15SCF和TGF-β反义寡核苷酸对CD34^+造血细胞的协同作用显示文摘转化生长因子-β1(TGF-β1)反义寡核苷酸能使骨髓中幼稚的CD34^+细胞从静息期进入细胞周期,从而促进CFU-GEMM及早期BFU-E的集落形成。由于干细胞因子(SCF)对CD34^+细胞的集落形成也有类似的作用,作者对SCF的作用是否与阻断TGF-β的抑制效应有关进行了研究。结果表明,与TGF-β1反义核酸、SCF单用相比,两者合用对骨髓CD34^+细胞集落形成的促进作用优于递加作用,对脐血的促进作用略低于递加作用。这说明SCF仅可使部分被TGF-β维持在静息期的脐血造血细胞进入细胞周期。当TGF-β1反义寡核苷酸和SCF合用时,脐血中CFU-GEMM的绝对数比骨髓中CD34^+细胞多4倍。这与我们曾观察到的脐血所含的CD34^+CD38^-细胞,即对TGF-β1反义寡核苷酸有反应的细胞群比骨髓中多4倍的结果相一致。李玛琳 AA Cardoso P Sansilvestri A Hatzfeld J-P Levesque J Hatzfeld EL Brown H Sookdo SC Clark 1996中国实验血液学杂志1996,4,3:1

17对脐血CD34^+CD38^-细胞的研究揭示脐血移植成人的可能性显示文摘通过在生长因子依赖的长期培养体系中加入转化生长因子β1(TGF-β1)的反义核酸或抗TGF-β1抗体以阻断TGF-β1的抑制作用,比较了脐血和骨髓的CD34^+CD38^-及CD34^+CD38^+亚群在长期培养中的增殖反应以及产生早期造血前体细胞的能力。结果显示:脐血中CD34^+CD38^-细胞占CD34^+的比例为4%,比骨髓多4倍;和成人骨髓移植平均所需的骨髓标本相比,一份常规脐血标本含有的CD34^+CD38^-细胞所产生的CFU-GEMM和骨髓相当,CFU-GM为骨髓的2倍,BFU-E为骨髓的3倍;由脐血产生的集落明显大于由骨髓产生的集落。提示脐血更富含造血干细胞并具有较高的增殖潜能。脐血中晚期祖细胞量低于骨髓。以上研究结果提示脐血可用于成人进行造血细胞移植。李玛琳 A ACardoso P Batard A Hatzfeld J-PLevesque B Panterne P Sansilvestri J Hatzfeld EL Brown H Sookdo SC Clark 1996中国实验血液学杂志1996,4,2:1
