| 2 | Selection and development of representative simple sequence repeat primers and multiplex SSR sets for high throughput automated genotyping in maize显示文摘In the current study, 1900 maize simple sequence repeat (SSR) primers published in MaizeGDB were screened utilizing reference literature, 15 representative Chinese maize inbred lines and 15 Chinese maize hybrids from national regional testing. In total, 500 highly polymorphic primers were identified and used to construct a genetic map. 100 evenly distributed primers, 10 primers per chromosome, were further selected as a set of universal SSR core primers, recommended as preferred primers for general studies. These core primers were then redesigned and used to construct a high throughput multiplex PCR system based on a five-color fluorescence capillary detection system. | 4 | Major and trace element geochemistry of the mid-Bay of Bengal surface sediments: implications for provenance显示文摘The major and trace elements in 110 surface sediment samples collected from the middle of the Bay of Bengal(mid-Bay of Bengal) are analyzed to investigate provenance. Si levels are highest, followed by Al, and the distributions of these two elements are identical. The average CIA*(chemical index of alteration) value is 72.07,indicating that the degree of weathering of the sediments in the study area is intermediate between those of sediments of the Himalayan and Indian rivers. Factor analyses and discrimination function analyses imply that the two main provenances are the Himalayan and the Indian continent. The inverse model calculation of the Tinormalized element ratios of the Bay of Bengal sediments indicate an estimated average contribution of 83.5%and 16.5% from the Himalayan and peninsular Indian rivers to the study area, respectively. The Himalayan source contributes more sediment to the eastern part of the study area, whereas the western part receives more sediment from the Indian Peninsula than did the eastern part. | 6 | Relationship between differ- ential gene expression pat- terns in functional leaves of maize (Zea mays L.) at milk filling stage and heterosis using cDNA-AFLP显示文摘To understand the molecular mechanism of maize heterosis, differential gene expression patterns in the functional leaves of 35 maize hybrids relative to their parents involving 10 elite inbreds at milk filling stage were analyzed by using cDNA-AFLP. The correlation analyses of various differential expression patterns with the performance and heterosis of main maize agronomic traits were evaluated. The main results were as follows: For uniparental specific expression, significant positive correlations were detected with the performance of seed weight per ear and 100-seed weight at 0.01 and 0.05 probability levels respectively. For biparental specific expression, significant negative correla-tions were detected with the performance of ear diameter and seed weight per ear at 0.01 probability level. For uni-parental specific expression, significant positive correlations were detected with the heterosis of ear diameter and seed weight per ear at 0.01 and 0.05 probability levels respectively. For biparental specific expression, significant negative cor-relation was detected with the heterosis of ear diameter at 0.05 probability level. | 7 | Construction of a genetic map and location of quantitative trait loci for dwarf trait in maize by RFLP markers显示文摘Using F2 population derived from the cross of tall inbred 7922 by dwarf inbred 5003, an RFLP linkage map of maize has been constructed, on which 85 markers are distributed among 10 linkage groups and span maize genome about 1827.8 cM with an average distance (24.4 cM) between markers. 106 F2:3 lines of the population were grown in a 10×11 simple rectangular lattice design of one-raw plots with two replications and evaluated for plant height (PH). With interval mapping procedure, 5 QTLs controlling plant height have been identified and their genetic effects and gene action determined. 2 major OTLs with opposite effect have been discovered. One for increasing plant height is ph 1 which is located at chromosome 2 and accounts for 51.8% of the total phenotypic variation; the other for decreasing plant height is ph3 which is located at chromosome 5 and accounts for 38.6% of the total phenotypic variation. | 8 | An expression atlas of miRNAs in Arabidopsis thaliana显示文摘MicroRNAs(miRNAs) are small non-coding RNAs that regulate a variety of biological processes. miRNA expression often exhibits spatial and temporal specificity. However, genome-wide miRNA expression patterns in different organs during development of Arabidopsis thaliana have not yet been systemically investigated. In this study, we sequenced small RNA libraries generated from 27 different organ/tissue types, which cover the entire life cycle of Arabidopsis. Analysis of the sequencing data revealed that most miRNAs are ubiquitously expressed, whereas a small set of miRNAs display highly specific expression patterns. In addition, different miRNA members within the same family have distinct spatial and temporal expression patterns. Moreover, we found that some miRNAs are produced from different arms of their hairpin precursors at different developmental stages. | 13 | Cloning and characterization of a novel R1-MYB transcription factor in maize显示文摘R1-MYB transcription regulatory factors play vital roles in transcriptional modification during higher plant metabolism and development. In the present study, an R1-MYB gene was isolated from the maize (Zea mays L.) inbred line C8605-2 based on expressed sequence tags (ESTs), which expressed differentially between the hybrid C8605-2 × W1445 and its two parents in an oligonucleotide microarray. The full-length cDNA, designated as ZmMYBE1 (GeneBank Accession No. FJ024049), consists of 1992 nucleotides and contains a 1296-bp open reading frame. One conserved MYB domain was detected near the N-terminus of the deduced amino acid sequence, where an acidic Ser/Thr-rich area was also observed in the downstream region. The sequence of ZmMYBE1, mapped to chromosome 5 (bin 5.03), was revealed belonging to a multi-gene family in the maize genome. Subcellular localization analysis determined that ZmMYBE1 was located in the nucleus. Expression analysis showed that ZmMYBE1 transcripts accumulated in various tissues examined, with high expression levels in the immature ear and low levels in the tassel, root, and stem. The expression