2Sequencing a Juglans regia×J.microcarpa hybrid yields high-quality genome assemblies of parental species显示文摘Members of the genus Juglans are monecious wind-pollinated trees in the family Juglandaceae with highly heterozygous genomes,which greatly complicates genome sequence assembly.The genomes of interspecific hybrids are usually comprised of haploid genomes of parental species.We exploited this attribute of interspecific hybrids to avoid heterozygosity and sequenced an interspecific hybrid Juglans microcarpa×J.regia using a novel combination of single-molecule sequencing and optical genome mapping technologies.The resulting assemblies of both genomes were remarkably complete including chromosome termini and centromere regions.Chromosome termini consisted of arrays of telomeric repeats about 8 kb long and heterochromatic subtelomeric regions about 10 kb long.The centromeres consisted of arrays of a centromere-specific Gypsy retrotransposon and most contained genes,many of them transcribed.Juglans genomes evolved by a whole-genome-duplication dating back to the Cretaceous-Paleogene boundary and consist of two subgenomes,which were fractionated by numerous short gene deletions evenly distributed along the length of the chromosomes.Fractionation was shown to be asymmetric with one subgenome exhibiting greater gene loss than the other.The asymmetry of the process is ongoing and mirrors an asymmetry in gene expression between the subgenomes.Given the importance of J.microcarpa×J.regia hybrids as potential walnut rootstocks,we catalogued disease resistance genes in the parental genomes and studied their chromosomal distribution.We also estimated the molecular clock rates for woody perennials and deployed them in estimating divergence times of Juglans genomes and those of other woody perennials.Tingting Zhu Le Wang Frank M.You Juan C.Rodriguez Karin R.Deal Limin Chen Jie Li Sandeep Chakraborty Bipin Balan Cai-Zhong Jiang Patrick J.Brown Charles ALeslie Mallikarjuna KAradhya Abhaya M.Dandekar Patrick E.McGuire Daniel Kluepfel Jan Dvorak Ming-Cheng Luo 2019Horticulture Research2019,6,1:7
3Epidural electrical stimulation for spinal cord injury显示文摘A long-standing goal of spinal cord injury research is to develop effective repair strategies,which can restore motor and sensory functions to near-normal levels.Recent advances in clinical management of spinal cord injury have significantly improved the prognosis,survival rate and quality of life in patients with spinal cord injury.In addition,a significant progress in basic science research has unraveled the underlying cellular and molecular events of spinal cord injury.Such efforts enabled the development of pharmacologic agents,biomaterials and stem-cell based therapy.Despite these efforts,there is still no standard care to regenerate axons or restore function of silent axons in the injured spinal cord.These challenges led to an increased focus on another therapeutic approach,namely neuromodulation.In multiple animal models of spinal cord injury,epidural electrical stimulation of the spinal cord has demonstrated a recovery of motor function.Emerging evidence regarding the efficacy of epidural electrical stimulation has further expanded the potential of epidural electrical stimulation for treating patients with spinal cord injury.However,most clinical studies were conducted on a very small number of patients with a wide range of spinal cord injury.Thus,subsequent studies are essential to evaluate the therapeutic potential of epidural electrical stimulation for spinal cord injury and to optimize stimulation parameters.Here,we discuss cellular and molecular events that continue to damage the injured spinal cord and impede neurological recovery following spinal cord injury.We also discuss and summarize the animal and human studies that evaluated epidural electrical stimulation in spinal cord injury.Elliot H.Choi Sandra Gattas Nolan J.Brown John D.Hong Joshua N.Limbo Alvin Y.Chan Michael Y.Oh 2021Neural Regeneration Research2021,16,12:5

6Locomotor analysis identifies early compensatory changes during disease progression and subgroup classification in a mouse model of amyotrophic lateral sclerosis显示文摘Amyotrophic lateral sclerosis is a motoneuron degenerative disease that is challenging to diagnose and presents with considerable variability in survival.Early identification and enhanced understanding of symptomatic patterns could aid in diagnosis and provide an avenue for monitoring disease progression.Use of the m SOD1 G93 A mouse model provides control of the confounding environmental factors and genetic heterogeneity seen in amyotrophic lateral sclerosis patients,while investigating underlying disease-induced changes.In the present study,we performed a longitudinal behavioral assessment paradigm and identified an early hindlimb symptom,resembling the common gait abnormality foot drop,along with an accompanying forelimb compensatory mechanism in the m SOD1 G93 A mouse.Following these initial changes,m SOD1 mice displayed a temporary hindlimb compensatory mechanism resembling an exaggerated steppage gait.As the disease progressed,these compensatory mechanisms were not sufficient to sustain fundamental locomotor parameters and more severe deficits appeared.We next applied these initial findings to investigate the inherent variability in B6 SJL m SOD1 G93 A survival.We identified four behavioral variables that,when combined in a cluster analysis,identified two subpopulations with different disease progression rates:a fast progression group and a slow progression group.This behavioral assessment paradigm,with its analytical approaches,provides a method for monitoring disease progression and detecting m SOD1 subgroups with different disease severities.This affords researchers an opportunity to search for genetic modifiers or other factors that likely enhance or slow disease progression.Such factors are possible therapeutic targets with the potential to slow disease progression and provide insight into the underlying pathology and disease mechanisms.Melissa M.Haulcomb Rena M.Meadows Whitney M.Miller Kathryn P.Mc Millan Me Kenzie J.Hilsmeyer Xuefu Wang Wesley T.Beaulieu Stephanie L.Dickinson Todd J.Brown Virginia M.Sanders Kathryn J.Jones 2017Neural Regeneration Research2017,12,10:3

