1Antihepatoma effect of alpha-fetoprotein antisense phosphorothioate oligodeoxyribonucleotides in vitro and in mice显示文摘AIM To evaluate antihepatoma effect ofantisense phosphorothioate oligodeo-xyribonucleotides (S-ODNs) targeted to alpha-fetoprotein (AFP) genes in vitro and in nudemice.METHODS AFP gene expression was examinedby immunocytochemical method or enzyme-linked immunosorbent assay. Effect of S-ODNson SMMC-7721 human hepatoma cell growth invitro was determined using microculturetetrazolium assay. In vivo antitumor activitiesof S-ODNs were monitored by measuring tumorweight differences in treated and control micebearing SMMC-7721 xenografts. Induction of cellapoptosis was evaluated by fluorescence-activated cell sorter (FACS) analysis.RESULTS Antisense S-ODN treatment led toreduced AFP gene expression. Specificantisense S-ODNs, but not control S-ODNs,inhibited the growth of heaptoma cells in vitro.In vivo. only antisense S-ODNs exhibitedobvious antitumor activities. FACS analysisrevealed that the growth inhibition by antisenseS. 2Growth-inhibiting effects of taxol on human liver cancer in vitro and in nude mice显示文摘AIM To investigate the effects of taxol onSMMC-7721 human hepatoma and itsmechanisms.METHODS In vitro cell growth was assessedby trypan blue exclusion method.Experimentalhepatoma model was established by seedingSMMC-7721 cells subcutaneously into Balb/c(nu/nu)nude mice.In vivo tumor growth wasdetermined by measurement of tumor diameterwith Vernier calipers.The syntheses of DNA,RNA and protein were analyzed by incorporationof ~3H-thymidine,~3H-uridine and ~3H-leucinerespectively.Using light and electronmicroscopes to observe the morphologicalchanges of cells including mitosis andapoptosis.RESULTS Taxol was effective against SMMC-7721 human hepatoma cell growth in the rangesof 2.5 nmol/L-10 nmol/L with mitotic arrestand apoptosis in vitro.DNA,RNA and proteinsyntheses in cells were also obviouslysuppressed by in vitro treatment of taxol for72 h.Taxol at 2.5 nmol/L reduced ~3H-thymidineuptake to about 34% of the control value(P<0.05).Increasing the dose of taxol to20 nmol/L resulted in a greater decrease in ~3H-thymidine incorporation to 60% of the controlvalue(P<0.01).At a concentration of 20 nmol/L,the ~3H-uridine and ~3H-leucine uptakeswere reduced to 52%(P<0.05)and 63%(P<0.01),respectively.In vivo,taxolsignificantly inhibited SMMC-7721 tumor growthat 10 mg/kg,i.p.,once daily for 10 d.A morethan 90% decrease in tumor volume wasobserved by day 11(P<0.01)similarly withmitotic arrest and cell apoptosis.CONCLUSION Taxol has a marked anticanceractivity in SMMC-7721 human hepatoma both invitro and in nude mice.Its mechanisms might beassociated with mitotic arrest,subsequently,apoptosis of the hepatoma cells.No obvioustoxicity was observed with in vivoadministration of taxoi.Jin Hui Yuan Ru Ping Zhang Ru Gang Zhang Li Xia Guo Xing Wang Wang Dan Luo Yong Xie Hong Xie 2000World Journal of Gastroenterology2000,6,2:18

4Gut microbiota dysbiosis-induced activation of the intrarenal renin-angiotensin system is involved in kidney injuries in rat diabetic nephropathy显示文摘Some studies have shown that gut microbiota along with its metabolites is closely associated with diabetic mellitus(DM).ln this study we explored the relationship between gut microbiota and kidney injuries of early diabetic nephropathy(DN)and its underlying mechanisms.Male SD rats were intraperitoneally injected with streptozotocin to induce DM.DM rats were orally administered compound broad-spectrum antibiotics for 8 wieeks.After the rats were sacrificed,their blood,urine,feces,and renal tssues were harvested for analyses.We found that compared with the control rats,DM rats had abnormal intestinalmicroflora,increased plasma acetate levels,increased proteinuria,thickened glomerular basement membrane,and podocyte foot process effacement in the kidneys.Furthermore,the protein levels of angiotensinⅡ,angiotensin-converting enzyme,and angiotensinⅡtype 1 receptor in the kidneys of DM rats were significantly increased.Administration of broad-spectrum antbiotics in DM rats not only completely killed most intestinal microflora,but also significanty lowered the plasma acetate levels,inhibited intrarenal RAS activation,and attenuated kidney damage.Finally,we showed that plasma acetate levels were positively correlated with intrarenal angiotensinⅡprotein expression(r=0.969,P<0.001).In conclusion,excessive acetate produced by disturbed gut microbiota might be involved in the kidney injuries of early DN through activating intrarenal RAS.Chen-chen Lu Ze-bo Hu Ru Wang Ze-hui Hong Jian Lu Pei-pei Chen Jia-xiu Zhang Xue-qi Li Ben-yin Yuan Si-jia Huang Xiong-zhong Ruan Bi-cheng Liu Kun-ling Ma 2020Acta Pharmacologica Sinica2020,41,8:12
