3Construction of Prokaryotic Expression Vector of Mouse Nanog Gene and Its Expression显示文摘The aim of this study is to construct a prokaryotic expression vector of mouse Nanog gene and to express it in E.coli.A pair of primers was designed according to digestion sites in plasmid pGEX-KG and the Nanog gene sequence published by GenBank.The DNA fragment of 918 bp was amplified by polymerase chain reaction(PCR)from the pNA992 recombinant plasmid with Nanog gene,then cloned into pGEX-KG and transformed into the host E.coli strain TGⅠ.The sequence of the fragment was matched with the original sequence of pNA992.It indicated that fusion expression vector,pGEX-KG- Nanog,was constructed successfully.The pGEX-KG-Nanog plasmid was extracted from E.coli strain TGⅠand was transformed into BL21(DE3)for expression.After induction by isopropy1-β-D-thiogalactoside(IPTG)at 37°C,the expression product of Nanog gene was identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),and the expression condition was optimized.Nanog fusion protein was successfully expressed in the form of inclusion bodies. The molecular weight of the inclusion body was 63 kDa.Meanwhile,the optimum condition for the expression of Nanog fusion protein was induced with 0.8 mmol L-1 IPTG for 5 h.The mouse Nanog gene was successfully expressed in E.coli, which laid a foundation for the purification of Nanog protein and for the preparation of polyclonal antibody.LI Jun Lü Chang-rong DOU Lin DOU Zhong-ying 2007Agricultural Sciences in China2007,6,4:3
4Decadal change of the East Asian summer monsoon and its related surface temperature in Asia-Pacific during 1880–2004显示文摘The East Asian summer monsoon(EASM)and its related change of surface temperature in the past century were not clearly addressed due to absence of atmospheric reanalysis data before 1948.On the benchmark of station-observed sea level pressure(SLP)in China,we utilized multiple SLP datasets and evaluated their qualities in measuring the SLP-based EASM index(EASMI).It is found that the EASMI based on the SLP of the Hadley center version 2(HadSLP2)has shown the best performance on the interannual and decadal time scales.Instead of showing a linear weakening trend pointed out by the previous study,the EASMI has likely exhibited the decadal variability,characterized by weakened trends during 1880–1906,1921–1936,and 1960–2004,and with enhanced trends during 1906–1921 and 1936–1960,respectively.Corresponding to the weakened and enhanced periods of EASMI since the 1920s,the surface air temperature(SAT)index(SATI)averaged in eastern China has likely shown a warming and a cooling trend,respectively.However,the decadal abrupt transitions between the two indices do not occur concurrently,which results in a weak correlation between two indices on the decadal time scale.Further analysis indicates that there are four key regions where the SAT is significantly correlated with the EASMI,suggesting the joint impact of surface temperature in Asia-Pacific on the EASM during 1880–2004.In which,the decadal change of SAT near the Lake Baikal plays an important role in the linear trends of the EASM before and after 1960.LIN Xiang ZHU CongWen Lü JunMei 2013Chinese Science Bulletin2013,58,35:2

11Simulation of the emission lines radiated from cataclysmic variables显示文摘Cataclysmic variables are special celestial bodies because they have particular light curves and spectra.The mechanisms for generating emission lines radiated from dwarf novae in their quiescent phases are studied.We assume that the incident radiation field which is emitted by a hot source(white dwarf and boundary layer)irradiates the gaseous layer evaporated from the accretion disk,and the emission lines are radiated from the gas.We model the fluxes of emission lines by using the photoionization code CLOUDY.Using this method,we input some reasonable parameters and get a series of simulated spectra.In order to find a simulated spectrum which is the best fit to an observed spectrum,we use a cross-correlation method to match them.After the calculation,we use the approximation that the parameters of the simulated spectrum can simulate the observed spectrum.Finally,we learn more about the physical conditions of the system.Lin He Guo-Liang Lü Yong-Heng Zhao 2013Research in Astronomy and Astrophysics2013,13,10:0
12Steiner Tree Based Optimal Resource Caching Scheme in Fog Computing显示文摘Fog Computing is a new platform that can serve mobile devices in the local area. In Fog Computing, the resources need to be shared or cached in the widely deployed Fog clusters. In this paper, we propose a Steiner tree based caching scheme, in which the Fog servers, when caching resources, first produce a Steiner tree to minimize the total path weight(or cost) such that the cost of resource caching using this tree could be minimized. Then we give a running illustration to show how the Fog Computing works and we compare the traditional shortest path scheme with the proposed one. 13Research progress on microstructure evolution of semi-solid aluminum alloys in ultrasonic field and their rheocasting显示文摘The effects of ultrasonic vibration(UV)treatment on microstructure of semi-solid aluminum alloys and the application of UV in rheocasting process are reviewed.Good semi-solid slurry can be produced by high-intensity UV process for aluminum alloys.The microstructures of Al-Si,Al-Mg and Al-Cu alloys produced by rheocasting assisted with UV are compact and with fine grains.The mechanical properties of the UV treated alloys are increased by about 20%-30%.Grain refinement of the alloys is generally considered because of cavitation and acoustic streaming caused by UV.Apart from these mechanisms,a hypothesis of the fuse of dendrite root caused by capillary infiltration in the ultrasonic field,as well as a mechanism of crystallites falling off from the mould-wall and crystal multiplication by mechanical vibration effect in indirect ultrasonic vibration are proposed to explain the microstructure evolution of the alloys.Wu Shusen Lin Chong Lü Shulin Sha Meng 2014China Foundry2014,11,4:7
