2Identification of Biocontrol Strain HJX1 and Determination of Its Antifungal Spectrum显示文摘A biocontrol strain HJX1,which had good inhibitory effect against Fusarium oxysporum f. sp. cubense,was identified as Bacillus subtilis based on traditional morphology,16 S rRNA gene sequence and fatty acid profiles. The sequence of 16 S rRNA gene had 99% homology to B. subtilis according to the DNA fragment of 1 521 bp amplified with a pair of universal primers,P0 and P6,of bacteria. 33D Printing of Well Dispersed Electrospun PLGA Fiber Toughened Calcium Phosphate Scaffolds for Osteoanagenesis显示文摘Although the toughening of Calcium phosphate(CaP)scaffold by the addition of fiber has been well recognized,integrated mechanical,structural and functional considerations have been neglected in the design and fabrication of CaP scaffold implant.The emerging 3D printing provides a promising technique to construct CaP scaffold with precise size and elaborate microstructure.However,the most challenge is to extrude smoothly the CaP paste containing fibers for frequently-used extrusion-based 3D printing.In this study,frozen section and chemical dispersant(Pluronic F127,F127)were employed jointly to prepare non-aggregated polylactic-co-glycolic acid(PLGA)fibers.The injectability of CaP pastes with well dispersed PLGA fibers was more than 90%when the content of PLGA fibers was no more than 3 wt%.Meanwhile rheological property of CAP pastes with well dispersed fibers showed shear thinning,which were both beneficial to extrude CaP paste with well dispersed fibers for 3D printing.Moreover,these CaP scaffolds showed ductile fracture behavior due to the pullout and bridging effect of PLGA fibers.The cell proliferation and alkaline phosphatase(ALP)activity indicated that 3D printed CAP scaffold containing PLGA fibers possesses excellent biocompatibility and facilitate osteogenic differentiation ability.Thus,it was feasible to print CaP pastes with well dispersed PLGA fibers to construct toughening CaP scaffolds with the higher shape fidelity and complex structures,which had significant clinical potentials in osteoanagenesis due to their higher toughness and excellent biocompatibility.Guoru Zhao Rongwei Cui You Chen Sijie Zhou Chen Wang Zhangmei Hu Xiaoke Zheng Maohong Li Shuxin Qu 2020Journal of Bionic Engineering2020,17,4:2
4Reconstruction of Surface Porous PEEK Decorated with Strontium-doped Calcium Phosphate Coatings for Enhancing Osteogenic Activity显示文摘The aim of this study was to reconstruct surface porous structure with hundreds of micrometers and then bio-mineralize Sr-doped Calcium Phosphate(Sr-doped CaP)on Polyetheretherketone(PEEK)profile to enhance its bioactivity.A surface porous structure was prepared on PEEK profile by embedding and acid-etching of SiO2 particles as porogen(SP-PEEK).Then the Sr-doped CaP was further decorated on the porous surface after sulfonation,introduction of Sr-doped CaP crystal seeds and bio-mineralization in 1.5 times simulated body fluid(BSSP-PEEK-CaP/Sr).It was feasible to reconstruct the surface porous structure with hundreds of micrometers on PEEK profile by the present method without damaging its mechanical properties.The Sr-doped CaP crystal seeds effectively promoted the bio-mineralization of bio-inertness PEEK.All as-prepared PEEK did not inhibit the proliferation of cells.ALP of bio-mineralized groups was significantly increased than that of the other groups.The BSSP-PEEK-CaP/Sr obviously affected the morphology and promoted the adhesion and spreading of cells.As a result,the cyto-biocompatibity and bioactivity of PEEK were improved after bio-mineralization.Sr-doped CaP on PEEK most likely was beneficial for cells,which was associated with the increasing of the hydrophilicity on PEEK.This study provided a candidate method to improve the osteogenesis of PEEK implants.Chengcheng Liang Xiong Xiong Rongwei Cui Yuanxiao Hong Xin Liu Guoru Zhao Lizi Ye Shuxin Qu 2021Journal of Bionic Engineering2021,18,4:2
