| 3 | Development of a Widely Applicable and Simple Prognostic Score for Patients with Acute-on-chronic Liver Failure显示文摘Background and Aims:Acute-on-chronic liver failure(ACLF)tends to progress rapidly with high short-term mortality.We aimed to create a widely applicable,simple prognostic(WASP)score for ACLF patients.Methods:A retrospective cohort of ACLF cases recruited from three centers in China were divided into training and validation sets to develop the new score.A prospective longitudinal cohort was recruited for further validation.Results:A total of 541 cases were included in the training set,and seven independent ACLF prognostic factors were screened to construct a new quantitative WASP-ACLF table.In the validation set of 671 cases,WASP-ACLF showed better predictive ability for 28-day and 90-day mortality than the currently used prognostic scores at baseline,day 3,week 1,and week 2.The predictive efficacy and clinical validity of the model improved over time.Patients were assigned to low-,intermediate-,and high-risk groups by their WASP-ACLF scores.Compared with the other two groups,intermediate-risk patients had a more uncertain prognosis,with a 90-day mortality of 44.4–50.6%.Sequential assessments at weeks 1 and 2 found the 90-day mortality of intermediate-risk groups was<20%forpatients with a≥2 point decrease in WASP-ACLF and was up to 56%for patients with a≥2 points increase.Similar results were observed in prospective data.Conclusions:The new ACLF prognostic score was simple,widely applicable,and had good predictive efficacy.Continuous assessments and trend of change in WASP-ACLF need to be considered,especially for intermediate-risk patients. | Zhenjun Yu Yu Zhang Yuhan Li Feng Zhou Manman Xu Shaoli You Yu Chen Bing Zhu Ming Kong Fangjiao Song Shaojie Xin Zhongping Duan Tao Han China Network for Severe Liver Diseases | 2022 | Journal of Clinical and Translational Hepatology2022,10,5: | 1 |

| 7 | LncRNA-Airn alleviates acute liver injury by inhibiting hepatocyte apoptosis via the NF-κB signaling pathway显示文摘Acute liver injury is a common and serious syndrome caused by multiple factors and unclear pathogenesis.If the injury persists,liver injury can lead to cirrhosis and liver failure and ultimately results in the development of liver cancer.Emerging evidence has indicated that long noncoding RNAs(lncRNAs)play an important role in the development of liver injury.However,the role of antisense Igf2r RNA(Airn)in acute liver injury and its underlying mechanism remain largely unclear.In this study,we show that Airn is upregulated in liver tissue and primary hepatocytes from an acute liver injury mouse model.Consistently,Airn is also overexpressed in serum samples of patients with acute-on-chronic liver failure and is negatively correlated with the Model for End-Stage Liver Disease(MELD)score.Moreover,gene knockout and rescue assays reveal that Airn alleviates CCl_(4)-induced liver injury by inhibiting hepatocyte apoptosis and oxidative stress in vivo.Further investigation reveals that Airn decreases H_(2)O_(2)-induced hepatocyte apoptosis in vitro.Mechanistically,we reveal that Airn represses CCl_(4)-and H_(2)O_(2)-induced enhancement of phosphorylation of p65 and IκBα,suggesting that Airn inhibits hepatocyte apoptosis by inactivating the NF-κB pathway.In conclusion,our results demonstrate that Airn can alleviate acute liver injury by inhibiting hepatocyte apoptosis via inactivating the NF-κB signaling pathway,and Airn could be a potential biomarker for acute liver injury. | Shuai Shao Yu Zhang Feng Zhou Xiaoxiang Meng Zhenjun Yu Guantong Li Lina Zheng Kun Zhang Yuhan Li Beichen Guo Qi Liu Mengxia Zhang Xiaoxiao Du Wei Hong Tao Han | 2022 | Acta Biochimica et Biophysica Sinica2022,54,11: | 1 |