14Comparison of continuous homogenous azeotropic and pressure-swing distillation for a minimum azeotropic system ethyl acetate/nhexane separation显示文摘Continuous homogenous azeotropic distillation(CHAD) and pressure-swing distillation(PSD) are explored to separate a minimum-boiling azeotropic system of ethyl acetate and n-hexane. The CHAD process with acetone as the entrainer and the PSD process with the pressures of 0.1 MPa and 0.6 MPa in two columns are designed and simulated by Aspen Plus. The operating conditions of the two processes are optimized via a sequential modular approach to obtain the minimum total annual cost(TAC). The computational results show that the partially heat integrated pressure-swing distillation(HIPSD) has reduced in the energy cost and TAC by 40.79% and 35.94%, respectively, than the conventional PSD, and has more greatly reduced the energy cost and TAC by 62.61% and 49.26% respectively compared with the CHAD process. 15Spacelike Mobius Hypersurfaces in Four Dimensional Lorentzian Space Form显示文摘In this paper, we first set up an alternative fundamental theory of M?bius geometry for any umbilic-free spacelike hypersurfaces in four dimensional Lorentzian space form, and prove the hypersurfaces can be determined completely by a system consisting of a function W and a tangent frame {E_i}. Then we give a complete classification for spacelike M?bius homogeneous hypersurfaces in four dimensional Lorentzian space form. They are either M?bius equivalent to spacelike Dupin hypersurfaces or to some cylinders constructed from logarithmic curves and hyperbolic logarithmic spirals. 16Effect of Tongxieyaofang decoction on colonic mucosal protein expression profiles in rats with visceral hypersensitivity显示文摘OBJECTIVE: To explore the underlying mechanism of action of Tongxieyaofang decoction in rats with visceral hypersensitivity using proteomics technology.METHODS: Twenty-four female Sprague-Dawley rats were randomly divided into three groups: control group, irritable bowel syndrome(IBS) group and Tongxieyaofang treatment group. An IBS model, characterized as visceral hypersensitivity, was established using the odour of mothballs as conditional stimulation and colorectal distension combined with classic physical restraint as non-conditional stimulation. Rats were intragastrically treated with Tongxieyaofang(2 or 4 m L · kg^(-1)· d^(-1))for 4 weeks. On the 45 th day, the rats were dissected and the colonic mucosal proteins were extracted. Differential protein spots were screened by fluorescent two-dimensional differential gel electrophoresis(2 D-DIGE), and identified by matrix-assisted laser desorption/ionisation time of flight mass spectrometry(MALDI-TOF-MS). Western blotting experiments were performed to verify the changes observed in 2 D-DIGE and MALDI-TOF-MS.RESULTS: It was found that the visceral sensitivity of rats in the Tongxieyaofang treatment group(4 m L/kg) was lower than that in the IBS group(P <0.01). Sixty-one protein spots were differentially expressed between the IBS group and the Tongxieyaofang treatment group. Of these, 23 spots were upregulated in the Tongxieyaofang treatment group, while 38 spots were downregulated. Three specific proteins were successfully identified from the five protein spots with the most obvious changes. The two upregulated proteins were transgelin(TAGLN) and acetaldehyde dehydrogenase 2(Aldh2) and the downregulated protein was cytokeratin 8(CK8).CONCLUSION: Tongxieyaofang can dose-dependently ameliorate visceral hypersensitivity in rats and the mechanism of action may involve the upregulation of TAGLN and Aldh2 and the downregulation of CK8.Lin Yongjun Ding Ying Lü Bin Liu Nan 2020Journal of Traditional Chinese Medicine2020,40,2:4
17Effects of high-pressure rheo-squeeze casting on the Fe-rich phases and mechanical properties of Al-17Si-(1,1.5)Fe alloys显示文摘The effects of high pressure rheo-squeeze casting(HPRC) on the Fe-rich phases(FRPs) and mechanical properties of Al-17 Si-(1,1.5)Fe alloys were investigated. The alloy melts were first treated by ultrasonic vibration(UV) and then formed by high-pressure squeeze casting(HPSC). The FRPs in the as-cast HPSC Al-17 Si-1 Fe alloys only contained a long, needle-shaped β-Al5 Fe Si phase at 0 MPa. In addition to the β-Al5 Fe Si phase, the HPSC Al-17 Si-1.5 Fe alloy also contained the plate-shaped δ-Al4 Fe Si2 phase. A fine, block-shaped δ-Al4 Fe Si2 phase was formed in the Al-17 Si-1 Fe alloy treated by UV. The size of FRPs decreased with increasing pressure. After UV treatment, solidification under pressure led to further refinement of the FRPs. Considering alloy samples of the same composition, the ultimate tensile strength(UTS) of the HPRC samples was higher than that of the HPSC samples, and the UTS increased with increasing pressure. The UTS of the Al-17 Si-1 Fe alloy formed by HPSC exceeded that of the Al-17 Si-1.5 Fe alloy formed in the same manner under the same pressure. 18DMMF: Dominant and Max-Min Fair Allocation in Satellite Networks显示文摘Resource allocation is crucial for satellite networks. In this paper, we propose a multi-resource fair allocation scheme, namely Dominant and Max-min Fair(DMMF), to efficiently and fairly allocate resources. It consists of two allocation stages, dominant resource fair(DRF) allocation stage and max-min fairness(MMF) allocation stage. The proposed DMMF scheme exhibits desirable properties, including share incentive, strategy proofness, envy freeness and Pareto optimality.
