| 1 | Protective effects of Yindanxinnaotong capsule in a rat model of myocardial ischemia/reperfusion injury显示文摘OBJECTIVE: To investigate the effects of Yindanxinnaotong capsule(YDXNTC) and main components compatibility and ratios on myocardium against ischemia/reperfusion injury and the effect's underlying mechanism.METHODS: Myocardial ischemia/reperfusion injury(MIRI) was induced by ischemia for 30 min and reperfusion for 30 min. Electrocardiogram data and coronary flow were recorded, and superoxide dismutase(SOD), malondialdehyde(MDA), lactate dehydrogenase, creatine kinase-MB, cardiac troponin T and I(cT nT, cT n I) and interleukin-1β, interleukin-8,interleukin-18(IL-1β, IL-8, IL-18) in myocardium were measured. Hypoxia/reoxygenation and hydrogen peroxide(H2O2) injury were induced by hypoxia for 3 h/reoxygenation for 2 h, and 100 μM H2O2 for 1 h, respectively, in vitro rat myocardial cells(H9c2). Cell viability, SOD, MDA, cT nT and inflamma-tory factors(IL-1β, IL-8 and IL-18) were determined,and Toll-like receptor 4(TLR-4) expression was measured by western blotting.RESULTS: In the isolated heart experiment, elevated heart function, coronary flow and SOD levels,and decreased MDA levels and inflammatory factors were noted in the YDXNTC, main components and main components compatibility groups. Ventricular tachycardia/ventricular fibrillation occurrence decreased in the ginkgo biloba extract(GBE),and GBE and salvia miltiorrhiza ethanol extract compatibility(SM-E, GSEC) groups. Lactic dehydrogenase levels decreased in the YDXNTC and aqueous extract of salvia miltiorrhiza(SM-H) groups. Creatine kinase-MB decreased with GBE, SM-E, SM-H and GSEC treatment, and cT n I and cT nT levels decreased with GSEC. In the in vitro cell study,YDXNTC and main components ratios improved cell viability and SOD levels, and suppressed MDA,cT nT and inflammatory factors. TLR-4 expression was down-regulated.CONCLUSION: YDXNTC and main components compatibility showed protective effects on MIRI in this rat model and in vitro study. | 2 | Developing controllable hypermutable Clostridium cells through manipulating its methyl-directed mismatch repair system显示文摘Development of controllable hypermutable cells can greatly benefit understanding and harnessing microbial evolution.However,there have not been any similar sys-tems developed for Clostridium,an important bacterial genus.Here we report a novel two-step strategy for de-veloping controllable hypermutable cells of Clostridium acetobutylicum,an important and representative indus-trial strain.Firstly,the mutS/L operon essential for methyl-directed mismatch repair(MMR)activity was inactivated from the genome of C.acetobutylicum to generate hy-permutable cells with over 250-fold increased mutation rates.Secondly,a proofreading control system carrying an inducibly expressed mutS/L operon was constructed.The hypermutable cells and the proofreading control system were integrated to form a controllable hypermut-able system SMBMutC,of which the mutation rates can be regulated by the concentration of anhydrotetracycline(aTc).Duplication of the miniPthl-tetR module of the proof-reading control system further signifi cantly expanded the regulatory space of the mutation rates,demonstrating hypermutable Clostridium cells with controllable mutation rates are generated.The developed C.acetobutylicum strain SMBMutC2 showed higher survival capacities than the control strain facing butanol-stress,indicating greatly increased evolvability and adaptability of the controllable hypermutable cells under environmental challenges. | Guodong Luan Zhen Cai Fuyu Gong Hongjun Dong Zhao Lin Yanping Zhang Yin Li | 2013 | Protein & Cell2013,4,11: | 4 |

| 4 | Methane in soil gas and its transfer to the atmosphere in the Yakela condensed gas field in the Tarim Basin,Northwest China显示文摘In this study,by analyzing CH4 concentration and δ13CCH4 in soil-gas profiles,the potentials of CH4 gas transfer from ground to atmosphere were studied at four representative sectors in the Yakela condensed gas field in the Tarim Basin,Xinjiang,China.These are:1) the oil-gas interface sector,2) fault sector,3) oil-water interface sector,4) an external area.Variation in CH4 in soil-gas profiles showed that CH4 microseepage resulted from the migration of subsurface hydrocarbon from deep-buried reservoirs to the earth's surface.It was found that CH4 from deep-buried reservoirs could migrate upwards to the surface through faults,fissures and permeable rocks,during which some CH4 was oxidized and the unoxidized methane remained in the soil or was emitted into the atmosphere.The lowest level of CH4 at the soil-gas profile was found at the CH4 gas-phase equilibrium point at which the CH4 migration upwards from deep-buried reservoirs and the CH4 diffusion downwards from the atmosphere met.The δ13CCH4 and ethane,propane in soil gas exhibited thermogenic characteristics,suggesting the occurrence of CH4 microseepage from deep-buried reservoirs.A linear correlation analysis between CH4 concentrations in soil gas and temperature,moisture,pH,Eh,Ec and particle size of soil indicated that both soil Eh and soil temperature could affect CH4 concentration in soil gas while soil pH could indirectly influence soil methanotrophic oxidation via impacting soil Eh. | Tang Junhong Wang Guojian Yin Haoyong Li Hongjun | 2013 | Petroleum Science2013,10,2: | 2 |

| 18 | A phylogenetic approach identifies patterns of beta diversity and floristic subregions of the Qinghai-Tibet Plateau显示文摘Patterns of taxonomic and phylogenetic beta diversity and their relationships with environmental correlates can help reveal the origin and evolutionary history of regional biota.The Qinghai-Tibet Plateau(QTP)harbors an exceptionally diverse flora,however,a phylogenetic perspective has rarely been used to investigate its beta diversity and floristic regions.In this study,we used a phylogenetic approach to identify patterns of beta diversity and quantitatively delimit floristic regions on the Qinghai-Tibet Plateau.We also examined the relationships between multifaceted beta diversity,geographical distance,and climatic difference,and evaluated the relative importance of various factors(i.e.,climate,topography and history)in shaping patterns of beta diversity.Sørensen dissimilarity indices indicated that patterns of species turnover among sites dominated the QTP.We also found that patterns of both taxonomic and phylogenetic beta diversity were significantly related to geographical distance and climatic difference.The environmental factors that contributed most to these patterns of beta diversity include annual precipitation,mean annual temperature,climatic gradients and climatic instability.Hierarchical dendrograms of dissimilarity and non-metric multidimensional scaling ordination based on phylogenetic beta diversity data identified ten floristic subregions in the QTP.Our results suggest that the contemporary environment and historical climate changes have filtered species composition among sites and eventually determined beta diversity patterns of plants in the QTP. | Haibin Yu Man Yang Zixin Lu Weitao Wang Fangyuan Yu Yonghua Zhang Xue Yin Hongjun Yu Junjie Hu David C.Deane | 2024 | Plant Diversity2024,46,1: | 0 |
