| 1 | Downregulation of lncRNA AFAP1-AS1 by oridonin inhibits the epithelial-to-mesenchymal transition and proliferation of pancreatic cancer cells显示文摘Recent studies have demonstrated that the expression of the long non-coding RNA(lncRNA)AFAP1-AS1 in pancreatic cancer is negatively correlated with survival and prognosis.However,the effects of oridonin and lncRNA AFAP1-AS1 on the epithelial-to-mesenchymal transition(EMT)and migration of pancreatic cancer cells have not been fully elucidated.Surgery is the only potentially curative method for pancreatic cancer,but postoperative recurrence and metastasis are common.The aim of the present study was to assess the effect of oridonin and lncRNA AFAP1-AS1 silencing on pancreatic cancer cells.The pancreatic cancer cell lines BxPC-3 and PANC-1 cells were transfected with siAFAP1-AS1 and its negative control(siNC).After that,oridonin was used to treat the siAFAP1-AS1-transfected cells.The expression of lncRNA AFAP1-AS1 was downregulated in the pancreatic cancer cell lines BxPC-3 and PANC-1.The apoptosis and cell cycle progression of pancreatic cancer cells were evaluated by flow cytometry and Hoechst 33258 staining.Metastasis and invasion of BxPC-3 and PANC-1 cells were detected by transwell migration assay,real-time cell analysis,and western blot analysis.Cells were transfected with the lentiviral siAFAP1-AS1 and siNC,and tumorigenesis was evaluated in BALB/C nude mice.Immunohistochemical examination was used to verify the effects of oridonin and siAFAP1-AS1 on pancreatic cancer.The results demonstrated that the combination of oridonin and siAFAP1-AS1 inhibited pancreatic cancer cell proliferation,induced apoptosis,arrested cell cycle progression,prevented the migration,regulated EMT-related protein expression in BxPC-3 and PANC-1 cells,and inhibited pancreatic cancer cell tumorigenicity and EMT in nude mice. | Songmei Lou Jian Xu Bili Wang Shuquan Li Jun Ren Zhengjun Hu Bin Xu Feng Luo | 2019 | Acta Biochimica et Biophysica Sinica2019,51,8: | 10 |

| 4 | Realignment of Slanted Fe Nanorods on Silicon Substrates by a Strong Magnetic Field显示文摘Slanted Fe nanorods prepared by glancing angle deposition on silicon substrates exhibited easy magnetization along their growth axis. By using a thin gold film on a silicon substrate as a buffer layer, slanted Fe nanorods can be realigned towards the substrate surface normal by a strong magnetic field. After realignment, the Fe nanorods retained the easy magnetization axis along their growth axis. The effects of the realignment by the strong magnetic field on the properties of the slanted Fe nanorods were also investigated. | 9 | A Case Report:Severe Bullous Skin Reaction Induced by Anti-PD-1 Antibody(Toripalimab)Therapy in a Patient with Thymic Carcinoma显示文摘The development of immune checkpoint inhibitors,such as those targeting programmed cell death protein 1(PD-1),represents a major breakthrough in cancer therapy.Although immune checkpoint blockade therapy has a favorable risk/benefit ratio,it causes significant immune-related adverse events(irAEs),such as cutaneous reactions,in particular,severe bullous skin reactions and toxic epidermal necrolysis.Here,we report a case of a 51-year-old woman with malignant thymoma who developed a severe bullous skin reaction(characterized by a systemic rash,bullae,epidermal desquamation,and Stevens-Johnson syndrome)as a result of treatment with the PD-1 inhibitor toripalimab.The patient was treated with high doses of glucocorticoid,intravenous immunoglobulin,and intensive care,and eventually recovered from the severe irAEs.The intravenous injection of anti-PD-1 antibodies induces cutaneous reactions,which are associated with higher mortality rates.High doses of glucocorticoid combined with intravenous immunoglobulin are effective in alleviating such irAEs.Thus,improving the level of care and preventing skin infections can effectively reduce the risk of death. | Chong TENG Xiaowei SONG Chengjuan FAN Yashuang ZHAO Lina DU Siqi MAN Yuanyuan HU Zhengjun JIANG Tao XIN | 2022 | Clinical Cancer Bulletin2022,1,3: | 0 |
