2Analysis of the intestinal flora in patientswith primary liver cancer显示文摘Objective To investigate the differences in intestinal flora of patients with primary liver cancer and ofhealthy individuals and to investigate the effect of the differential flora on the development of liver cancer.Methods Overall, 67 patients with primary liver cancer who received systematic and complete treatmentbetween January 2019 and December 2020 at the Sixth People’s Hospital of Qingdao and had completeclinical data were enrolled in this study, and 26 individuals who were healthy on physical examination inthe same period were used as healthy controls. Macro genome and 16s ribosome Deoxyribo Nucleic Acid(rDNA) high-throughput sequencing were performed on the stool flora of the enrolled patients and controls,and the differences in the intestinal flora were analyzed using the LEfSe bioinformatics software.Results Compared with the control samples, all the tested patient samples showed statistically significantdifferences in the number of colonies of 5 bacterial phyla, 5 orders, 8 families, 11 genera, and 14 species(P < 0.05).Conclusion Compared with healthy people, patients with primary liver cancer have significant differencesin the intestinal flora composition. 3Correlation analysis of breast fibroadenoma and the intestinal flora based on 16S rRNA sequencing显示文摘Objective To analyze the characteristics of the intestinal microflora in patients with breast fibroadenoma using 16S ribosomal RNA(rRNA)high-throughput sequencing.Methods Fecal samples from 20 patients with breast fibroadenoma and 36 healthy subjects were randomly collected and analyzed using high-throughput sequencing technology for 16S rRNA V4 region sequencing,and the alpha diversity(Chao index,Shannon index)was calculated using Mothur(v.1.39.5)software.Beta diversity was analyzed using QIIME(v1.80).SPSS software(version 23.0)and the t-test of two independent samples were used to analyze differences in the abundance of bacteria between the two groups.Results Compared with that in the healthy control group,theαdiversity of the intestinal microflora in breast fibroadenoma patients increased,but the difference was not statistically significant(P>0.05).At the phylum level,significant differences were observed between the two groups.The abundance of Firmicutes was higher in the breast fibroadenoma group(P<0.05),whereas the abundance of Synergistetes was higher in the healthy control group(P<0.005).A total of five bacterial genera showed significant differences between the two groups:the breast fibroadenoma group showed higher levels of Bautia(P<0.005),Coprococcus(P<0.005),Roseburia(P<0.05),and Ruminococcus(P<0.005),whereas Sutterella was more abundant in the healthy control group than in the breast fibroadenoma group(P<0.05).Conclusion The diversity and abundance of the intestinal flora in patients with breast fibroadenoma are significantly different from those in healthy subjects,suggesting that an imbalance in the intestinal flora is correlated with the occurrence of breast fibroadenoma.Bingdong Wang Xin Liu Yahong Bian Guoxin Sun Huizhe Wang Jingjin Zhang Zhengfu Zhang Xiao Zou 2021Oncology and Translational Medicine2021,7,6:0
4Peripheral nerve allograft prepared by an acellular,hypotonic tissue engineering method显示文摘BACKGROUND:Tissue engineered acellular nerves are good autologous nerve substitutes.Acellular peripheral nerves prepared using a conventional chemical extraction method cause a great deal of damage to nerve structures,and the allograft affects the nerve regeneration following transplantation.OBJECTIVE:To prepare peripheral nerve grafts through an acellular tissue engineering method,and observe their histology,ultrastructure,protein components and histocompatibility.DESIGN,TIME AND SETTING:A randomized,controlled,in vivo nerve tissue engineering experiment was performed at the Department of Biochemistry and Molecular Biology,Shenyang Medical College,China,from September 2006 to June 2007.MATERIALS:Triton X-100,Pepstatin A,Aprotinin and Leupeptin were purchased from Sigma,USA;Tris(hydroxymethyl) aminomethane was purchased from Gibco,USA.METHODS:The bilateral sciatic nerves of Wistar rats were harvested,treated with 0.05 mol/L Tris-HCl buffer,followed by proteinase inhibitor and Triton X-100 to prepare acellular peripheral nerves.The nerves were implanted in the quadriceps femoris muscle of healthy Wistar rats.MAIN OUTCOME MEASURES:Tissue structure and ultrastructure of acellular peripheral nerves were observed by optical microscopy and scanning electron microscopy.Growth associated proteins were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis.Nerve allograft and the surrounding muscles were observed by hematoxylin-eosin staining.RESULTS:Acellular treatment eliminated Schwann cells,epineurium or perineurium cells,myelin sheaths and axons of nerve fibers in normal peripheral nerves,while the spatial structure,comprising basement membrane tubes of Schwann cells and the extracellular matrix of perineurium and nerve fascicles was maintained.Protein bands at the region of 30 kD were no longer visible,had slightly decreased at 43 kD and remained unchanged at 65 kD.Following implantation for 7 days,epineurium cells were absorbed.However,increased fibroblasts,decreased newly-generated capillaries and maturation of granulation tissue were observed.CONCLUSION:The acellular nerve allograft prepared through the use of a hypotonic,acellular method displays good histocompatibility,eliminates immune substances and retains growth associated proteins that induce the growth of the neural axis.In addition,this method provides an ideal scaffold to construct artificial nerves.Huizhe Sun Wenying Dai Jie Wang Xiaojie Tong Wei Tian Jun Xu 2010Neural Regeneration Research2010,5,8:0