5Isolation,Screening and Identification of Antagonistic Actinomycetes against Ustilago scitaminea Syd显示文摘[Objectives] This study was conducted to obtain actinomycetes strains having antagonistic effect on Ustilago scitaminea Syd. [Methods]At first,actinomycetes strains were isolated from 22 soil samples in Hainan sugarcane regions. Then,antagonistic actinomycetes against U. scitaminea were screened by confrontation culture. Finally,the taxonomic status of antagonistic actinomycetes was determined using 16 S rD NA. [Results] From the 22 samples,984 actinomycetes strains were isolated. From all the isolated strains,23 antagonistic actinomycetes strains were obtained through primary screening,and one strains with better antagonistic effect was then obtained through secondary screening,and designated FAS. 16 S rD NA identification showed that strain FAS shared 99% sequence similarity with Streptomyces cealestis US24. A phylogenetic tree was built with MAGE 7. 0 software,and the results showed that strain FAS had the shortest genetic distance with S. caelestis US24. Therefore,the actinomycetes FAS was determined as S. caelestis. 6Colonization Ability of Bacillus subtilis HAS in Sugarcane显示文摘Bacillus subtilis HAS had good control effects on sugarcane smut in the field. Colonization dynamics of B. subtilis HAS were studied through biotics marker and pot culture, to understand the bio-control mechanism of the strain and provide experimental basis for commercialization.The results showed that B. subtilis HAS had strong colonization ability, which could be colonized in sugarcane root, stem, leaf and rhizosphere for long time, but the colonization levels were different. The colonization density of B. sutilis HAS remained 103 CFU/g soil in the rhizosphere and 102 CFU/g(fresh weight) in the root after inoculation of 60 d. The colonization in the rhizosphere decreased first, then increased and decreased again,and finally stabilized. The colonization in the root first increased then decreased. 7Study on Acute Toxicity of Purified Protein HAS1 Expression in KM Mice显示文摘[Objectives]The antifungal protein HAS1 newly obtained was evaluated for acute toxicity safety in KM mice according to relevant national regulations,so as to eliminate people s concerns about the safety of transgenic plants.[Methods]The acute toxicity of the purified protein HAS1 was observed by intragastric administration of mice,and the poisoning symptoms,poisoning degree,recovery and death were observed.[Results]No abnormal toxicity symptoms were observed in the test group,the vehicle control group and the blank control group.The main tissues and organs were not abnormal in gross anatomy.The average body weight of each group showed an increasing trend compared with before administration at 1,3,7,11 and 14 d after administration.It was found that after giving the purified protein HAS1 to KM mice at a cumulative dose of 64 mg/kg a day,no obvious toxicity was observed in the acute toxicity test,indicating that the test substance was non-toxic by oral administration.[Conclusions]This study provides a basis for further use of the protein and its coding genes.Guoru XIONG Gengfeng ZHAO Jungang WANG Guilan XING Shuzhen ZHANG 2019Agricultural Biotechnology2019,8,4:1

9Effects of Differential Expression of Monosaccharide Transporter Genes on Yield Formation of Virus-free Sugarcane Seedlings显示文摘The yield increase effect was analyzed by planting disease-free sugarcane seedlings,and the differential expression of three monosaccharide transporter genes( SGT1,SGT2 and PST2a) in disease-free and untreated sugarcane seedlings was studied to clarify the functions of these genes on sugarcane yield. The yield was increased by 27. 2% by planting disease-free sugarcane seedlings. The expression levels of SGT1,SGT2 and PST2 a in leaves of virus-free plants were1-20 times higher than those in untreated sugarcane plants during the whole growth period. In addition,the expression levels of SGT1 and SGT2 in 1-8 internodes of sugarcane at elongation stage and maturity stage were 1-45 times higher than those in untreated plants. And the expression levels of PST2 a in 4-8 internodes of sugarcane plants at elongation stage were 4-7 times higher than those in untreated plants,but there was no difference in expression levels at maturity stage. The functions of different monosaccharide transporters varied among different growth stages. 