analysis for ZmMYBE1 in immature ears showed the highest expression level at the earliest vegetative developmental stage of the plant. Expression of ZmMYBE1 higher than that of the parents was observed in the hybrid C8605-2 × W1445. And also the expression level similar to the lowest of the parents in the hybrid C8605-2 × W245 was observed, revealing different expression patterns for the same gene in various hybrids combined from diversified inbred lines. | 15 | Detection of quantitative trait loci and heterotic loci for plant height using an immortalized F_2 population in maize显示文摘A set of recombinant inbred lines (RIL) derived from Yuyu22, an elite hybrid widespread in China, was used to construct an immortalized F2 (IF2) population comprising 441 different crosses. Genetic linkage maps were constructed containing 10 linkages groups with 263 simple sequence repeat (SSR) molecular markers. Twelve and ten quantitative trait loci (QTL) were detected for plant height in the IF2 and RIL populations respectively, using the composite interval mapping method, and six same QTL were identified in the two populations. In addition, ten unique heterotic loci (HL) located on seven different chromosomes were revealed for plant height using the mid-parent heterosis as the input data. These HL explained 1.26%―8.41% of the genotypic variance in plant height heterosis and most expressed overdominant effects. | 18 | The chromosome-based lavender genome provides new in sights into Lamiaceae evolution and terpenoid biosynthesis显示文摘The aromatic shrub Lavandula angustifolia produces various volatile terpenoids that serve as resources for essential oils and function in plant-insect communication.To better understand the genetic basis of the terpenoid diversity in lavender,we present a high-quality reference genome for the Chinese lavender cultivar‘Jingxun 2'using PacBio and Hi-C technologies to anchor the 894.50 Mb genome assembly into 27 pseudochromosomes.In addition to the y triplication event,lavender underwent two rounds of whole-genome duplication(WGD)during the Eocene-Oligocene(29.6 MYA)and Miocene-Pliocene(6.9 MYA)transitions.As a result of tandem duplications and lineage-specific WGDs,gene families related to terpenoid biosynthesis in lavender are substantially expanded compared to those of five other species in Lamiaceae.Many terpenoid biosynthesis transcripts are abundant in glandular trichomes.We further integrated the contents of ecologically functional terpenoids and coexpressed terpenoid biosynthetic genes to construct terpenoid-gene networks.Typical gene clusters,including TPS-TPS,TPS-CYP450,and TPS-BAHD,linked with compounds that primarily function as attractants or repellents,were identified by their similar patterns of change during flower development or in response to methyl jasmonate.Comprehensive analysis of the genetic basis of the production of volatiles in lavender could serve as a foundation for future research into lavender evolution,phytochemistry,and ecology. | Jingrui Li Yiming Wang Yanmei Dong Wenying Zhang Di Wang Hongtong Bai Kui Li Hui Li Lei Shi | 2021 | Horticulture Research2021,8,1: | 5 |
| 19 | Heparanase mRNA expression and point mutation in hepatocellular carcinoma显示文摘AIM: To explore the expression of heparanase mRNA and point mutation in hepatocellular carcinoma (HCC). METHODS: Reverse transcription polymerase chain reaction was used to measure the expression of heparanase mRNA in the primary tumor tissues and surrounding liver tissues of 33 HCC patients. T-A cloning and sequencing were used to detect whether there was any mutation in the amplified PCR products. RESULTS: The expression of heparanase mRNA waspositive in 16 primary tumor tissues of HCC, and the positive rate was 48.5%, which was significantly higher than that in the surrounding liver parenchyma (P<0.01). The positive rate for heparanase gene in high-tendency to metastatic recurrence group (71.4%, 10/14) was obviously higher than that in low-tendency to metastatic recurrence group (31.6%, 6/19) (P= 0.023). The positive rate for heparanase gene in paltients with metastatic recurrence during postoperative follow-up (78.6%, 11/14) was also significantly higher than that in those without metastatic recurrence (21.4%, 3/14) (P = 0.003). Sequence analysis of the HPA PCR products was made in 7 patients, and 2-point mutations were found in 4 patients, one of which was sense mutation, neither base insertion nor deletion was detected. The mutation rate was 57.1% (4/7). CONCLUSION- The expression rate of heparanase mRNA increases in HCC, and HPA mRNA may be one of the reliable markers for the metastatic activity gained by the liver tumor cells and could be used clinically in predicting metastatic recurrence of HCC. | 20 | Bamboo charcoal enhances cellulase and urease activities during chicken manure composting:Roles of the bacterial community and metabolic functions显示文摘Microbial enzymes are crucial for material biotransformation during the composting process.In this study,we investigated the effects of adding bamboo charcoal(BC)(i.e.,at 5%,10%,and 20%corresponding to BC5,BC10,and BC20,respectively)on the enzyme activity levels during chicken manure composting.The results showed that BC10 could increase the cellulose and urease activities by 56%and 96%,respectively.The bacterial community structure in BC10 differed from those in the other treatments,and Luteivirga,Lactobacillus,Paenalcaligenes,Ulvibacter,Bacillus,Facklamia,Pelagibacterium,Sporosarcina,Cellvibrio,and Corynebacterium had the most important roles in composting.Compared with other treatments,BC10 significantly enhanced the average rates of degradation of carbohydrates(Dxylose(40%)andα-D-lactose(44%))and amino acids(L-arginine(16%),L-asparagine(14%),and L-threonine(52%)).We also explored the associations among the bacterial community and their metabolic functions with the changes in the activities of enzymes.Network analysis demonstrated that BC10 altered the co-occurrence patterns of the bacterial communities,where Ulvibacter and class Bacilli were the keystone bacterial taxa with high capacities for degrading carbon source,and they were related to increases in the activities of cellulase and urease,respectively.The results obtained in this study may help to further enhance the efficiency of composting. | Yanan Yin Chao Yang Jingrui Tang Jie Gu Haichao Li Manli Duan Xiaochang Wang Rong Chen | 2021 | Journal of Environmental Sciences2021,33,10: | 4 |