5Effect of heparin on apoptosis in human nasopharyngeal carcinoma CNE2 cells显示文摘In order to study the mechanism of the effect of heparin on apoptosis in carcinoma cells, the nasopharyngeal carcinoma cell line CNE2 was used to identify the effect of heparin on apoptosis associated with the expression of c-myc, bax, bcl-2 proteins by use of Hoechst 33258 staining, terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), agarose gel electrophoresis, and flow cytometry, as well as Western blot analysis. The results showed that heparin induced apoptosis of CNE2 cells including the morphologic changes such as reduction in the volume, and the nuclear chromatin condensation, as well as the “ladder pattern” revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner.The number of TUNEL-positive cells was dramatically increased to 33.6±1.2% from 2.8±0.3% by treat ment with heparin in different concentrations (10~40 kU/L). The apoptotic index was increased to 32.5% from 3.5% by detecting SubG1 peaks on flow cytometry. 6Using a non-radioisotopic, quantitative TRAP-based method detecting telomerase activities in human hepatoma cells显示文摘A non-radioisotopic, quantitative TRAP-based telomerase activity assay was established mainly by using SYBR Green-I staining instead of radioisotope. Comparing with conventional radioisotope based method, it was better in reproducibility and accuracy. 7Elevated Resting Heart Rate is Associated with Dyslipidemia in Middle-aged and Elderly Chinese显示文摘Objective To study the relationship between resting heart rate and blood lipid level.Methods A total of 9 415 subjects aged≥40 years were included in the present study.Their resting heart rate was monitored and their serum levels of triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)were measured to define dyslipidemia according to the 2007 Chinese Guidelines on Prevention and Treatment of Dyslipidemia in Adults.Results The subjects were divided into group A with their resting heart rate<70 beats/min,group B with their resting heart rate=70-79 beats/min,group C with their resting heart rate=80-89 beats/min,and group D with their resting heart rate≥90 beats/min.High TG,TC,and LDL-C were presented across the resting heart rate(Ptrend<0.01).Multiple logistic regression analysis revealed that the risk of high TG and TC was higher in subjects with their resting heart rate≥90 beats/min than in those with their resting heart rate<70 beats/min(OR=1.42;95%CI:1.16-1.74 and OR=1.33;95%CI:1.09-1.64,respectively).Conclusion Elevated resting heart rate is associated with high TG and TC in middle-aged and elderly Chinese subjects.SUN Ji Chao HUANG Xiao Lin DENG Xin Ru LV Xiao Fei LU Jie Li CHEN Yu Hong BI Yu Fang WANG Wei Qing XU Min NING Guang 2014Biomedical and Environmental Sciences2014,27,8:5

10Inhibitory effects of lapachol on rat C6 glioma in vitro and in vivo by targeting DNA topoisomerase Ⅰ and topoisomerase Ⅱ显示文摘OBJECTIVE The aim of this study is to investigate the inhibitory effects of lapachol on rat C6 glioma both in vitro and in vivo,as well as the potential mechanisms.METHODS First,the model of C6 glioma in Wistar rats was established and verified by hemotoxylin and eosin staining,immunohistochemical staining and magnetic resonance imaging(MRI).Then different doses of lapachol were gavaged and tumor volumes of the C6 glioma were detected by MRI.The effects of lapachol on C6 cell proliferation,apoptosis and DNA damage were detected by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS)/phen-azinemethosulfate(PMS)assay,Hoechst33358 staining,AnnexinⅤ-FITC/PI staining,and comet assay.Effects of lapachol on topoisomeraseⅠ(TOPⅠ)and topoisomeraseⅡ(TOPⅡ)activities were detected