1Endocrine impact of Helicobacter pylori:Focus on ghrelin and ghrelin o-acyltransferase显示文摘Ghrelin is predominantly produced by the gastric enteroendocrine cell compartment and is octanoylated by the recently discovered ghrelin o-acyltransferase (GOAT) before secretion into the bloodstream. This octanoylation is essential for many of the biological properties of ghrelin including appetite stimulation and anti-inflammatory properties as only the acylated form of ghrelin binds to the ghrelin receptor, the growth hormone secretagogue receptor (GHS-R). Given the gastric location of ghrelin production, it is perhaps not surprising that insult to the gastric mucosa affects circulating ghrelin levels in humans. Helicobacter pylori (H. pylori) infects more than f ifty percent of the world's population and once established within the gastric mucosa, can persist for life. Infection is associated with chronic gastritis, gastric atrophy and ulceration, reduced appetite and a lower body mass index (BMI). The large majority of studies investigating levels of circulating ghrelin and ghrelin expression in the stomach in patients with H. pylori infection indicate that the bacterium has a negative impact on ghrelin production and/or secretion. Eradication of infection restores ghrelin, improves appetite and increases BMI in some studies, however, a causative relationship between H. pylori-associated serum ghrelin decline and food intake and obesity has not been established. Most studies measure total ghrelin in the circulation although the measurement of the ratio of acyl/total ghrelin gives a clearer indication that the ghrelin acylation process is altered during infection and atrophy. 2Axonal regeneration after spinal cord injury in zebrafish and mammals: differences, similarities, translation显示文摘Spinal cord injury (SCI) in mammals results in functional deficits that are mostly permanent due in part to the inability of severed axons to regenerate. Several types of growth-inhibitory molecules expressed at the injury site contribute to this regeneration failure. The responses of axons to these inhibitors vary greatly within and between organisms, reflecting axons' characteristic intrinsic propensity for regeneration. In the zebrafish (Danio rerio) many but not all axons exhibit successful regeneration after SCi. This review presents and compares the intrinsic and extrinsic determinants of axonal regeneration in the injured spinal cord in mammals and zebrafish. 3局部缓释IUdR治疗恶性星形细胞瘤实验研究(英文)显示文摘目的研究脑局部缓释125碘标记的5-碘脱氧尿苷(IUdR)的作用。方法为了控制释放125碘-IUdR,合成了25μCi的羧基苯氧丙烷:癸二酸(PCPPSA)(2080)125碘-IUdR聚合体。体外将合成的三聚体缓释剂(10mgPCPPSA)置于37℃磷酸盐缓冲溶液(PBS)中孵育不同时间后移出溶剂并置换为新的PBS,将移出液于放射计数器上测量数值。在体研究采用雄性裸鼠(6周)颅内接种U251人恶性星形细胞瘤及皮下接种对比研究。将颅内荷瘤鼠瘤腔内及皮下接种肿瘤组织内分别应用三聚体缓释剂及空白缓释剂,于不同时间分别测量动物头颅及皮下结节的放射活性,间接定量研究缓释状况;采血了解全身代谢情况。对于多聚磷酸酯(PPE)缓释剂研究采用皮下肿瘤结节内局部应用并测量其缓释的方法,放射自显影研究及全身不同器官缓释剂的吸收代谢研究采用接种后2、4和8d收集标本,分别行切片放射自显影和器官放射活性测定。结果体内和体外研究均显示PCPP:SA缓释剂对125碘-IUdR的控制性释放作用,尤其在体研究分别显示颅内和皮下接种的肿瘤组织对标记的IUdR的释放程度存有极大的差异,提示颅内局部应用的优越性。放射自显影定性研究显示了随距离的增加被标记的IUdR的递减趋势,为确定缓释剂施放部位和间距提供了参考数据。各主要脏器放射性测定的结果显示早期小肠内放射性较高,其次为脾脏,提示IUdR主要的代谢渠道,为处理应用其而可能引发的其他脏器损害或作用提供了线索。结论放射标记的IUdR缓释剂主要集中在接种部位且缓释作用确切。马晓东 Jerry R.Williams Larry E.Dillehay Jeffery A Williams 2005中华神经外科疾病研究杂志2005,4,4:3
4经颈内静脉肝内门腔分流术远期疗效分析显示文摘目的:探讨经颈静脉肝内门脉分流术(TIPS)后病人的远期分流道通畅率,生存期和生活质量。材料及方法:51例TIPS术后病人,随访时间平均为16.8±12.8月(1天-49月),用Kaplan-Meier法分析TIPS后病人的远期生存率,和分流道通畅率。用COX模型将生存期和通畅率与Child~Pugh肝功能分级和主要症状作相关分析,并预测影响生存期和分流道通畅率的因素。用SF~36问卷评估病人的生活质量。结果:1-4年累计生存率分别为65%,56%,32%,19%。累计原发通畅率分别为65%,56%,32%,19%。42个月原发再次通畅率为94%,18个月4继发通畅率为7l%。肝功能Child-Pugh分级与累积生存率有显著性差异。影响TIPS术后90天生存率的相关因素有:酒精性肝硬化,腹水,急性出血,分流腔道直径,静脉曲张需栓塞者,肝性脑病,分流通道再狭窄及再发出血。TIPS术后生活质量的所有9项指标均较术前提高,其中4项有显著性差异。结论:TIPS对控制出血,腹水及改善近期生活质量有肯定的疗效,但是对病人远期生活率的确切作用尚有待于进一步的研究。滕皋军 Michael A Bettmann Robert F Jeffery James K St.George 1997介入放射学杂志1997,6,3:3
5Dysfunctional stem and progenitor cells impair fracture healing with age显示文摘Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. 6Discretization Approach in Integrated Hydrologic Model for Surface and Groundwater Interaction显示文摘The commonly used discretization approaches for distributed hydrological models can be broadly categorized into four types,based on the nature of the discrete components:Regular Mesh,Triangular Irregular Networks(TINs),Representative Elementary Watershed(REWs) and Hydrologic Response Units(HRUs).In this paper,a new discretization approach for landforms that have similar hydrologic properties is developed and discussed here for the Integrated Hydrologic Model(IHM),a combining simulation of surface and groundwater processes,accounting for the interaction between the systems.The approach used in the IHM is to disaggregate basin parameters into discrete landforms that have similar hydrologic properties.These landforms may be impervious areas,related areas,areas with high or low clay or organic fractions,areas with significantly different depths-to-water-table,and areas with different types of land cover or different land uses.Incorporating discrete landforms within basins allows significant distributed parameter analysis,but requires an efficient computational structure.The IHM integration represents a new approach interpreting fluxes across the model interface and storages near the interface for transfer to the appropriate model component,accounting for the disparate discretization while rigidly maintaining mass conservation.The discretization approaches employed in IHM will provide some ideas and insights which are helpful to those researchers who have been working on the integrated models for surface-groundwater interaction.ZHANG Jing Mark A ROSS Jeffery GEURINK 2012Chinese Geographical Science2012,22,6:3
