Molecular diversity and biogeography of benthic ciliates in the Bohai Sea and Yellow Sea显示文摘This study explored the molecular diversity and biogeography of benthic ciliates in Chinese marginal seas, the Bohai Sea(BHS), North Yellow Sea(NYS) and South Yellow Sea(SYS). From a previous 18S rRNA gene pyrosequencing dataset of the benthic microeukaryotes, we retrieved the sequences affiliated with phylum Ciliophora and analyzed alpha and beta diversities of ciliate communities. We found that BHS had the highest ciliate operational taxonomic unit(OTU) richness than NYS and SYS, whereas the richness was not significantly different between summer and winter. Among all the measured environmental variables, water depth showed consistently the strongest correlations with alpha diversities. Overall, the class Spirotrichea(mostly Choreotrichia and unassigned lineages within the class) dominated the communities in terms of both relative proportion of sequences(77.0%) and OTU richness(66.5%). OTU-level ciliate community structure was significant different among the three basins, but not between the seasons. Structurally, significant differences in relative proportion among the basins were detected for the class Litostomatea, but not for other classes. Partial Mantel tests demonstrated that water depth difference was more important than geographic and environment distances in shaping the community structure of benthic ciliates in the studied area. About 60% OTUs were not assigned at a class or order level and at least 45% OTUs shared a sequence similarity no more than 97% with the described species, indicating a great potential for ciliate species discovery in the offshore sediments. Compared with previous morphology-based surveys, the spatial pattern of ciliate diversity(decreasing from NYS to SYS) is also identified in the present study. Nevertheless, structurally, the dominant class appeared to be Spirotrichea in the sequencing dataset, which differs from previous morphology-based results(dominance of classes Prostomatea and Karyorelictea in biomass). The potential causes for the discrepancies between molecular and morphological findings are also discussed. | Guihao Li Lei Su Qianqian Zhang Xiaoli Zhang Jun Gong | 2019 | Acta Oceanologica Sinica2019,38,2: | 4
Scaled-up synthesis of defect-rich layered double hydroxide monolayers without organic species for efficient oxygen evolution reaction显示文摘The scaled-up synthesis of organic-free monolayer nanomaterials is highly desirable, especially in obtaining green energy by electrocatalysis. In this study,a method for the scaled-up synthesis of the series of monolayer layered double hydroxides(LDHs)without the addition of organic solvents is reported via the separate nucleation and aging steps process. The resulting monolayer LDHs with the thicknesses of less than 1 nm showed a narrow thickness distribution.X-ray absorption fine-structure revealed that monolayer NiFe-LDH nanosheets have a number of oxygen and metal vacancies defects. As a practical application, monolayer NiFe-LDH nanosheets containing defects showed an enhanced electrocatalytic water oxidation activity compared with that of bulk NiFe-LDH.Density functional theory calculations uncovered that excellent catalytic activity is attributed to vacancies defects. The proposed method is an economical and universally applicable strategy for the scaled-up production of monolayer LDHs. | Haoyuan Chi Jingwen Dong Tian Li Sha Bai Ling Tan Jikang Wang Tianyang Shen Guihao Liu Lihong Liu Luyi Sun Yufei Zhao Yu-Fei Song | 2022 | Green Energy & Environment2022,7,5: | 2
Spatiotemporal distributions and environmental drivers of diversity and community structure of nosZ-type denitrifiers and anammox bacteria in sediments of the Bohai Sea and North Yellow Sea, China显示文摘Denitrification and anammox processes are major nitrogen removal processes in coastal ecosystems. However, the spatiotemporal dynamics and driving factors of the diversity and community structure of involved functional bacteria have not been well illustrated in coastal environments, especially in human-dominated ecosystems. In this study, we investigated the distributions of denitrifiers and anammox bacteria in the eutrophic Bohai Sea and the northern Yellow Sea of China in May and November of 2012 by constructing clone libraries employing nosZ and 16S rRNA gene biomarkers. The diversity of nosZ-denitrifier was much higher at the coastal sites compared with the central sites, but not significant among basins or seasons. Alphaproteobacteria were