10Cloning and Analysis of Chitinase Gene in Bacillus subtilis HAS显示文摘[Objective] The paper was to study the possible mechanism of chitinase gene. [Method] The chitinase gene with inhibitory effect against a variety of pathogenic fungi was cloned and analyzed in this study. [Result] The genome of HAS strain contained an ORF of 834 bp. 11Molecular Detection of Sugarcane Ratoon Stunting Disease in Hainan Sugarcane-growing Areas显示文摘[Objective]The paper was to clarify the occurrence of sugarcane ratoon stunting disease(RSD)in Hainan sugarcane-growing areas.[Method]In 2019,270 samples of sugarcane leaves were collected from six main sugarcane-growing areas in Hainan Province,and RSD was detected by PCR assay with specific primers.[Result]RSD was detected out in 41 out of 270 sugarcane samples,with an average detection rate of15.19%.The detection rates of RSD were different in six sugarcane-growing areas;the detection rate of RSD in Danzhou sugarcane-growing area was the highest of 22.00%;the detection rate of RSD in Lingao sugarcane-growing area was the lowest of 9.26%.RSD was detected out in 8 out of10 main sugarcane cultivars,among which Xintaitang 22 suffered the heaviest damage,with the positive detection rate of 45.83%;RSD had not been detected out in Zhongtang 1 and Zhongtang 2,while the positive detection rates of RSD in the remaining seven sugarcane cultivars were10.00%-31.25%.[Conclusion]RSD commonly occurs in Hainan sugarcane-growing areas.The research results provide a basis for scientific prevention and control of RSD and promotion and application of healthy virus-free sugarcane seedlings.Shen Linbo Xiong Guoru Feng Xiaoyan Wang Wenzhi Feng Cuilian Zhao Tingting Wang Jungang Wu Nannan Zhang Shuzhen 2020Plant Diseases and Pests2020,11,1:0
12Expression of Invertase Genes in Virus-free Sugarcane显示文摘Virus-free sugarcane seedlings have improved biomass and sucrose content compared with ordinary seedlings,and sucrose invertases are key enzymes regulating sugarcane growth and sucrose accumulation.In this study,the differences in the expression levels of 3 invertase genes,CWI,SAI and NI,between virusfree and ordinary sugarcane seedlings were analyzed.Compared with ordinary sugarcane plants,the expression of CWI was mainly up-regulated in immature leaves and stems at elongation stage and leaves and immature internodes at maturation stage,and especially,greatly up-regulated in immature internodes at maturation stage of virus-free plants.The expression of SAI and NI were mainly up-regulated in leaves and immature internodes of virus-free plants at maturation stage,which might be beneficial to sugar accumulation and rapid utilization of monosaccharide in the stalks of virus-free plants.It is further indicated that virus-free treatment could significantly improve the expression of sucrose invertases at late growth period,and might facilitate the increase of plant biomass.Jungang WANG Tingting ZHAO Benpeng YANG Wenwei CAI Cuilian FENG Jun ZENG Guoru XIONG Shuzhen ZHANG 2017Agricultural Biotechnology2017,6,4:0

14Preliminary Study on Production Conditions and Action Mechanism of Antifuingal Protein from Strain HJX1显示文摘Antagonistic bacterium HJX1 is a Bacillus subtilis strain isolated from field rhizosphere soil where banana wilt was serious,with certain control efficacy on banana Fusarium wilt.The crude extract obtained from its fermentation broth after ammonium sulfate precipitation has an inhibitory effect on various pathogenic fungi.The culture conditions for this strain were optimized in this study with Fusarium oxysporum f.sp.cubense as an indicator,and the results showed that the fermentation broth obtained from culture in LB liquid medium under 30 ℃ at 170 r / min for 48 h had the strongest inhibition against F.oxysporum f.sp.cubense.It was observed that the inhibition was mainly reflected by causing breakage and distortion of mycelia,followed by appearance of vesicles and swelling,ablation,and leakage of protoplasm.This study provides a scientific basis for the study on the action mechanism of crude protein from bacterial strain HJX1.Gengfeng ZHAO Yimin QIU Yunxia ZHANG Guoru XIONG 2016Agricultural Biotechnology2016,5,5:0