by TOPⅠand TOPⅡmediated supercoiled p BR322 DNA relaxation assay.Molecular docking was used to predict the interaction of lapachol-TOPⅠand lapachol-TOPⅡ.TOP I and TOPⅡexpression levels in C6 cells were determined by Enzymelinked immunosorbent assay kits and real-time polymerase chain reaction(RT-PCR).RESULTS The rat C6 glioma model was successfully established.High dose lapachol showed significant inhibitory effect on the C6 glioma in Wistar rats(P<0.05).MTS/PMS assay,Hoechst 33258 staining,AnnexinⅤ-FITC/PI staining,and comet assay showed that lapachol could inhibit proliferation,induce apoptosis and DNA damage of C6 cells in dose dependent manners.Lapachol could inhibit the activities of both TOPⅠandⅡ.Molecular docking showed that lapachol-TOPⅠshowed relatively stronger interaction than that of lapachol-TOPⅡ.Enzyme-linked immunosorbent assay and RT-PCR showed that lapachol could inhibit TOPⅡexpression levels,but not TOPⅠexpression levels.CONCLUSION These results showed that lapachol could significantly inhibit C6 glioma both in vivo and in vitro,which might be related with inhibiting TOPⅠand TOPⅡactivities,as wel as TOPⅡexpression.XU Huan-li CHEN Qun-ying WANG Hong XU Ping-xiang YUAN Ru LI Xiao-rong BAI Lu XUE Ming 2016中国药理学与毒理学杂志2016,30,10:3

12Serum Calcium Level is Associated with Brachial-ankle Pulse Wave Velocity in Middle-aged and Elderly Chinese显示文摘Objective To study the relation between serum calcium level and elevated BaPWV in Chinese subjects.Methods The relation between serum calcium level and elevated BaPWV was studied in 9 615subjects.The mean value of left and right BaPWV was analyzed.BaPWV was defined as high when it was?1 752.5 cm/s(the upper quartile)either side.Results The BaPWV and its elevated percentage progressively increased across the quartiles of the serum calcium level(P<0.05).The prevalence of elevated BaPWV was significantly higher in subjects of the second,third and highest quartiles than in those of the lowest quartile(26.9%,28.4%,and 33.2%vs23.7%,P=0.0116,P=0.0004,and P<0.0001).Logistic regression analysis revealed that the risk of elevated BaPWV was 1.32-fold higher in subjects of the highest quartile than in those of the lowest quartile(OR=1.32,95%CI:1.08-1.60).Conclusion The elevated serum calcium level is related to an elevated BaPWV and a higher risk of arterial stiffness,independent of conventional risk factors,in middle-aged and elderly Chinese subjects.DENG Xin Ru ZHANG Yin Fei WANG Tian Ge XU Bai Hui SUN Ji Chao ZHAO Lie Bin XU Min CHEN Yu Hong WANG Wei Qing BI Yu Fang LU Jie Li 2014Biomedical and Environmental Sciences2014,27,8:2

14An Encapsulation of the Rare Earth Complex in Modified Mesoporous Molecular Sieve MCM-41显示文摘The encapsulation of a rare earth (RE) complex Eu(DBM)3phen in modified Si- MCM41 with 3 -aminopropyltriethoxy st.lane is reported for the first time. 17Influence of Iron Supplementation on DMT1(IRE)-induced Transport of Lead by Brain Barrier Systems in vivo显示文摘Objective To investigate the potential involvement of DMT1(IRE) protein in the brain vascular system in vivo during Pb exposure. Methods Three groups of male Sprague-Dawley rats were exposed to Pb in drinking water, among which two groups were concurrently administered by oral gavage once every other day as the low and high Fe treatment group, respectively, for 6 weeks. At the same time, the group only supplied with high Fe was also set as a reference. The animals were decapitated, then brain capillary-rich fraction was isolate from cerebral cortex. Western blot method was used to identify protein expression, and RT-PCR to detect the change of the m RNA. Results Pb exposure significantly increased Pb concentrations in cerebral cortex. Low Fe dose significantly reduced the cortex Pb levels, However, high Fe dose increased the cortex Pb levels. Interestingly, changes of DMT1(IRE) protein in brain capillary-rich fraction were highly related to the Pb level, but those of DMT1(IRE) m RNA were not significantly different. Moreover, the consistent changes in the levels of p-ERK1/2 or IRP1 with the changes in the levels of DMT1(IRE). Owing to the improvement of molecular biological technique, there are some updated findings on the characteristics of PDA.
