Atg5 regulates late endosome and lysosome biogenesis显示文摘Autophagy is an evolutionarily conserved lysosome-based degradation process.Atg5 plays a very important role in autophagosome formation. Here we show that Atg5 is required for biogenesis of late endosomes and lysosomes in an autophagy-independent manner. In Atg5 cells, but not in other essential autophagy genes defecting cells, recycling and retrieval of late endosomal components from hybrid organelles are impaired, causing persistent hybrid organelles and defective formation of late endosomes and lysosomes.Defective retrieval of late endosomal components from hybrid organelles resulting from impaired recruitment of a component of V1-ATPase to acidic organelles blocks the pH-dependent retrieval of late endosomal components from hybrid organelles.Lowering the intracellular pH restores late endosome/lysosome biogenesis in Atg5 cells. Our data demonstrate an unexpected role of Atg5 and shed new light on late endosome and lysosome biogenesis.PENG JunYa ZHANG Ran CUI YiTong LIU HaoDong ZHAO XiaoXin HUANG Lei HU MingXu YUAN XiaoXi MA BenYu MA XiaoWei TAKASHI Ueno MASAAKI Komatsu LIANG XingJie YU Li 2014Science China(Life Sciences)2014,57,1:6
A review of progress in coupled ocean-atmosphere model developments for ENSO studies in China显示文摘El Nino-Southern Oscillation(ENSO) is the strongest interannual signal that is producedby basinscale processes in the tropical Pacific, with significant effects on weather and climate worldwide. In the past, extensive and intensive international efforts have been devoted to coupled model developments for ENSO studies.A hierarchy of coupled ocean-atmo sphere models has been formulated;in terms of their complexity, they can be categorized into intermediate coupled models(ICMs),hybrid coupled models(HCMs),and fully coupled general circulation models(CGCMs).ENSO modeling has made significant progress over the past decades, reaching a stage where coupled models can now be used to successfully predict ENSO events 6 months to one year in advance.Meanwhile,ENSO exhibits great diversity and complexity as observed in nature, which still cannot be adequately captured by current state-of-the-art coupled models, presenting a challenge to ENSO modeling. We primarily reviewed the long-term efforts in ENSO modeling continually and steadily made at different institutions in China;some selected representative examples are presented here to review the current status of ENSO model developments and applications, which have been actively pursued with noticeable progress being made recently. As ENSO simulations are very sensitive to model formulations and process representations etc.,dedicated efforts have been devoted to ENSO model developments and improvements. Now,different ocean-atmosphere coupled models have been available in China, which exhibit good model performances and have already had a variety of applications to climate modeling, including the Coupled Model Intercomparison Project Phase 6(CMIP6).Nevertheless,large biases and uncertainties still exist in ENSO simulations and predictions, and there are clear rooms for their improvements, which are still an active area of researches and applications. Here, model performances of ENSO simulations are assessed in terms of advantages and disadvantages with these differently formulated coupled models, pinpointing to the areas where they need to be further improved for ENSO studies. These analyses provide valuable guidance for future improvements in ENSO simulations and predictions.ZHANG Rong-Hua YU Yongqiang SONG Zhenya REN Hong-Li TANG Youmin QIAO Fangli WU Tongwen GAO Chuan HU Junya TIAN Feng ZHU Yuchao CHEN Lin LIU Hailong LIN Pengfei WU Fanghua WANG Lin 2020Journal of Oceanology and Limnology2020,38,4:4
Exploring sensitive area in the tropical Indian Ocean for El Niño prediction: implication for targeted observation显示文摘Based on initial errors of sea temperature in the tropical Indian Ocean that are most likely to induce spring predictability barrier(SPB)for the El Niño prediction, the sensitive area of sea temperature in the tropical Indian Ocean for El Niño prediction starting from January is identified using the CESM1.0.3(Community Earth System Model),a fully coupled global climate model. The sensitive area locates mainly in the subsurface of eastern Indian Ocean. The effectiveness of applying targeted observation in the sensitive area is also evaluated in an attempt to improve the El Niño prediction skill. The results of sensitivity experiments indicate that if initial errors exist only in the tropical Indian Ocean, applying targeted observation in the sensitive area in the Indian Ocean can significantly improve the El Niño prediction. In particular, for SPB-related El Niño events, when initial errors of sea temperature exist both in the tropical Indian Ocean and the Pacific Ocean, which is much closer to the realistic predictions,if targeted observations are conducted in the sensitive area of tropical Pacific, the prediction skills of SPB-related El Niño events can be improved by 20.3%in general. Moreover,if targeted observations are conducted in the sensitive area of tropical Indian Ocean in addition, the improvement of prediction skill can be increased by 25.2%.Considering the volume of sensitive area in the tropical Indian Ocean is about 1/3 of that in the tropical Pacific Ocean, the prediction skill improvement per cubic kilometer in the sensitive area of tropical Indian Ocean is competitive to that of the tropical Pacific Ocean.Additional to the sensitive area of the tropical Pacific Ocean, sensitive area of the tropical Indian Ocean is also a very effective and cost-saving area for the application of targeted observations to improve El Niño forecast skills.ZHOU Qian DUAN Wansuo HU Junya 2020Journal of Oceanology and Limnology2020,38,6:1