15Optimization of Streptomyces caelestis Culture Conditions and Preliminarily Determination of Its Antifungal Components显示文摘Streptomyces caelestis FAS is an actinomyces strain isolated with Ustilago scitaminea as an indicator. With GAUZE's medium as the basic medium,carbon source and nitrogen source in the medium and ventilation volume were optimized,and the antifungal substances produced by S. caelestis FAS were preliminarily determined through organic solvent extraction and ammonium sulfate precipitation. The results showed that the antifungal components produced by S. caelestis FAS were proteins,and the optimal culture conditions were using corn flour as carbon source and KNO_3 as nitrogen source and culturing in 250 ml triangular flask added with 70 ml of culture medium. 16Study on Wide Antifungal Property of Bacillus subtilis HAS显示文摘Bacillus subtilis HAS is a biocontrol strain with better inhibitory effect on Sporisorium scitaminea Sydow. In order to further understand its action range,with fungal pathogens in sugarcane production as targets,confrontation tests and tests of inhibitory effect of secondary metabolites on growth of pathogens were carried out. The results showed that B. subtilis HAS not only had better inhibitory effect on S. Scitaminea,but also inhibited the growth of various fungal pathogens in sugarcane production. The inhibitory effect was the best on Ceratocystis adiposum,Thielaviopsis paradoxa,Fusarium moniliforme and Colletotrichum fuleatum,which was reflected by the radius of inhibitory zone over 10 mm. The inhibitory rates of the secondary metabolites were about 70%. 17Skin Anaphylaxis Test of Purified Protein HAS1 in Cavies显示文摘The antifungal protein HAS1 is a new antifungal protein isolated from the genome of Bacillus subtilis HAS,which has a good inhibitory effect on various pathogenic fungi in sugarcane.This study aimed at evaluating the immunological transfer reaction of the test sample through repeated skin contact by observing whether the cavy skin repeatedly exposed to purified protein HAS1 suffers from allergic reaction and how strong is the allergic reaction.The results showed that the test group and the vehicle control group exhibited no allergic reaction in the skin immediately and at 24,48 and 72 h and had an mean reaction score and a sensitization rate both of 0,so the results of the naked eye observation were both nonallergenic to cavy skin;and in the positive control group,the cavies were observed to be highly sensitized immediately after removing the drug,mildly sensitized at 24 h,and not sensitized at 48 and 72 h,which meant the skin allergy was alleviated with the observation time.It is suggested that the purified protein HAS1 is negative for cavy skin anaphylaxis test (nonallergenic to cavy skin),which provides an experimental basis for further utilization of the protein and its coding gene.Guoru XIONG Gengfeng ZHAO Jungang WANG Guilan XING Shuzhen ZHANG 2019Agricultural Biotechnology2019,8,5:0
18Screening and Identification of Antagonistic Strain HAS against Sporisorium scitaminea Sydow显示文摘The present study was to screen and identify a biocontrol strain with antagonistic effect against Sporisorium scitaminea Sydow. 16 S rDNA gene fragments were amplified by universal primers P0 and P6. The results showed that 16 S rDNA gene fragment with the size of 1 533 bp was obtained,and its sequence had the homology of 99% with the sequence of Bacillus subtilis. The morphological characteristics of the strain were as follows: the mycelium of strain HAS was short-rod shape,and could move; Gram staining was positive; 3% KOH solubility test was negative; flagellum was grown around the cells; spores were grown in the middle,elliptic,and cysts were slightly swelled. The strain was preliminarily identified as B. subtilis. Dual culture with pathogenic fungi showed that the strain had good inhibitory effect against various pathogenic fungi causing diseases on sugarcane and other crops. 20Fungistatic Activity of Crude Protein Extracted from Bacillus subtilis HJX1显示文摘Bacillus subtilis HJX1,a biocontrol strain of Fusarium oxysporum f. sp. cubense,could inhibit growth of several plant pathogenic fungi in vitro. To understand its mechanism of fungistasis,we extracted crude protein from fermentation broth of the strain HJX1 through ammonium sulfate precipitation,and preliminarily studied fungistatic activities at different conditions. The results showed that the crude protein was insensitive to protease K,trypsin or ultraviolet radiation.