| 9 | Dynamics,stability,and control of a four-cable mount system for wind tunnel test显示文摘A four-cable mount system is proposed for full-model wind tunnel flutter tests,which may adjust the pitch and roll attitude of the aircraft scaled model and ensure that the model is not subjected to cable tension.The system provides sufficient support to simulate the free flight of the aircraft by applying appropriate spring stiffness and cable tensions.The proposed fourcable mount system is modeled based on Lagrange mechanics,and its dynamics equations consider aerodynamic effects.The singularity of the system and its bifurcation characteristics under flow conditions are analysed to determine the supercritical bifurcation phenomenon for different tension levels and distances from the front suspension point to the mass centre of the model.The mathematical expressions of the longitudinal flight stability of the cable mount system are derived by linearising the system dynamics equations using small perturbations.The influence of the cable tension,spring stiffness,suspension point position,and other factors on the flight stability of the aircraft are analysed.A feedforward control algorithm is proposed to minimize the total elastic potential energy of the system.The results show that the model is in the level flight state when the elastic potential energy of the four-cable mount system is minimized.A feedback control design method is proposed based on the Lyapunov stability theory to derive the closed-loop stability conditions.The system dynamics model that includes the aircraft rigid body model,flexible cables,pulleys,springs,aerodynamic model,and servo motor control is established using the flexible multibody dynamics method.A multibody dynamics solver and Simulink are used to simulate the attitude adjustment of the model in the wind tunnel and verify the supercritical bifurcation characteristics of the system and the effectiveness of the feedback and feedforward control. | Zhenjun ZHAO Zhanwei ZHAO Yu YAN Kaichun ZENG Xiaojun SHI Han WU | 2023 | Chinese Journal of Aeronautics2023,36,2: | 0 |

| 12 | High-purity magnesium screws modulate macrophage polarization during the tendon–bone healing process in the anterior cruciate ligament reconstruction rabbit model显示文摘Magnesium(Mg)screws perform clinical potential in anterior cruciate ligament(ACL)reconstruction,and promote fibrocartilaginous entheses regeneration at the femoral entrance.We aim to prove that high-purity Magnesium(HP Mg)screws modulate macrophage polarization in fibrocartilage interface regeneration both in vitro and in vivo.HP Mg extracts performed good cytocompatibility and significantly promoted M2 macrophage polarization in the flow cytometry and ELISA assays.M2 macrophages stimulated fibrochondrocyte differentiation of cocultured hBMSCs,and HP Mg extracts had synergistic effect on the process.Then we applied HP Mg screws,with Ti screws as control,in the ACL reconstruction rabbit model.In the histological and immunofluorescence analysis,HP Mg screws inhibited M1 polarization at 2 weeks and highly promoted M2 polarization at 2 and 4 weeks at the tendon–bone interface.Furthermore,regeneration of fibrocartilaginous entheses,rather than the fibrovascular scar interface,was detected in the HP Mg group at 12 weeks.For further mechanism study via RNA-seq detection and WB assays,we found that AKT1 was highly activated in M2 polarization,and HP Mg could stimulate AKT1 expression,rather than AKT2,in the early phase of tendon–bone healing.Our study elucidated macrophage polarization during tendon–bone healing process and emphasized HP Mg on M2 polarization and fibrocartilage interface regeneration via the selective activation of AKT1 and PI3K/AKT pathway. | PengFei Cheng ZhenJun Weng Musha Hamushan Weijie Cai Yubo Zhang Zun Ren Yunchu Sun XiaoNong Zhang Hao Shen Pei Han | 2022 | Regenerative Biomaterials2022,9,1: | 0 |
| 13 | Characteristics of outdoor pollutants intrusion and ventilation control in sentry buildings with normal openings显示文摘Sentry buildings have windows that are often open to facilitate communication between personnel.It also provides the possibility for the intrusion of pollutants such as vehicle exhaust emissions.To prevent the intrusion of outdoor pollutants and create an excellent indoor environment,internal circulation with double-attached ventilation(IC-DAV)and external circulation with double-attached ventilation(EC-DAV)are proposed for such buildings,and the isolation effect of two attached ventilation modes on pollutants is compared with that of natural ventilation.A computational fluid dynamics(CFD)method was used to simulate the transportation process and indoor distribution of outdoor pollutants intruding into sentry buildings from the regular openings under different outdoor wind directions,wind velocities,and states of the doors and ventilation modes.The results indicate that the leeward airflow of the three wind directions caused the largest pollutants to invade the room.The amount of pollutants intrusion increased with increasing wind velocity.When the leeward airflow and the windward airflow blow through the building,opening the door increased the amount of pollutants intruding into the room by 3.34 times and 8.85 times,respectively,compared with closing the door.However,the IC-DAV can isolate 81.7% of the pollutants while the EC-DAV can isolate 99.92% of the pollutants as compared with natural ventilation.Applying double attached ventilation mode in buildings can effectively prevent the intrusion of outdoor pollutants into the room,reduce the harm of outdoor pollutants to the health of indoor personnel,and provide a new idea for buildings to improve the indoor air quality. | Linfeng Liang Haiguo Yin Da Zhang Yuekun Han Yanyan Li Jingying Zhang Yongshou Zhang Zhenjun Ma Angui Li | 2023 | Building Simulation2023,16,3: | 0 |
