| 2 | Urinary donor-derived cell-free DNA as a non-invasive biomarker for BK polyomavirus-associated nephropathy显示文摘BK polyomavirus-associated nephropathy(BKPyVAN)is a common cause of allograft failure.However,differentiation between BKPyVAN and type I T cell-mediated rejection(TCMR)is challenging when simian virus 40(SV40)staining is negative,because of the similarities in histopathology.This study investigated whether donor-derived cell-free DNA(ddcfDNA)can be used to differentiate BKPyVAN.Target region capture sequencing was applied to detect the ddcfDNAs of 12 recipients with stable graft function,22 with type I TCMR,21 with proven BKPy VAN,and 5 with possible Py VAN.We found that urinary ddcfDNA levels were upregulated in recipients with graft injury,whereas plasma ddcfDNA levels were comparable for all groups.The median urinary concentrations and fractions of ddcfDNA in proven BKPyVAN recipients were significantly higher than those in type I TCMR recipients(10.4 vs.6.1 ng/mL,P<0.001 and 68.4%vs.55.3%,P=0.013,respectively).Urinary ddcfDNA fractions(not concentrations)were higher in the BKPyVAN-pure subgroup than in the BKPyVAN-rejection-like subgroup(81.30%vs.56.64%,P=0.025).With a cut-off value of 7.81 ng/m L,urinary ddcf DNA concentrations distinguished proven BKPyVAN from type I TCMR(area under the curve(AUC)=0.848,95%confidence interval(95%CI):0.734 to 0.963).These findings suggest that urinary ddcf DNA is a non-invasive biomarker which can reliably differentiate BKPy VAN from type I TCMR. | Jia SHEN Luying GUO Wenhua LEI Shuaihui LIU Pengpeng YAN Haitao LIU Jingyi ZHOU Qin ZHOU Feng LIU Tingya JIANG Huiping WANG Jianyong WU Jianghua CHEN Rending WANG | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,11: | 4 |

| 4 | Ataxia,acute mountain sickness,and high altitude cerebral edema显示文摘Previous investigations suggest that ataxia is common and often one of the most reliable warning signs of high altitude cerebral edema(HACE). The aim of this study was to investigate the diagnostic role of ataxia in acute mountain sickness(AMS)and HACE among mountain rescuers on the quake areas,and in approaching the relation between AMS and HACE. After the earthquake on April 14,2010,approximately 24 080 lowland rescuers were rapidly transported from sea level or lowlands to the mountainous rescue sites at 3 750 ~ 4 568 m,and extremely hardly worked for an emergency treatment after arrival. Assessments of acute altitude illness on the quake areas were using the Lake Louise Scoring System. 73 % of the rescuers were found to be developed AMS. The incidence of high altitude pulmonary edema(HAPE)and HACE was 0.73 % and 0.26 %,respectively,on the second to third day at altitude. Ataxia sign was measured by simple tests of coordination including a modified Romberg test. The clinical features of 62 patients with HACE were analyzed. It was found that the most frequent,serious neurological symptoms and signs were altered mental status(50/62,80.6 %)and truncal ataxia(47/62,75.8 %). Mental status change was rated slightly higher than ataxia,but ataxia occurred earlier than mental status change and other symptoms. The earliest sign of ataxia was a vague unsteadiness of gait,which may be present alone in association with or without AMS. Advanced ataxia was correlated with the AMS scores,but mild ataxia did not correlate with AMS scores at altitudes of 3 750~4 568 m. Of them,14 patients were further examined by computerized tomographic scanning of the brain and cerebral magnetic resonance imagines were examined in another 15 cases. These imaging studies indicated that the presence of the cerebral edema was in 97 % of cases who were clinically diagnosed as HACE(28/29). | 6 | Microbial community variation in phytoremediation of triazophos by Canna indica Linn. in a hydroponic system显示文摘Phytoremediation of triazophos (O,O-diethyl-O-(1-phenyl-1,2,4-triazole-3-base) sulfur phosphate, TAP) pollution by Canna indica Linn. in a hydroponic system has been well studied, whereas the microbial mechanism on TAP degradation is still unknown. The variation in microbial community compositions was investigated by analyzing phospholipid fatty acids (PLFAs) profiles in microbes under TAP exposure. The TAP exposure resulted in an increase in proportions of fatty acid 16:0 and decrease in fatty acid 18:2ω9,12c, indicating that TAP may stimulate the reproduction of microorganisms and inhibit the growth of fungi to some degree. Significant correlation was found between the ratio of fungi to bacteria and TAP removal (r 2 = 0.840, p < 0.01). In addition, the microbial community in the phytoremediation system with C. indica was dominated by Gram negative bacteria, which possibly contributed to the degradation of TAP. | 8 | Studies on monitoring hemodynamics and oxygen dynamics of adult respiratory distress syndrome secondary to high altitude pulmonary edema显示文摘To study monitoring hemodynamics and oxygen dynamics of adult respiratory distress syndrome (ARDS) secondary to high altitude pulmonary edema (HAPE),we performed clinic and laboratory studies in 8 patients who preliminarily developed high altitude cerebral edema (HACE) and then ARDS occurred at an altitude of 4 500 m. After an initial emergency treatment on high mountains,all the patients were rapidly transported to a hospital at a lower altitude of 2 808 m. The right cardiac catheterizations were carried out within 5 h after hospitalized. The monitoring hemodynamics and oxygen dynamics were studied via a thermodilution Swan-Gaze catheter. The results showed that before treatments at the beginning of monitoring,there presented a significant pulmonary artery hypertension with a decreased cardiac function,and a lower oxygen metabolism in all the 8 patients. However,after some effective treatments,including mechanical ventilation and using dexamethasone,furosemide,etc,four days later the result of a repeated monitoring showed that their pulmonary artery pressure had been decreased with an improved cardiac function with all the oxygen metabolic indexes increased significantly. | 9 | A comparison on the phytoremediation ability of triazophos by different macrophytes显示文摘The strategy of choosing suitable plants should receive great performance in phytoremediation of surface water polluted by triazophos(O,O-diethyl-O-(1-phenyl-1,2,4-triazol-3-base) sulfur phosphate, TAP), which is an organophosphorus pesticide widespread applied for agriculture in China and moderately toxic to higher animal and fish. The tolerance, uptake, transformation and removal of TAP by twelve species of macrophytes were examined in a hydroponic system and a comprehensive score(CS) of five parameters(relative growth rate(RGR), biomass, root/shoot ratio, removal capacity(RC), and bio-concentration factor(BCF)) by factor analysis was employed to screen the potential macrophyte species for TAP phytoremediation. The results showed that Thalia dealbata, Cyperus alternifolius, Canna indica and Acorus calamus had higher RGR values, indicating these four species having stronger growth capacity under TAP stress. The higher RC loading in Iris pseudacorus and Cyperus rotundus were 42.11 and 24.63 μg/(g fw·day), respectively. The highest values of BCF occurred in A. calamus(1.17), and TF occurred in Eichhornia crassipes(2.14). Biomass and root/shoot ratio of plant showed significant positive correlation with first-order kinetic constant of TAP removal in the hydroponic system, indicating that plant biomass and root system play important roles in remediation of TAP.
