| 1 | Dysregulation of gastric H,K-ATPase by cigarette smoke extract显示文摘AIM:To test whether the expression and activity of H,K-ATPase in parietal cells would be affected by cigarette smoke extract.METHODS: Extracts of cigarette smoke were administered into mice by gastric gavage (5 mg/kg body weight/day) for 3 d or in drinking water for 7 or 14 d. For the latter, each day a mouse consumed 5 mL water containing extracts of two cigarettes, on average. Control littermate mice received only vehicle. To compare the amount of H,K-ATPase in control and smoke-treated mice, the stomach was processed for Western blotting and immunohistochemical analysis using monoclonal antibodies specific for α- or β-subunits of H,K-ATPase. The p-nitrophenylphospatase activity assay was used as a measurement for K-dependent H,K-ATPase activity.RESULTS: Probed transblots showed an increase in the amount of H,K-ATPase in smoke-treated mice which was confirmed by immunohistochemistry and was found to be due to increased amounts of protein per parietal cell rather than an increased parietal cell number. The increase in the amount of H,K-ATPase was associated with an enhancement of its enzymatic activity. | 2 | Porous media grain size distribution and hydrodynamic forces effects on transport and deposition of suspended particles显示文摘The effects of porous media grain size distribution on the transport and deposition of polydisperse suspended particles under different flow velocities were investigated. Selected Kaolinite particles(2–30 μm) and Fluorescein(dissolved tracer) were injected in the porous media by step input injection technique. Three sands filled columns were used: Fine sand,Coarse sand, and a third sand(Mixture) obtained by mixing the two last sands in equal weight proportion. The porous media performance on the particle removal was evaluated by analysing particles breakthrough curves, hydro-dispersive parameters determined using the analytical solution of convection–dispersion equation with a first order deposition kinetics, particles deposition profiles, and particle-size distribution of the recovered and the deposited particles. The deposition kinetics and the longitudinal hydrodynamic dispersion coefficients are controlled by the porous media grain size distribution. Mixture sand is more dispersive than Fine and Coarse sands. More the uniformity coefficient of the porous medium is large, higher is the filtration efficiency. At low velocities, porous media capture all sizes of suspended particles injected with larger ones mainly captured at the entrance.A high flow velocity carries the particles deeper into the porous media, producing more gradual changes in the deposition profile. The median diameter of the deposited particles at different depth increases with flow velocity. | 3 | Modelling the spatial distribution of snake species in northwestern Tunisia using maximum entropy(Maxent) and Geographic Information System(GIS)显示文摘We used GIS and maximum entropy to predict the potential distribution of six snake species belong to three families in Kroumiria(Northwestern Tunisia): Natricidae(Natrix maura and Natrix astreptophora), Colubridae(Hemorrhois hippocrepis, Coronella girondica and Macroprotodon mauritanicus), and Lamprophiidae(Malpolon insignitus). The suitable habitat for each species was modelled using the maximum entropy algorithm, combining presence field data(collected during 16 years:2000–2015) with a set of seven environmental variables(mean annual precipitation, elevation, slope gradient,aspect, distance to watercourses, land surface temperature and normalized Differential Vegetation Index. The relative importance of these environmental variables was evaluated by jackknife tests and the predictive power of our models was assessed using the area under the receiver operating characteristic. The main explicative variables of the species distribution were distance from streams and elevation, with contributions ranging from 60 to 77 and from 10 to 25%,respectively. | 4 | Clinical and epidemiological characteristics of norovirus gastroenteritis among hospitalized children in Lebanon显示文摘AIM To assess the burden of norovirus(No V) and to determine the diversity of circulating strains among hospitalized children in Lebanon. METHODS Stool samples were collected from children presenting with acute gastroenteritis to six major hospitals in Lebanon. A total of 739 eligible stool samples, testing negative for diarrhea caused by rotavirus as a possible viral pathogen, were collected between January 2011 and June 2013. A standardized questionnaire including demographic, epidemiological and clinical observations was used at the time of hospitalization of children presenting with diarrhea. Viral RNA was extracted from stool samples followed by reverse transcription polymerase chain reaction and nucleotide sequencing of a fragment of the viral protein 1 capsid gene. Multiple sequence alignments were carried out and phylogenetic trees were constructed using the MEGA 6 software.RESULTS Overall, 11.2% of stool samples collected from children aged < 5 years tested positive for No V genogroups Ⅰ(GⅠ) and Ⅱ(GⅡ). GⅡ accounted for 10.6% of the gastroenteritis cases with only five samples being positive for GⅠ(0.7%). The majority of hospitalized children showed symptoms of diarrhea, dehydration, vomiting and fever. Upon sequencing of positive samples and based on their clustering in the phylogenetic tree, 4/5 of GⅠ gastroenteritis cases were designated GⅠ.3 and one case as GⅠ.4. GⅡ.4 was predominantly detected in stool of our study participants(68%). We report a JB-15/KOR/2008 GⅡ.4 Apeldoorn 2008-like variant strain circulating in 2011; this strain was replaced between 2012 and 2013 by a variant sharing homology with the Sydney/NSW0514/2012/AUS GⅡ.4 Sydney 2012 and Sydney 2012/FRA GⅡ.4 strains. We also report the co-circulation of non-GⅡ.4 genotypes among hospitalized children.
