1Use of recombinant microRNAs as antimetabolites to inhibit human non-small cell lung cancer显示文摘During the development of therapeutic microRNAs(miRNAs or miRs),it is essential to define their pharmacological actions.Rather,miRNA research and therapy mainly use miRNA mimics synthesized in vitro.After experimental screening of unique recombinant miRNAs produced in vivo,three lead antiproliferative miRNAs against human NSCLC cells,miR-22-3p,miR-9-5p,and miR-218-5p,were revealed to target folate metabolism by bioinformatic analyses.Recombinant miR-22-3p,miR-9-5p,and miR-218-5p were shown to regulate key folate metabolic enzymes to inhibit folate metabolism and subsequently alter amino acid metabolome in NSCLC A549 and H1975 cells.Isotope tracing studies further confirmed the disruption of one-carbon transfer from serine to folate metabolites by all three miRNAs,inhibition of glucose uptake by miR-22-3p,and reduction of serine biosynthesis from glucose by miR-9-5p and-218-5p in NSCLC cells.With greater activities to interrupt NSCLC cell respiration,glycolysis,and colony formation than miR-9-5p and-218-5p,recombinant miR-22-3p was effective to reduce tumor growth in two NSCLC patient-derived xenograft mouse models without causing any toxicity.These results establish a common antifolate mechanism and differential actions on glucose uptake and metabolism for three lead anticancer miRNAs as well as antitumor efficacy for miR-22-3p nanomedicine,which shall provide insight into developing antimetabolite RNA therapies.Yixin Chen Mei-Juan Tu Fangwei Han Zhenzhen Liu Neelu Batra Primo N.Lara Hong-Wu Chen Huichang Bi Ai-Ming Yu 2023Acta Pharmaceutica Sinica B2023,13,10:0
2Physical structure and phytoplankton community off the eastern Hainan coast during summer 2015显示文摘Based on satellite remote sensing dataset and survey data during upwelling season of 2015,the spatial structures of phytoplankton biomass and community for the first time in the eastern Hainan upwelling(EHU)and its adjacent area,the eastern Leizhou Peninsula upwelling(ELPU)were illustrated.It is found that a significant cold tongue with high salinity and low temperature along the eastern Hainan coast driven by upwelling-favorable summer monsoon.The ELPU was relative weaker than the EHU because of its wide and gentle continental slope.Due to mixing by tides and waves,DO concentration with high value(>6.0 mg/L)were almost homogenous from surface to 30 m depth at the EHU.Beneath that,low DO water(<6.0 mg/L,anoxia)were pumped upward from bottom by the upwelling.The ELPU has worse DO condition compared with the EHU where bottom DO were lower than 3.5 mg/L owing to abundant DO consumption.The phytoplankton biomass reached maximal value about 1.5 mg/m3 at 30 m depth layer rather than surface layer at the EHU indicating the impact limit of upwelling on phytoplankton growth and DO distribution.Nourished by rich nutrient input,the phytoplankton biomass at the ELPU were much higher than the EHU where the maximal value can reach about 4.0 mg/m3.The phytoplankton biomass were reduced to about 0.2–0.3 mg/m3 at the offshore areas of the EHU and ELPU which were close to the value at open sea.At the inshore of the EHU,the phytoplankton community was dominated by diatom which accounted for about 50%of phytoplankton biomass.And prokaryotes(about 40%),green algae(about 20%)and prochlorococcus(about 20%)became main species at the offshore of the EHU.At the ELPU,diatom accounted for about 80%of phytoplankton biomass followed by green algae,indicating a different ecosystem at this region compared with the EHU.Sumin Liu Bo Hong Guifen Wang Weiqiang Wang Qiang Xie Zekai Ni Liu Yu Huichang Jiang Tong Long Hongzhou Xu 2020Acta Oceanologica Sinica2020,39,11:0
3Overview of Earth-Moon Transfer Trajectory Modeling and Design显示文摘TheMoon is the only celestial body that human beings have visited.The design of the Earth-Moon transfer orbits is a critical issue in lunar exploration missions.In the 21st century,new lunar missions including the construction of the lunar space station,the permanent lunar base,and the Earth-Moon transportation network have been proposed,requiring low-cost,expansive launch windows and a fixed arrival epoch for any launch date within the launch window.The low-energy and low-thrust transfers are promising strategies to satisfy the demands.This review provides a detailed landscape of Earth-Moon transfer trajectory design processes,from the traditional patched conic to the state-of-the-art low-energy and low-thrust methods.Essential mechanisms of the various utilized dynamic models and the characteristics of the different design methods are discussed in hopes of helping readers grasp thebasic overviewof the current Earth-Moon transfer orbitdesignmethods anda deep academic background is unnecessary for the context understanding.Jiye Zhang Huichang Yu Honghua Dai 2023Computer Modeling in Engineering & Sciences2023,,4:0
