| 1 | Four new species of the genus Hymenoscyphus (fungi) based on morphology and molecular data显示文摘Four new species of Hymenoscyphus (H. brevicellulus, H. hyaloexcipulus, H. microcaudatus, and H. subsymmetricus) and a new Chinese record (H. subpallescens) are described. These five species share common characteristics: small apothecia (<3mm in diameter); hymenium whitish, pale yellow, to yellow in color; ectal excipulum of textura prismatica; asci arising from simple septa; ascospores scutuloid and guttulate; saprophytic nutrition; and leaf habitats, except for H. subsymmetricus, which grows on herbaceous stems. Phylogenetic analyses of internal transcribed spacer nuclear ribosomal DNA sequences, the universal DNA barcode for fungi, for 16 species in the genus indicated that these taxa were closely related to H. microserotinus, in accordance with their morphological features, but represented independent species. | 3 | Linkage of Aromatic Ring Structures in Saturates, Aromatics, Resins and Asphaltenes Fractions of Vacuum Residues Determined by Collision-Induced Dissociation Technology显示文摘The linkage of aromatic ring structures in vacuum residues was important for the refining process. The Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR MS) combined with collision-induced dissociation(CID) is a powerful method to characterize the molecular structure of petroleum fractions. In this work, model compounds with different aromatic ring structures were measured by CID FT-ICR MS. The cracking of the parent ions and the generated fragment ions were able to distinguish different linkage of the model compounds. Then, vacuum residues were separated into saturates, aromatics, resins and asphaltenes fractions(SARA), and each fraction was characterized by CID technology. | 5 | In-Situ Chemosynthesis of ZnO Nanoparticles to Endow Wood with Antibacterial and UV-Resistance Properties显示文摘Hybrid wood materials have attracted considerable attention because they have combined advantages of both wood and inorganic compounds. This work investigated the microstructural morphology, thermal stability, ultraviolet(UV) stability, and antibacterial property of composites made from wood/ZnO hybrid materials through a facile in-situ chemosynthesis methods. The X-ray diffraction(XRD) and thermogravimetric analysis(TGA) results indicated that the synthesized ZnO particles had an average grain size of about 10.8 nm. The scanning electron microscopy(SEM) observations showed that ZnO nanoflowers self-assembled with nanosheets were presented in wood cell lumens and increased with increasing Zn^(2+)concentrations. ZnO nanoparticles were also generated in the wood cell wall, which was confirmed by the results of energy-dispersive spectroscopy(EDS). The TGA tests also indicated that the thermal stability of wood/ZnO hybrid materials was improved after the formation of ZnO inorganic particles. | 7 | Structural Characterization of Petroleum Molecules by CID FT-ICR MS with Narrow Isolation Window显示文摘High resolution tandem mass spectrometry has been applied to obtain the structure information of petroleum samples. Here, we report a method for structural characterization of crude oil molecules by the collision-induced dissociation(CID) technology coupled with the high-field Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR MS). The ion isolation window was narrowed down to 1 Da to distinguish the complex homologues contained in petroleum. Aromatic model compounds and crude oil samples were measured by CID FT-ICR MS at different collision energy levels. The fragmentation of model compounds with alkyl side-chains was found to be related to the size of the aromatic rings. The fragmentation of model compounds with archipelago structures depended on the length of the bridge alkylene chain. The prevalent reaction pathway of model compounds with naphthenic rings was mainly determined by the position of naphthenic rings in the molecules. On the basis of the fragmentation pathways, the structure differences of two crude oils were recognized as different content of naphthenic rings by CID technology with 1 Da isolation window. The NMR analysis was also applied to confirm the CID results. | 8 | HDAC I/IIb selective inhibitor Purinostat Mesylate combined with GLS1 inhibition effectively eliminates CML stem cells显示文摘Purinostat Mesylate(PM)is a novel highly selective and active HDAC I/IIb inhibitor,and the injectable formulation of PM(PMF)based on the compound prescription containing cyclodextrin completely can overcome PM’s poor solubility and improves its stability and pharmacokinetic properties.Here,we showed that PM effectively repressed the survival of Ph+leukemia cells and CD34+leukemia cells from CML patients in vitro.In vivo studies demonstrated that PMF significantly prevented BCR-ABL(T315I)induced CML progression by restraining leukemia stem cells(LSCs),which are insensitive to chemotherapy and responsible for CML relapse.Mechanism studies revealed that targeting HDAC I/IIb repressed several important factors for LSCs survival including c-Myc,β-Catenin,E2f,Ezh2,Alox5,and mTOR,as well as interrupted some critical biologic processes.Additionally,PMF increased glutamate metabolism in LSCs by increasing GLS1.The combination of PMF and glutaminase inhibitor BPTES synergistically eradicated LSCs by altering multiple key proteins and signaling pathways which are critical for LSC survival and self-renewal.Overall,our findings represent a new therapeutic strategy for eliminating LSCs by targeting HDAC I/IIb and glutaminolysis,which potentially provides a guidance for PMF clinical trials in the future for TKI resistance CML patients. | Qiang Qiu Linyu yang Yunyu Feng Zejiang Zhu Ning Li Li Zheng Yuanyuan Sun Cong Pan Huandi Qiu Xue Cui Wei He Fang Wang Yuyao Yi Minghai Tang Zhuang Yang Yunfan Yang Zhihui Li Lijuan Chen Yiguo Hu | 2023 | Bioactive Materials2023,,3: | 1 |

