosis related-protein Bcl-X/L and cell apoptosis in situ from 66 cases of esophageal squamous cell carcinoma and their upper cut edge and paracancerous mucosa.RESULTS: Egr-1 gene in situ hybridization, Bcl-X/L immunohistochemistry positive products were located in the cytoplasm, while Egr-1 immunohistochemistry and TUNEL positive signal were located in the nuclei. The apoptosis index(AI) and the frequency of apoptosis occurrence were increased gradually from precancerous lesion to cancer (P<0.01) and the expression of Egr-1mRNA and Egr-1 protein in dysplasia was the highest among all specimens (P<0.01).The AI of Egr-1 positive cancer tissues was much higher than that of Egr-1 negative cancer tissues (P<0.01), while the AI of Bcl-X/L positive cancer tissues was much lower than that of Bcl-X/L negative cancer tissues (P<0.01). The AI and Egr-1 expression were not correlated with invasiveness and lymphatic metastasis in EC.CONCLUSION: Cell apoptosis was present through esophageal carcinogenesis. The expression of Egr-1 mRNA and Egr-1 protein were high in precancerous lesion of esophagus. The AI was increased significantly in Egr-1 positive squamous cell carcinoma. Egr-1 might promote apoptotic effect. | 20 | A selective tropism of transfused oval cells for liver显示文摘AIM: To explore the biological behaviors of hepatic oval cells after transfused into the circulation of experimental animals.METHODS: Oval cells from male SD rat were transfused into the circulation of a female rat which were treated by a 2-AAF/CCl4 program, through caudal vein. Sex-determining gene sry which located on Y chromosome was examined by PCR and in situ hybridization technique in liver, kidney and spleen of the experimental animals, respectively.RESULTS: The results of the cell-transplant experiment showed that the srygene was detectable only in the liver but not in spleen and kidney of the experimental rats, and no signals could be detected in the control animals. It can be also morphologically proved that some exogenous cells had migrated into the parenchyma of the liver and settled there.CONCLUSION: The result means that there are exogenous cells located in the liver of the experimental animal and the localization is specific to the liver. This indicates that some 'signal molecules' must exist in the circulation of the rats treated by 2-AAF/CCl4.