9GPR3,GPR6,and GPR12 as novel molecular targets:their biological functions and interaction with cannabidiol显示文摘The G protein-coupled receptors 3,6,and 12(GPR3,GPR6,and GPR12)comprise a family of closely related orphan receptors with no con firmed en dogenous liga nds.These receptors are con stitutively active and capable of signaling through G protein-mediated and non-G protein-mediated mechanisms.These orphan receptors have previously been reported to play important roles in many normal physiological functions and to be involved in a variety of pathological conditions.Although they are orphans,GPR3,GPR6,and GPR12 are phylogenetically most closely related to the cannabinoid receptors.Using β-arrestin2 recruitment and cAMP accumulati on assays,we recently found that the non psychoactive phytoca nnabinoid cann abidiol(CBD)is an inverse agonist for GPR3,GPR6,and GPR12.This discovery highlights these orphan receptors as potential new molecular targets for CBD,provides novel mechanisms of action,and suggests new therapeutic uses of CBD for illnesses such as Alzheimer's disease,Parkinson's disease,can cer,and infertility.Furthermore,identificati on of CBD as a new in verse agonist for GPR3,GPR6,and GPR12 provides the initial chemical scaffolds upon which potent and efficacious agents acting on these receptors can be developed,with the goal of developing chemical tools for studying these orphan receptors and ultimately new therapeutic agents.Alyssa S.Laun Sarah H.Shrader Kevin J.Brown Zhao-Hui Song 2019Acta Pharmacologica Sinica2019,40,3:2
10Effects of Climate Change and Shifts in Forest Composition on Forest Net Primary Production显示文摘Forests are dynamic in both structure and species composition,and these dynamics are strongly influenced by climate.However,the net effects of future tree species composition on net primary production (NPP) are not well understood.The objective of this work was to model the potential range shifts of tree species (DISTRIB Model) and predict their impacts on NPP (PnET-II Model) that will be associated with alterations in species composition.We selected four 200 × 200 km areas in Wisconsin,Maine,Arkansas,and the Ohio-West Virginia area,representing focal areas of potential species range shifts.PnET-II model simulations were carried out assuming that all forests achieved steady state,of which the species compositions were predicted by DISTRIB model with no migration limitation.The total NPP under the current climate ranged from 552 to 908 g C/m2 per year.The effects of potential species redistributions on NPP were moderate (-12% to +8%) compared with the influence of future climatic changes (-60% to +25%).The direction and magnitude of climate change effects on NPP were largely dependent on the degree of warming and water balance.Thus,the magnitude of future climate change can affect the feedback system between the atmosphere and biosphere.Jyh-Min Chiang Louts R.Iverson Anantha Prasad Kim J.Brown 2008Journal of Integrative Plant Biology2008,50,11:2

13Therapeutic potential of neuromodulation for demyelinating diseases显示文摘Neuromodulation represents a cutting edge class of both invasive and non-invasive therapeutic methods which alter the activity of neurons.Currently,several different techniques have been developed-or are currently being investigated–to treat a wide variety of neurological and neuropsychiatric disorders.Recently,in vivo and in vitro studies have revealed that neuromodulation can also induce myelination,meaning that it could hold potential as a therapy for various demyelinating diseases including multiple sclerosis and progressive multifocal leukencepalopathy.These findings come on the heels of a paradigm shift in the view of myelin's role within the nervous system from a static structure to an active co-regulator of central nervous system plasticity and participant in neuron-mediated modulation.In the present review,we highlight several of the recent findings regarding the role of neural activity in altering myelination including several soluble and contact-dependent factors that seem to mediate neural activitydependent myelination.We also highlight several considerations for neuromodulatory techniques,including the need for further research into spatiotemporal precision,dosage,and the safety and efficacy of transcranial focused ultrasound stimulation,an emerging neuromodulation technology.As the field of neuromodulation continues to evolve,it could potentially bring forth methods for the treatment of demyelinating diseases,and as such,further investigation into the mechanisms of neuron-dependent myelination as well as neuro-imaging modalities that can monitor myelination activity is warranted.Elliot H.Choi Chioma Nwakalor Nolan J.Brown Joonho Lee Michael Y.Oh In Hong Yang 2021Neural Regeneration Research2021,16,2:1
14The descriptive epidemiology of sitting in Chilean adults: Results from the National Health Survey 2009-2010显示文摘Background: Although evidence on the health effects of sedentary behavior(SB) has grown systematically in recent years, few developing countries have reported population levels of SB, especially in South America. Our objective was to describe time spent sitting in a representative sample from Chile categorized by age, gender, educational level, and body mass index(BMI).Methods: A national health survey was conducted in Chile in a nationally representative sample(n = 5411) in 2009-2010. Sitting time(ST) was measured with the Global Physical Activity Questionnaire Version 2.Results: Data were from 5031 participants(43.26±0.41 years, mean±SE; 40.3% male). Overall, there were no gender differences in mean ST(men: 158.10±5.80 min/day, women 143.45±4.77 min/day; p = 0.05). ST was lower in those who lived in rural areas compared with urban areas(99.4 min/day vs. 160.0 min/day; p = 0.001). ST increased significantly with increasing BMI, but only in men(p = 0.009), and was positively related to years of education in both men and women(p < 0.0001).Conclusion: The findings were different from those reported in other countries and contexts, reinforcing the need for international surveillance and monitoring over time to inform policy makers.