| 12 | A Qualitative Study of the Triage of Patients with Non-Traumatic Acute Abdomen显示文摘Objective: To explore the work experience of medical staff in the emergency department of a general hospital in the triage of patients with non-traumatic acute abdomen to formulate corresponding intervention measures and branch evaluation tools. Methods: With descriptive phenomenology as the research method, semi-structured interviews were conducted with the medical staff in a tertiary hospital in Nanjing from February 1st to 10th, 2023, and Colaizzi seven-step analysis was used to analyze the data. Results: A total of 17 emergency medical staff were interviewed in this study. Four themes were derived from the analysis of the data: the etiology of acute abdomen is complex, so it is difficult to categorize them: acute abdomen requires immediate treatment, but the treat will be delayed if the categorization is inaccurate;the high pressure of nurses and the accuracy in categorizing the patients are problems that should be addressed. Conclusion: The categorization of patients with non-traumatic acute abdomen is challenging. | 13 | Effect of Co-substrate on Degradation of Benzothiazole in MEC显示文摘Due to its persistence and bio-toxicity,benzothiazole(BTH) cannot be biodegraded efficiently.Recent work has shown that removal rates of biorefractory organics can be enhanced by the addition of cosubstrates.In this work,ethanol,acetate,propionate and butyrate were added as co-substrates in order to promote the degradation of BTH in microbial electrolysis cell(MEC).By probing the changes in degradation rates of BTH in the presence of different co-substrates,it was observed that all the four co-substrates can enhance the BTH degradation in MEC,both the efficiency(EBTH) and the rate(RBTH).It was also found that acetate is more effective than others,which made the degradation efficiency of BTH up to 90% with acetate-C at350 mg/L(measuring by the carbon content of co-substrate,the same below),within 6 h and the degradation rate of BTH arrived 0.001 2/(mg·h).The microbacteria in MEC have also been influenced by different cosubstrates.This metabolism of the co-substrates enables the microbacteria on anode to generate ATP and thus grow to ensure the microbacteria activity.Therefore,this work showed that the addition of co-substrates such as acetate can be a novel and efficient approach for improving the elimination of BTH from wastewaters by MEC system. | Huandi Huang Jie Ding Xianshu Liu Guojun Xie Dihui Song | 2016 | Journal of Harbin Institute of Technology(New Series)2016,23,5: | 0 |

| 15 | TSPAN32 suppresses chronic myeloid leukemia pathogenesis and progression by stabilizing PTEN显示文摘We report herein that TSPAN32 is a key node factor for Philadelphia (Ph+) leukemia pathogenesis. We found that TSPAN32expression was repressed by BCR-ABL and ectopic TSPAN32 expression upon Imatinib treatment inhibited the proliferation of Ph+cell lines. Tspan32 overexpression significantly prevented BCR-ABL induced leukemia progression in a murine model and impairedleukemia stem cell (LSC) proliferation. LSCs represent an obstacle for chronic myeloid leukemia (CML) elimination, which continuallyreplenish leukemia cells and are associated with disease relapse. Therefore, the identification of essential targets that contribute tothe survival and self-renewal of LSCs is important for novel curative CML. Mechanistically, TSPAN32 was shown to interact withPTEN, increased its protein level and caused a reduction in PI3K-AKT signaling activity. We also found that TSPAN32 was repressedby BCR-ABL via the suppression of an important transcription factor, TAL1. Ectopic expression of TAL1 significantly increasedTSPAN32 mRNA and protein level, which indicated that BCR-ABL repressed TSPAN32 transcription by decreasing TAL1 expression.Overall, we identified a new signaling axis composed of “BCR-ABL-TAL1-TSPAN32-PTEN-PI3K-AKT”. Our findings furthercomplement the known mechanisms underlying the transformation potential of BCR-ABL in CML pathogenesis. | 17 | Expression of Fragaria ananassa Osmotin-like Protein(FaOLP2) in E. coli:Purification and Antifungal Activity显示文摘[Objective]The FaOLP2(Fragaria ananassa osmotin-like protein)is a functional homolog of PR5-like protein.This study was undertaken to produce recombinant FaOLP2 and to identify its antifungal activity.[Method]The ORF of FaOLP2(accession number DQ325524)was cloned into pET22b vector to construct the pET22b-FaOLP2 plasmid.The recombinant mature FaOLP2 was expressed in E.coli Rosetta-gami B(DE3)by inducing with 1 mmol/L IPTG and found exclusively in insoluble inclusion bodies.As FaOLP2 requires the correct formation of eight disulfide bonds,but there were no obvious effect to correctly form these by expression at different temperatures and high osmotic pressure(supplemented Betaineand and D-Sorbitol),we used an in vitro method to refold E.coli expressed FaOLP2 by gradually elution using reduced:oxidized gluthatione redox buffer,followed by 8 mol/L urea solubilized His6-tagged mature FaOLP2 protein,which was affinity-purified by an immobilized-metal(Ni2+)affinity chromatography(IMAC)column.[Result]This method generated biologically active conformations of the recombinant mature FaOLP2 that displayed antifungal activity against Ustilaginoides virens,a plant pathogenic fungus,which causes rice false smut.[Conclusion]This study laid the foundation for further biotechnological application of the novel protein. | Bai LI Sufang LI Huandi LI Junwei SUN Keqin ZOU | 2013 | Agricultural Biotechnology2013,2,6: | 0 |
