1The expressions of Bcl-2 and Bax in gastric carcinoma and precancerous lesions显示文摘Objective: To study the expressions of Bcl-2 and Bax proteins and explore the relationships between them and apoptosis in gastric carcinoma and precancerous lesions. Methods: TUNEL method was used to detect apoptosis and im- munohistochemical staining method was used to detect the expressions of Bcl-2 and Bax proteins in 70 cases of chronic gastritis, 49 cases of intestinal metaplasia, 64 cases of dysplasia and 81 cases of gastric carcinoma. Results: The positive incidences of Bcl-2 and Bax were the highest in severe dysplasia. In intestinal metaplasia and dysplasia, positive incidences of Bcl-2 and Bax were higher than those in chronic gastritis and gastric carcinoma. During the process of evolvement from chronic gastritis to intestinal metaplasia then to dysplasia, apoptosis indexes gradually rise up to mid-high dysplasia, which reached the highest point, then it began to fall down, when it reached the lowest point in gastric carcinoma. There were nega- tive correlations between the expressions of Bcl-2, Bax proteins and apoptosis. Conclusion: The expression of Bcl-2 may be an early behavior in the gastric carcinogenesis. Bax had antagonistic effect to Bcl-2. 4肿瘤坏死因子α致肝细胞凋亡效应及其机制的研究(英文)显示文摘Objective: To investigate the apoptosis effect of TNF-α on HepG2 cells and its mechanism in vitro. Methods: HepG2 cells were treated with TNF-α (400 U/mL). HepG2 cells treated with TNF-α for 24 h and apoptosis was proved by DNA fragments on gel electrophoresis. Fluorescent quantitative real-time PCR was used to detect Fas and FasL expression. HepG2 cells treated by TNF-α were co-cultivated with normal HepG2 cells. Apoptosis of HepG2 cells was determined by the method of FACS. Results: After 24 h TNF-α treatment, DNA was collapsed into fragments to form DNA ladder in gel elec- trophoresis; FasL expression increases and induces HepG2 cells apoptosis. By FACS, 98.4% of the co-cultivated cells were apoptosis, but 16.5% cells in control group were apoptosis. Conclusion: TNF-α can induce apoptosis of HepG2 cells in vitro. FasL expression on TNF-α pre-treated HepG2 cells increased and these cells can lead normal HepG2 cells to apoptosis. 9Residue substitution enhances the immunogenicity of neoepitopes from gastric cancers显示文摘Objective:Neoantigens arising from gene mutations in tumors can induce specific immune responses,and neoantigen-based immunotherapies have been tested in clinical trials.Here,we characterized the efficacy of altered neoepitopes in improving immunogenicity against gastric cancer.Methods:Raw data of whole-exome sequencing derived from a patient with gastric cancer were analyzed using bioinformatics methods to identify neoepitopes.Neoepitopes were modified by P1Y(the first amino acid was replaced by tyrosine)and P2L(the second amino acid was replaced by leucine).T2 binding and stability assays were used to detect the affinities between the neoepitopes and the HLA molecules,as well as the stabilities of complexes.Dendritic cells(DCs)presented with neoepitopes stimulated naïve CD8+T cells to induce specific cytotoxic T lymphocytes.ELISA and carboxyfluorescein succinimidyl ester were used to detect IFN-γand TNF-αlevels,and T cell proliferation.Perforin was detected by flow cytometry.The cytotoxicity of T cells was determined using the lactate dehydrogenase assay.Results:Bioinformatics analysis,T2 binding,and stability assays indicated that residue substitution increased the affinity between neoepitopes and HLA molecules,as well as the stabilities of complexes.DCs presented with altered neoepitopes stimulated CD8+T cells to release more IFN-γand had a greater effect on promoting proliferation than wild-type neoepitopes.CD8+T cells stimulated with altered neoepitopes killed more wild-type neoepitope-pulsed T2 cells than those stimulated with wild-type neoepitopes,by secreting more IFN-γ,TNF-α,and perforin.Conclusions:Altered neoepitopes exhibited greater immunogenicity than wild-type neoepitopes.Residue substitution could be used as a new strategy for immunotherapy to target neoantigens.Huahui Yu Jieyu Li Yuan Yuan Yu Chen Jingwen Hong Chunmei Ye Wansong Lin Huijing Chen Zengqing Guo Bo Li Yunbin Ye 2021Cancer Biology & Medicine2021,18,4:0

11Distributed robust MPC for nonholonomic robots with obstacle and collision avoidance显示文摘Considering that the inevitable disturbances and coupled constraints pose an ongoing challenge to distributed control algorithms,this paper proposes a distributed robust model predictive control(MPC)algorithm for a multi-agent system with additive external disturbances and obstacle and collision avoidance constraints.In particular,all the agents are allowed to solve optimization problems simultaneously at each time step to obtain their control inputs,and the obstacle and collision avoidance are accomplished in the context of full-dimensional controlled objects and obstacles.To achieve the collision avoidance between agents in the distributed framework,an assumed state trajectory is introduced for each agent which is transmitted to its neighbors to construct the polyhedral over-approximations of it.Then the polyhedral over-approximations of the agent and the obstacles are used to smoothly reformulate the original nonconvex obstacle and collision avoidance constraints.And a compatibility constraint is designed to restrict the deviation between the predicted and assumed trajectories.Moreover,recursive feasibility of each local MPC optimization problem with all these constraints derived and input-to-state stability of the closed-loop system can be ensured through a sufficient condition on controller parameters.Finally,simulations with four agents and two obstacles demonstrate the efficiency of the proposed algorithm.Li Dai Yanye Hao Huahui Xie Zhongqi Sun Yuanqing Xia 2022Control Theory and Technology2022,20,1:0