predominant and prevalent in the sediments, whereas Betaproteobacteria primarily occurred at the site near the Huanghe (Yellow) River estuary. Anammox bacteria Candidatus Scalindua was predominant in the sediments, and besides, Candidatus Brocadia and Candidatus Kuenenia were also detected at the site near the Huanghe River estuary that received strong riverine and anthropogenic impacts. Salinity was the most important in structuring communities of nosZ-denitrifier and anammox bacteria. Additionally, anthropogenic perturbations (e.g. nitrogen overloading and consequent high primary productivity, and heavy metal discharges) contributed significantly to shaping community structures of denitrifier and anammox bacteria, suggesting that anthropogenic activities would influence and even change the ecological function of coastal ecosystems. | CAI Youjun ZHANG Xiaoli LI Guihao DONG Jun YANG Anjing WANG Guangyu ZHOU Xiaojian | 2019 | Journal of Oceanology and Limnology2019,37,4: | 2

Diversity and distribution of bacterioplankton in the coastal upwelling waters off Hainan Island,China显示文摘The diversity, community composition and 16 S rRNA gene abundance of bacterioplankton along a transect across an upwelling area off the eastern coast of Hainan Island(the Qiongdong upwelling)were investigated in August of 2016 using high throughput sequencing and quantitative PCR assay of 16 S rRNA genes.Compared with the offshore stations, the inner-shelf stations had higher bacterial gene abundance(up to 3 fold)and operational taxonomic unit richness,a result of the influence of upwelled and fresher waters.Overall,a majority of the reads were affiliated with Gammaproteobacteria(11%-74%)and Alphaproteobacteria(14%-43%).The structure of the bacterial community was significantly affected by salinity, dissolved oxygen,NO_(3)^(-) and NH_(4)^(+),which also defined the physicochemical features of the upwelled waters.Horizontally, the relative abundances and gene abundances of Rhodobacteraceae,SAR86,Cyanobacteria and Bacteroidetes in eutrophic zone decreased from the inner to outer shelves and slope, whereas Alteromonas,Vibrio and Oceanospirillaceae exhibited an opposite trend. This study stresses the riverine influence on the oceanographic condition and spatial variability of bacterioplankton diversity and distribution in the Qiongdong upwelling. | Fahui Gong Qixing Ji Guihao Li Kedong Yin Jun Gong | 2022 | Acta Oceanologica Sinica2022,41,3: | 0
Diversity, community structure, and quantity of eukaryotic phytoplankton revealed using 18S rRNA and plastid 16S rRNA genes and pigment markers:a case study of the Pearl River Estuary显示文摘Understanding consistencies and discrepancies in characterizing diversity and quantity of phytoplankton is essential for better modeling ecosystem change. In this study, eukaryotic phytoplankton in the Pearl River Estuary,South China Sea were investigated using nuclear 18S rRNA and plastid 16S or 23S rRNA genes and pigment analysis. It was found that 18S abundance poorly explained the variations in total chlorophyll a(Chl-a).However,the ratios of log-transformed 18S abundance to Chl-a in the major phytoplankton groups were generally environment dependent, suggesting that the ratio has potential as an indicator of the physiological state of phytoplankton. The richness of 18S-based operational taxonomic units was positively correlated with the richness of 16S-based amplicon sequence variants of the whole phytoplankton community, but insignifcant or weak for individual phytoplankton groups.Overall, the 18S based, rather than the 16S based, community structure had a greater similarity to pigment-based estimations.Relative to the pigment data, the proportion of haptophytes in the 18S dataset, and diatoms and cryptophytes in the 16S dataset, were underestimated. This study highlights that 18S metabarcoding tends to refect biomass-based community organization of eukaryotic phytoplankton.Because there were lower copy numbers of plastid 16S than 18S per genome, metabarcoding of 16S probably approximates cell abundance-based community organization.Changes in biomass organization of the pigment-based community were sensitive to environmental changes.Taken together, multiple methodologies are recommended to be applied to more accurately profle the diversity and community composition of phytoplankton in natural ecosystems. | Shumin Xu Guihao Li Cui He Yi Huang Dan Yu Huiwen Deng Zhuyin Tong Yichong Wang Christine Dupuy Bangqin Huang Zhuo Shen Jie Xu Jun Gong | 2023 | Marine Life Science & Technology2023,5,3: | 0