A Hybrid Coupled Ocean–Atmosphere Model and Its Simulation of ENSO and Atmospheric Responses显示文摘A new hybrid coupled model(HCM) is presented in this study, which consists of an intermediate tropical Pacific Ocean model and a global atmospheric general circulation model. The ocean component is the intermediate ocean model(IOM)of the intermediate coupled model(ICM) used at the Institute of Oceanology, Chinese Academy of Sciences(IOCAS). The atmospheric component is ECHAM5, the fifth version of the Max Planck Institute for Meteorology atmospheric general circulation model. The HCM integrates its atmospheric and oceanic components by using an anomaly coupling strategy. A100-year simulation has been made with the HCM and its simulation skills are evaluated, including the interannual variability of SST over the tropical Pacific and the ENSO-related responses of the global atmosphere. The model shows irregular occurrence of ENSO events with a spectral range between two and five years. The amplitude and lifetime of ENSO events and the annual phase-locking of SST anomalies are also reproduced realistically. Despite the slightly stronger variance of SST anomalies over the central Pacific than observed in the HCM, the patterns of atmospheric anomalies related to ENSO,such as sea level pressure, temperature and precipitation, are in broad agreement with observations. Therefore, this model can not only simulate the ENSO variability, but also reproduce the global atmospheric variability associated with ENSO, thereby providing a useful modeling tool for ENSO studies. Further model applications of ENSO modulations by ocean–atmosphere processes, and of ENSO-related climate prediction, are also discussed.Junya HU Rong-Hua ZHANG Chuan GAO 2019Advances in Atmospheric Sciences2019,36,6:1
Different El Niño Flavors and Associated Atmospheric Teleconnections as Simulated in a Hybrid Coupled Model显示文摘A previously developed hybrid coupled model(HCM)is composed of an intermediate tropical Pacific Ocean model and a global atmospheric general circulation model(AGCM),denoted as HCMAGCM.In this study, different El Niño flavors, namely the Eastern-Pacific(EP)and Central-Pacific(CP)types,and the associated global atmospheric teleconnections are examined in a 1000-yr control simulation of the HCMAGCM.The HCMAGCM indicates profoundly different characteristics among EP and CP El Niño events in terms of related oceanic and atmospheric variables in the tropical Pacific, including the amplitude and spatial patterns of sea surface temperature(SST),zonal wind stress, and precipitation anomalies. An SST budget analysis indicates that the thermocline feedback and zonal advective feedback dominantly contribute to the growth of EP and CP El Niño events, respectively.Corresponding to the shifts in the tropical rainfall and deep convection during EP and CP El Niño events, the model also reproduces the differences in the extratropical atmospheric responses during the boreal winter. In particular, the EP El Niño tends to be dominant in exciting a poleward wave train pattern to the Northern Hemisphere, while the CP El Niño tends to preferably produce a wave train similar to the Pacific North American(PNA)pattern. As a result, different climatic impacts exist in North American regions, with a warm-north and cold-south pattern during an EP El Niño and a warm-northeast and cold-southwest pattern during a CP El Niño,respectively. This modeling result highlights the importance of internal natural processes within the tropical Pacific as they relate to the genesis of ENSO diversity because the active ocean–atmosphere coupling is allowed only in the tropical Pacific within the framework of the HCMAGCM.Junya HU Hongna WANG Chuan GAO Rong-Hua ZHANG 2024Advances in Atmospheric Sciences2024,41,5:0
Targeted B-domain deletion restores F8 function in human endothelial cells and mice显示文摘Dear Editor,Hemophilia A(HA)is the most common genetic bleeding disorders and affects 1 in 5000 male births.1 HA patients suffer spontaneous soft-tissue and joint bleeding or life-threatening intracranial hemorrhage. The main clinical treatment for severe HA patients is life-long maintenance of protein replacement therapy, which increase the risks of infection and induce FVIII inhibitors.Gene therapy is expected to cure HA,however, the current AAVbased gene addition strategy is limited by its loading capacity, immunogenicity of capsids, and the risk of insertional mutagenesis.2 Therefore, the aim of this study is to overcome these limitations by a combination of gene editing and ex vivo strategy for HA gene therapy.Zhiqing Hu Zhuo Li Yong Wu Junya Zhao Lingqian Wu Miaojin Zhou Desheng Liang 2022Signal Transduction and Targeted Therapy2022,7,7:0
Synergistic Interdecadal Evolution of Precipitation over Eastern China and the Pacific Decadal Oscillation during 1951-2015显示文摘By using the multi-taper method(MTM)of singular value decomposition(SVD),this study investigates the interdecadal evolution(10-to 30-year cycle)of precipitation over eastern China from 1951 to 2015 and its relationship with the North Pacific sea surface temperature(SST).Two significant interdecadal signals, one with an 11-year cycle and the other with a 23-year cycle, are identified in both the precipitation and SST fields. Results show that the North Pacific SST forcing modulates the precipitation distribution over China through the effects of the Pacific Decadal Oscillation(PDO)-related anomalous Aleutian low on the western Pacific subtropical high(WPSH)and Mongolia high(MH).During the development stage of the PDO cold phase associated with the 11-year cycle,a weakened WPSH and MH increased the precipitation over the Yangtze River Basin, whereas an intensified WPSH and MH caused the enhanced rain band to move northward to North China during the decay stage.During the development stage of the PDO cold phase associated with the 23-year cycle,a weakened WPSH and MH increased the precipitation over North China, whereas an intensified WPSH and the weakened MH increased the precipitation over South China during the decay stage. The 11-year and 23-year variabilities contribute differently to the precipitation variations in the different regions of China,as seen in the 1998flooding case. The 11-year cycle mainly accounts for precipitation increases over the Yangtze River Basin, while the 23-year cycle is responsible for the precipitation increase over Northeast China. These results have important implications for understanding how the PDO modulates the precipitation distribution over China, helping to improve interdecadal climate prediction.Minmin WU Rong-Hua ZHANG Junya HU Hai ZHI 2024Advances in Atmospheric Sciences2024,41,1:0
