1Verification of Mutagen Function of Zeocin in Nannochloropsis oceanica Through Transcriptome Analysis显示文摘Zeocin can cause double strand breaks of DNA and thus is frequently used as a selective antibiotic of eukaryotic Sh ble transformants. In non-transformation system, Zeocin may function as a mutagen if not totally lethal. To verify such function of Zeocin, we mutated Nannochloropsis oceanica by increasing the concentration of Zeocin in medium gradually, and isolated a N. oceanica strain(single cell culture) which survived Zeocin up to 10.0μg mL^(-1). The Zeocin-tolerant strain entered the exponential growth phase later and grew slower than the wild strain. Transcriptome profiling showed that the Zeocin-tolerant N. oceanica strain survived Zeocin mainly by adapting(heritable), rather than acclimating(plastic) to Zeocin. Hence mutating N. oceanica with Zeocin was approved effective. Meanwhile, the physiological characteristics of this Zeocin-tolerant strain were demonstrated. As we proposed, N. oceanica tolerated Zeocin by strengthening its protein degradation and antioxidation. The genes controlling cell division and cellular response to stimuli may also have played important roles in the reduction of growth and the tolerance to Zeocin. Our findings evidenced that Zeocin can serve as an appropriate mutagen of microalgae. 2Influence of macroscopic graphite particulates on the damping properties of Zn-Al eutectoid alloy显示文摘The paper presents in detail the effects of macroscopic graphite (Gr) particulates on the damping behavior of Zn-Al eutectoid alloy (Zn-Al). Macroscopic defects are graphite particulates with sizes of the order of a millimeter (0.5 mm and 1.0 mm). Macroscopic graphite particulate-reinforced Zn-Al eutectoid alloys were prepared by the air pressure infiltration process. The damping characterization was conducted on a multifunction internal friction apparatus (MFIFA). The internal friction (IF), as well as the relative dynamic modulus, was measured at different frequencies over the temperature range of 20 to 400℃. The damping capacity of the Zn-Al/Gr, with two different volume fractions of macroscopic graphite particulates, was compared with that of bulk Zn-Al eutectoid alloy. The damping capacity of the materials is shown to increase with increasing volume fraction of macroscopic graphite particulates. Two IF peaks were found in the IF-temperature curves. The first is a grain boundary peak, which is associated with the diffusive flux on a boundary between like phases, Al/Al. Its activation energy has been calculated to be 1.13±0.03 eV and the pre-exponential factor is 10?14 s in IF measurements. The second is a phase transition peak, which results from the transformation of Zn-Al eutectoid. 3Structural Variation Analysis of Mutated Nannochloropsis oceanica Caused by Zeocin Through Genome Re-Sequencing显示文摘Zeocin can cause double strand breaks of DNA and thus may be employed as a mutagen.In this study,two strains of Nannochloropsis oceanica,the wild and the Zeocin-tolerant strains,were re-sequenced to verify such function of Zeocin.The results showed that Zeocin can mutate the N.oceanica genome and cause the structural variation.Zeocin either swept away or selected the alleles of genes functioning in ubiquitin-mediated proteolysis,alpha-linolenic acid metabolism,ascorbate and aldarate metabolism,ribosome biogenesis,and circadian rhythm,indicating that N.oceanica may have adjusted its metabolic performances for protein,carbohydrate,and lipid,and changed its ribosome biosynthesis and living rhythm to survive in Zeocin containing medium.In addition,Zeocin caused mutation may have influenced the expression of a set of transcription factors.It was concluded that Zeocin effectively caused the structural variation of the genome of N.oceanica,and forced the microalgae to select out the alleles of a set of genes around these variations in order to adapt to Zeocin containing medium.Further studies on the genetic basis of the phenotypic adaptation of this haploid and asexual microalga and the application of Zeocin to its genetic improvement are very important.LIN Genmei ZHANG Zhongyi GUO Li DING Haiyan YANG Guanpin 2018Journal of Ocean University of China2018,17,5:3
4Using Chain of Mobile Access Gateway to Reduce Delay for PMIPv6 Protocol Applied in WLAN显示文摘Wireless LAN controller(WLC) is used to manage and control Access points(APs) in Wireless local area network(WLAN).Proxy mobile IPv6(PMIPv6)protocol supports network-layer mobility in WLC based WLAN.However,it introduces extra delay in delivering packets from the APs to the WLC.We use Mobile access gateway(MAG) chain to reduce packet delay.The handoff delay and packet delivery delay under the proposed scheme are derived,based on which we formulate the delay minimization problem whose solution leads to the optimal MAG chain length.Numerical analysis results indicate that the proposed scheme outperforms the existing scheme in terms of delay in the case when the delay between Local mobility anchor(LMA) and WLC is relatively greater than the delay between two neighboring WLCs.The proposed scheme is able to reduce packet loss resulting from the traditional handoff procedure introduced in the PMIPv6 protocol and that due to delay limitation.ZHU Yihua ZHU Deqing PAN Genmei CHI Kaikai LI Yanjun 2017Chinese Journal of Electronics2017,26,5:1
