1Systematically investigating the key features of the DNase deactivated Cpf1 for tunable transcription regulation in prokaryotic cells显示文摘With a unique crRNA processing capability,the CRISPR associated Cpf1 protein holds great potential for multiplex gene regulation.Unlike the well-studied Cas9 protein,however,conversion of Cpf1 to a transcription regulator and its related properties have not been systematically explored yet.In this study,we investigated the mutation schemes and crRNA requirements for the DNase deactivated Cpf1(dCpf1).By shortening the direct repeat sequence,we obtained genetically stable crRNA co-transcripts and improved gene repression with multiplex targeting.A screen of diversity-enriched PAM library was designed to investigate the PAMdependency of gene regulation by dCpf1 from Francisella novicida and Lachnospiraceae bacterium.We found novel PAM patterns that elicited strong or medium gene repressions.Using a computational algorithm,we predicted regulatory outputs for all possible PAM sequences,which spanned a large dynamic range that could be leveraged for regulatory purposes.These newly identified features will facilitate the efficient design of CRISPR-dCpf1 based systems for tunable multiplex gene regulation.Chensi Miao Huiwei Zhao Long Qian Chunbo Lou 2019Synthetic and Systems Biotechnology2019,4,1:1
2SERVQUAL Model Based Evaluation Analysis of Railway Passenger Transport Service Quality in China显示文摘Railway is the backbone of Chinese transportation system,but its poor quality of services for passengers cause complains now and then.This study first analyzed the influencing factors of service quality on railway passenger,and its quality characteristics was also explained,and finally we proposed an evaluation system of service quality on railway passenger transport.Through the statistical analysis and processing of the basic information from survey data from railway station,trains and the official website of the ticket purchase,the evaluation score of question naire was converted into the score in evaluation index system,which was based on SERVQUAL model.Finally,the evaluation index system was applied to the field test,and all levels of indicators and the overall evaluation of railway passenger transport service quality was obtained.The relevant results show that the evaluation model of this study is concise and practical,and the method has certain practicability and promotion value,which is beneficial to the department of management supervision in railway transportation.Huiwei Niu Jiao Yao Jing Zhao Jin Wang 2019Journal on Big Data2019,1,1:1
3A Study on Remediation Technologies for Cadmium Contamination in Greenhouse Vegetable Fields显示文摘Based on the literature and experimental results, three kinds of amendments, namely, rice biochar, hydroxyapatite and potassium dihydrogen phosphate(KH2PO4), and deeper ploughing were selected to evaluate the field application effect of soil amendments and agronomic measures on the remediation of Cd contamination in greenhouses. Cd-contaminated greenhouse screening was conducted from 2015 to 2017. In September 2017, comparative tests of eight treatments were performed in September 2017, and a selective test was performed in September 2018. The results of the screening of contaminated areas indicated that the distributions of over-standard sites and Cd content were significantly different. Greenhouse No. 4, in which the highest over-standard rate was 83.33% and the average content of Cd in soil was 0.535 mg/kg, was used as a comparative test experimental site for eight treatments. Greenhouse No. 1 was selected as the optimum test greenhouse for selective testing, with three plots having average Cd concentrations of 0.530, 0.568 and 0.792 mg/kg. The 8 months following comparative treatment showed that the content of available Cd following the treatment of rice biochar+hydroxyapatite+deeper ploughing(treatment T6) was reduced by 32.55% and 24.96% compared with the CK(the control) and 2 months following treatment, respectively. The content of available Cd using rice biochar+KH2PO4+deeper ploughing(treatment T7) treatment decreased by 47.88% and 31.00% compared with the CK and 2 months following treatment. The selective test results showed that in rice biochar+hydroxyapatite+deeper ploughing: the total Cd content of decreased from 0.489 to 0.372 mg/kg(23.86% lower), compared with the CK, and the mean content of available Cd decreased by 10.71% after 8 months, which was the lowest available Cd measured(0.133 mg/kg). In addition, the bean plant Cd content, bioconcentration factor(BCF) and translocation coefficient(TF) decreased by 15.86%, 23.68% and 25.77%, respectively. 4A survey of public needs for government information service in libraries显示文摘Purpose:This paper describes a comprehensive user survey carried out in China to investigate public needs for government information service in libraries.It is expected to provide insights into the ways to help public libraries meet the user needs.Design/methodology/approach:The paper is founded on a survey among library users,librarians and non-library users.Paper questionnaires were distributed in the National Library of China and 32 provincial public libraries in China and electronic questionnaires were sent to non-library users.A total of 909 valid questionnaires were collected.Findings:A lot of attention is paid to policies,laws and regulations relating to common people's work and lives such as science,technology,education,culture,finance,market supervision,health care,sports,urban construction and environment protection.A wide variety of government information resources is needed and information delivered to users shall be accurate and timely.Internet has become the main means of accessing government information and mobile phones the preferred Web access channel,while printed materials are considered important information sources.Users need assistance and training in the use of library information resources and services.Research limitations:The sample was limited to respondents from provincial-level public libraries,but city and county-level library users were not included.Practical implications:The survey results are beneficial for the Chinese public libraries to design service programs and improve their services.Originality/value:The paper is one of the comprehensive surveys on public needs for government information service in the Chinese libraries and some of die findings will be of wider interest for other libraries.Yunpeng QU Huiwei LIANG Lei SA Yumei XU Wenjie DUN Rongrong ZHAO Qiuhui CHEN 2014Chinese Journal of Library and Information Science2014,,1:0

In addition, the Cd content, bioconcentration factor(BCF) and translocation coefficient(TF) of bean aboveground plants were decreased by 15.86%, 23.68% and 25.77%, respectively when compared with CK.
