1Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection,histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection,tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection,and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys).In 30 of them,both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern,tumor staging,lymph node metastasis,grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma,andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. 2Analysis of carbon isotope in phytoliths from C3 and C4 plants and modern soils显示文摘The analysis of carbon isotope in phytoliths from modern plants and surface soils in China shows that the values of carbon isotope are consistent with those from C3 and C4 plants, and the processes of photosynthesis of the original plants can be clearly identified by carbon isotope in phytoliths. The value of carbon isotope varied from -23.8‰ to -28‰, with the maximum distributed in the latitude zone from 34°N to 40°N in North China and East China areas, and the minimum in the Northeast China and South China regions. The values of carbon of phytoliths tend to increase from low to high and then reduce to low value again as the latitude increases. 3Hydrodynamic process of Tibetan Plateau lake revealed by grain size:Case study of Pumayum Co显示文摘Under the background of global warming,some lakes on the Tibetan Plateau(TP) are potentially sensitive to temperature change.With a case study of Pumayum Co,where glacier meltwater is important to supply(we call this a glacier-fed lake hereafter),we analyze the sensitivity of lake sediment grain size to temperature change.This is done by resolving the modern hydrodynamic process,coupled with comparison of paleoclimatic proxies.According to the spatial distributions of parameters,percentage of grain size and the grain size frequency distribution curve,hydrodynamic processes are analyzed.Five clastic sedimentation types are thereby discriminated.In the open lake area,suspended load transport is the main transport agent.Grain Size Trend Analysis(GSTA),a sediment dynamics model,reveals a trend toward eastward transport.This indicates that the largest and glacier-fed river,the Jiaqu River,influences the entire lake(not just the subsurface alluvial fan),and that lake sediment grain size may serve as a temperature indicator.Time series comparison between grain size of a short core from the central lake and meteorological data confirms this temperature indication,which in turn shows reliability of the method discriminating the hydrodynamic process.This case study will improve the ability of paleoclimatic reconstruction using lake sediment in glacier-fed lakes on the Tibetan Plateau.JU JianTing ZHU LiPing FENG JinLiang WANG JunBo WANG Yong XIE ManPing PENG Ping ZHEN XiaoLin LÜXinMiao 2012Chinese Science Bulletin2012,57,19:7
4Development and characterization of an inducible Dicer conditional knockout mouse model of Parkinson’s disease:validation of the antiparkinsonian effects of a sigma-1 receptor agonist and dihydromyricetin显示文摘Parkinson’s disease(PD)is a common neurodegenerative disease characterized by motor impairment and progressive loss of dopamine(DA)neurons.At present,the acute application of neurotoxic drugs such as 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)and 6-hydroxydopamine(6-OHDA)are commonly used to simulate the pathology of PD;however,it is dif?cult to induce the progressive pathogenesis of PD with these models.In this study,we employed DAT promoter-mediated Cre transgenic mice to establish tamoxifen-inducible Dicer conditional knockout(cKO)mice in an effort to mimic the progressive loss of DA neurons and the development of PD-like behavioral phenotypes.The results showed that Dicer cKO mice exhibited progressive loss of DA neurons in the substantia nigra(SN)following tamoxifen administration.Signi?cant DA loss was observed 6 weeks after tamoxifen administration;accordingly,progressive motor function impairment was also observed.We also found that a signi?cant neuroin?ammatory response,as evidenced by microglial proliferation,another hallmark of PD pathogenesis,accompanied the loss of DA neurons.The acute application of levo-DOPA(L-DOPA)relieved the PD-like motor impairments in Dicer cKO mice to exert its antiparkinsonian action,indicating that the model can be used to evaluate the antiparkinsonian ef?cacy of PD drugs.To further elucidate the potential application of this novel PD animal model for PD drug development,we employed the powerful neuroprotective agent dihydromyricetin(DHM)(10 mg/kg)and the selective sigma-1 receptor agonist PRE-084(1 mg/kg),both of which were previously shown to produce antiparkinsonian effects.The results indicated that the chronic administration of either DHM or PRE-084 attenuated the Dicer cKO-induced loss of DA neurons and motor impairments,although the two drugs acted through different mechanisms.These data indicate that the Dicer cKO mouse model may be a useful model for investigating the pathological development of PD and intervention-mediated changes.In conclusion,this transgenic mouse model appears to simulate the progressive pathogenesis of PD and may be a potentially useful model for PD drug discovery.Chen-hong Guo Ting Cao Long-tai Zheng John L Waddington Xue-chu Zhen 2020Acta Pharmacologica Sinica2020,41,4:5
5Dual-layered metasurfaces for asymmetric focusing显示文摘Asymmetric transmission,defined as the difference between the forward and backward transmission,enables a plethora of applications for on-chip integration and telecommunications.However,the traditional method for asymmetric transmission is to control the propagation direction of the waves,hindering further applications.Metasurfaces,a kind of two-dimensional metamaterials,have shown an unprecedented ability to manipulate the propagation direction,phase,and polarization of electromagnetic waves.Here we propose and experimentally demonstrate a metasurface-based directional device consisting of a geometric metasurface with spatially rotated microrods and metallic gratings,which can simultaneously control the phase,polarization,and propagation direction of waves,resulting in asymmetric focusing in the terahertz region.These dual-layered metasurfaces for asymmetric focusing can work in a wide bandwidth ranging from 0.6 to 1.1 THz.The flexible and robust approach for designing broadband asymmetric focusing may open a new avenue for compact devices with potential applications in encryption,information processing,and communication.BINGSHUANG YAO XIAOFEI ZANG ZHEN L LIN CHEN JINGYA XIE YIMING ZHU SONGLIN ZHUANG 2020Photonics Research2020,8,6:2
