18The metabolite, alpha-ketoglutarate inhibits non-alcoholic fatty liver disease progression by targeting lipid metabolism显示文摘Background:Non-alcoholic liver disease is of increased concern and contributing to economic burdens not only in developing countries but in developed countries as well.Identifying the biomarker of early diagnosis and early intervention approaches for non-alcoholic liver disease is unmet and required further investigation.Although the alpha-ketoglutarate(a-KG)is recently proposed to be a potential biomarker in differentiating patients with obesity from those with non-alcoholic liver disease,how a-ketoglutatate is involved in the fatty liver progression is not clear.Methods:A high-fat diet(HFD)feeding animal model,liver functional assays,and molecular approaches were adopted to clarify the impact of a-KG in fatty liver progression.Results:In the current study,it was found that dietary a-KG would inhibit weight gain in male and female mice fed with a normal chew or HFD.HFD feeding caused fatty liver in male mice,but a-KG treatment could substantially inhibit hepatic steatosis progression.Biochemical studies revealed the possible linkage of a-KG protective functions to lipid metabolism.Further analysis identified the important role of peroxisome proliferator-activated receptors in beneficial a-KG-mediated effects on fatty liver progression.Conclusions:The current study demonstrates the therapeutic potential of a-KG and how it may be used,via dietary supplementation,as a preventive intervention for non-alcoholic liver disease in obese patients.Katsuya Nagaoka Joud Mulla Kevin Cao Zhixiang Cheng Dan Liu William Mueller Amalia Bay Grace Hildebrand Shaolei Lu Chiung-Kuei Huang 2020Liver Research2020,4,2:0
19Human mesenchymal stem cells exhibit altered mitochondrial dynamics and poor survival in high glucose microenvironment显示文摘BACKGROUND Mesenchymal stem cells(MSCs)are a type of stem cells that possess relevant regenerative abilities and can be used to treat many chronic diseases.Diabetes mellitus(DM)is a frequently diagnosed chronic disease characterized by hyperglycemia which initiates many multisystem complications in the long-run.DM patients can benefit from MSCs transplantation to curb down the pathological consequences associated with hyperglycemia persistence and restore the function of damaged tissues.MSCs therapeutic outcomes are found to last for short period of time and ultimately these regenerative cells are eradicated and died in DM disease model.AIM To investigate the impact of high glucose or hyperglycemia on the cellular and molecular characteristics of MSCs.METHODS Human adipose tissue-derived MSCs(hAD-MSCs)were seeded in low(5.6 mmol/L of glucose)and high glucose(25 mmol/L of glucose)for 7 d.Cytotoxicity,viability,mitochondrial dynamics,and apoptosis were deplored using specific kits.Western blotting was performed to measure the protein expression of phosphatidylinositol 3-kinase(PI3K),TSC1,and mammalian target of rapamycin(mTOR)in these cells.RESULTS hAD-MSCs cultured in high glucose for 7 d demonstrated marked decrease in their viability,as shown by a significant increase in lactate dehydrogenase(P<0.01)and a significant decrease in Trypan blue(P<0.05)in these cells compared to low glucose control.Mitochondrial membrane potential,indicated by tetramethylrhodamine ethyl ester(TMRE)fluorescence intensity,and nicotinamide adenine dinucleotide(NAD+)/NADH ratio were significantly dropped(P<0.05 for TMRE and P<0.01 for NAD+/NADH)in high glucose exposed hAD-MSCs,indicating disturbed mitochondrial function.PI3K protein expression significantly decreased in high glucose culture MSCs(P<0.05 compared to low glucose)and it was coupled with significant upregulation in TSC1(P<0.05)and downregulation in mTOR protein expression(P<0.05).Mitochondrial complexes I,IV,and V were downregulated profoundly in high glucose(P<0.05 compared to low glucose).Apoptosis was induced as a result of mitochondrial impairment and explained the poor survival of MSCs in high glucose.CONCLUSION High glucose impaired the mitochondrial dynamics and regulatory proteins in hAD-MSCs ensuing their poor survival and high apoptosis rate in hyperglycemic microenvironment.Ejlal Abu-El-Rub Fatimah Almahasneh Ramada R Khasawneh Ayman Alzu'bi Doaa Ghorab Rawan Almazari Huthaifa Magableh Ahmad Sanajleh Haitham Shlool Mohammad Mazari Noor S Bader Joud Al-Momani 2023World Journal of Stem Cells2023,15,12:0
20Continuous Mobile User Authentication Using a Hybrid CNN-Bi-LSTM Approach显示文摘Internet of Things (IoT) devices incorporate a large amount ofdata in several fields, including those of medicine, business, and engineering.User authentication is paramount in the IoT era to assure connecteddevices’ security. However, traditional authentication methods and conventionalbiometrics-based authentication approaches such as face recognition,fingerprints, and password are vulnerable to various attacks, including smudgeattacks, heat attacks, and shoulder surfing attacks. Behavioral biometrics isintroduced by the powerful sensing capabilities of IoT devices such as smartwearables and smartphones, enabling continuous authentication. ArtificialIntelligence (AI)-based approaches introduce a bright future in refining largeamounts of homogeneous biometric data to provide innovative user authenticationsolutions. This paper presents a new continuous passive authenticationapproach capable of learning the signatures of IoT users utilizing smartphonesensors such as a gyroscope, magnetometer, and accelerometer to recognizeusers by their physical activities. This approach integrates the convolutionalneural network (CNN) and recurrent neural network (RNN) models to learnsignatures of human activities from different users. A series of experiments areconducted using the MotionSense dataset to validate the effectiveness of theproposed method. Our technique offers a competitive verification accuracyequal to 98.4%.We compared the proposed method with several conventionalmachine learning and CNN models and found that our proposed modelachieves higher identification accuracy than the recently developed verificationsystems.
