| 1 | Holocene aeolian stratigraphic sequences in the eastern portion of the desert belt(sand seas and sandy lands) in northern China and their palaeoenvironmental implications显示文摘This paper presents the environmental history and its responses to palaeoclimatic changes since the start of the Holocene in the eastern portion of the desert belt(sand seas and sandy lands) in northern China by comparing the aeolian sandpalaeosol sequences and their palaeoclimatic proxies. The optically stimulated luminescence(OSL) ages of the aeolian sandpalaeosol sedimentary sequences and a series of palaeoenvironmental proxies show that:(1) The large-scale dune landscape currently in the Kubuqi Sand Sea was formed during the Holocene in general;and the palaeosol was generally developed during the period of 4–2 ka, indicating conditions favorable for vegetation growth, soil development, and organic carbon accumulation due to increased precipitation or effective moisture and weakened aeolian activities;the large-scale expansion of dunes in the recent 2 ka is closely linked to human activities. The variable discharge of the Yellow River with diversions for irrigation may have resulted in a more consistent supply of aeolian particles for dune field expansion.(2) The dune landscape of the Hunshandake Sandy Land was likely formed around 12 ka, and before this, the western part of the Hunshandake Sandy Land would have been covered by a single large lake;it was obviously wetter than today in the sandy land during the period of 9.6–3 ka and the palaeosols were developed at the same time. But the aeolian activities have not been completely dormant in this long-lasting wetter epoch;because the Holocene wetter period was likely time-transgressive across the region.(3) The palaeosol of the Hulunbuir Sandy Land began to develop as early as 14.5 ka, probably continuing until the last 2 ka. The palaeosol development of various dune fields in the eastern portion of the desert belt(sand seas and sandy lands) in northern China is spatially heterogeneous, and even the palaeosol development time in different locations within each sandy land is inconsistent. During the middle Holocene(especially the 7.5–3.5 ka), all the sandy lands were stabilized in general and the intensity of aeolian activities was significantly weakened. The number of palaeoenvironmental records in the eastern portion of the desert belt(sand seas and sandy lands) in northern China has increased rapidly in the past decade, but the amount of published data still does not match the vast extent of the dune fields. | 5 | Experimental optimization of phase noise performance of optoelectronic oscillator based on directly modulated laser显示文摘The experimental optimization of a 10 GHz optoelectronic oscillator (OEO) based on a 1.55 μm directly modulated distributed feedback (DFB) laser is demonstrated in this paper. The phase noise of the directly modulated laser (DML) based OEO is significantly reduced by proper selection of the laser's bias current and by using a dispersion shifted fiber as a delay line. The phase noise performance of the DML OEO achieved after optimization is close to that of a conventional OEO constructed using a commercial DFB laser and a LiNbO 3 Mach-Zehnder modulator. | 6 | A competitive RT-PCR-based approach to quantifying promoter activity of stress gene显示文摘Chloramphenicol acetyltransferase (CAT) as a stable reporter has been broadly adopted to detect promoter activity of many stress genes in eukaryotic cells. A simplified, convenient and relatively accurate assay to reveal CAT activities driven by heat shock gene promoter upon cell stress is reported here. A CAT mutant control plasmid pMCAT is constructed out of pBLCAT3, with a DNA segment within the CAT gene deleted. The control plasmid pMCAT along with the wild type CAT reporter gene driven by the regulatory sequence of hsp90β gene are co-transfected into Jurkat cells. Following total cellular RNA extracted and reverse transcribed, PCR is carried out to amplify two individual fragments with the same pair of synthesized primers, which bind to the identical sequences in both pMCAT and the CAT reporter plasmids. The two PCR fragments amplified from the reporter and control RNAs are identified by their size differences in agarose gel electrophoresis. The ratio of intensity of the two PCR bands in the same lane is taken as 'relative units of promoter efficiency' of a gene. The transcripts instead of CAT activity thus detected, may not only provide a closer look at the promoter activity of the stress gene but also normalize the efficiency of transfection simultaneously. With experimental conditions optimized, visible and reliable results can be obtained without using any sophisticated instruments. Since the results obtained with the RT-PCR approach are reproducible, much less troublesome and time-consuming, and comparable to our previous data, we thus take it as a conventional procedure for promoter activity assay of human hsp gene upon heat shock. | 7 | Synaptic development in the injured spinal cord cavity following co-transplantation of fetal spinal cord cells and autologous activated Schwann cells显示文摘Transplantation of activated transgenic Schwann cells or a fetal spinal cord cell suspension has been widely used to treat spinal cord injury. However, little is known regarding the effects of co-transplantation. In the present study, autologous Schwann cells in combination with a fetal spinal cord cell suspension were transplanted into adult Wistar rats with spinal cord injury, and newly generated axonal connections were observed ultrastructurally. Transmission electron microscopic observations showed that the neuroblast first presented cytoplasmic processes, followed by pre- and postsynaptic membranes with low electron density forming a dense projection. The number and types of synaptic vesicles were increased. Synaptic connections developed from single cell body-dendritic synapses into multiple cell body-dendritic and dendrite-dendritic synapses. In addition, the cell organs of the transplanted neuroblast, oligodendroblast and astroblast matured gradually. The blood-brain barrier appeared subsequently. Moreover, neurofilament, histamine, calcitonin-gene-related peptides, and glial fibrillary acidic protein positive fibers were observed in the transplant region.
