1How is mRNA expression predictive for protein expression? 2Preparation of fine copper powders and their application in BME-MLCC显示文摘The preparation of fine copper powders by chemical reduction method was investigated. The reaction of [Cu(NH3)4] 2+ complex with hydrazine hydrate gives spherical monodispersed fine copper powders. The spherical copper powder with a uniform size of 3.5 ± 0.5 μm was processed to obtain flake copper powder having a uniform size of 8-10 μm, excellent dispersibility and uniform shape. The spherical copper powder of 2.5 ± 0.3 μm in size, flake copper, glass frit and vehicle were mixed to prepare copper paste, which was fired in 910-920°C to obtain BME-MLCC (base metal multilayer ceramic capacitor) with a dense surface of end termination, high adhesion and qualified electrical behavior. Polarized light photo and SEM were employed to observe the copper end termination of BME-MLCC. 3Evaluation of strategy for analyzing mouse liver plasma membrane proteome显示文摘Plasma membrane (PM) proteome is one of the major subproteomes present in the cell,and is very important in liver function. In the present work, C57 mouse liver PM was purified by density-gradient centrifugation. The purified PM was verified by electron microscope analysis and Western blotting. The results showed that the PM was enriched by more than 20-fold and the contamination of mitochondria was reduced by 2-fold compared with the homogenization fraction. Proteins were separated by 2DE and 1DE, trypsin-digested and submitted to ESI-Q-TOF and MALDI-TOF-TOF mass spectrometry or directly digested in solution and analyzed by LC-ESI ion trap mass spectrometry. In all, 547 non-redundant mouse liver PM proteins were identified, of which 34% contributed to plasma membrane or plasma membrane-related proteins. 4Ambient temperature alters body size and gut microbiota of Xenopus tropicalis显示文摘Temperature is important to determine physiological status of ectotherms. However, it is still not fully understood how amphibians and their symbiotic microbiota acclimate to ambient temperature. In this study, we investigated the changes of gut microbiota of Xenopus tropicalis at different temperatures under controlled laboratory conditions. The results showed that microbial communities were distinct and shared only a small overlap among froglet guts, culture water and food samples.Furthermore, the dominant taxa harbored in the gut exhibited low relative abundance in water and food. It indicates that bacterial taxa selected by amphibian gut were generally of low abundance in the external environment. Temperature could affect betadiversity of gut microbiota in terms of phylogenetic distance, but it did not affect alpha diversity. The composition of gut microbiota was similar in warm and cool treatments. However, signature taxa in different temperature environments were identified. 10Differential protein expression in low-grade astrocytomas and peritumoral human brain tissues显示文摘Differential protein expression between various pathological grades of glioma has been shown in studies of glioma proteomics. However,very little data is available regarding normal brain tissues and glioma differential protein expression,because normal human brain tissues are difficult to harvest. The present study selected samples from low-grade astrocytomas and peritumoral brain tissues to analyze differential protein expression by two-dimensional(2D) electrophoresis and mass spectrometry techniques. Results revealing 36 protein spots by 2D electrophoresis,including 23 spots revealing increased expression and 13 spots revealing decreased expression. However,25 differential proteins were identified by mass spectrometry,including 16 proteins with increased expression and 9 with decreased expression. Western blot analysis confirmed the mass spectrometry results,i.e.,heat shock protein 70(HSP70) and human transthyretin(TTR) expressions were increased,but glial fibrillary acidic protein(GFAP) was decreased,in astrocytomas. The present study constructed a 2D electrophoresis pattern between low-grade astrocytomas in the human brain and peritumoral tissues. 11Effects of Arg26 and Lys27 mutation on the bioactivity of HNTX-Ⅳ显示文摘Hainantoxin-Ⅳ(HNTX-Ⅳ)was isolated from the Chinese bird spider Ornithoctorcs hainana and identified as a novel antagonist of tetrodotoxin-sensitive(TTX-S)sodium channels.As revealed by the solution structure of HNTX-Ⅳ solved by two-dimensional nuclear magnetic resonance(2D-NMR),HNTX-Ⅳ adopts an inhibitor cystine knot motif.To check the role of basic residues during HNTX-Ⅳ’s interaction with TTX-S sodium channels,R26A and K27A mutants of HNTX-Ⅳ were constructed by solid-phase chemical synthesis.The synthesized peptides were purified and refolded under optimized oxidation conditions.Correct synthesis and folding were confirmed by MALDI-TOF mass spectrometry and NMR spectroscopy,respectively.Using the whole-cell patch-clamp technique,Lys27 but not Arg26 was identified as a key residue for HNTX-Ⅳ’s bioactivity against TTX-S sodium channels,because R26A-HNTX-Ⅳ showed slightly reduced activity and K27A-HNTX-Ⅳ showed almost no inhibition.XIONG Xia XU Xia LI Dongling CHEN Ping LIANG Songping 2007Frontiers in Biology2007,2,1:0
12Leaf Morphology Genes SRL1 and RENL1 Co-Regulate Cellulose Synthesis and Affect Rice Drought Tolerance显示文摘The morphological development of rice(Oryza sativa L.)leaves is closely related to plant architecture,physiological activities,and resistance.However,it is unclear whether there is a co-regulatory relationship between the morphological development of leaves and adaptation to drought environment.In this study,a drought-sensitive,roll-enhanced,and narrow-leaf mutant(renl1)was induced from a semi-rolled leaf mutant(srl1)by ethyl methane sulfonate(EMS),which was obtained from Nipponbare(NPB)through EMS.Map-based cloning and functional validation showed that RENL1 encodes a cellulose synthase,allelic to NRL1/OsCLSD4.The RENL1 mutation resulted in reduced vascular bundles,vesicular cells,cellulose,and hemicellulose contents in cell walls,diminishing the water-holding capacity of leaves.In addition,the root system of the renl1 mutant was poorly developed and its ability to scavenge reactive oxygen species(ROS)was decreased,leading to an increase in ROS after drought stress.Meanwhile,genetic results showed that RENL1 and SRL1 synergistically regulated cell wall components.Our results revealed a theoretical basis for further elucidating the molecular regulation mechanism of cellulose on rice drought tolerance,and provided a new genetic resource for enhancing the synergistic regulation network of plant type and stress resistance,thereby realizing simultaneous improvement of multiple traits in rice.LIU Dan ZHAO Huibo WANG Zi’an XU Jing LIU Yiting WANG Jiajia CHEN Minmin LIU Xiong ZHANG Zhihai CEN Jiangsu ZHU Li HU Jiang REN Deyong GAO Zhenyu DONG Guojun ZHANG Qiang SHEN Lan LI Qing QIAN Qian HU Songping ZHANG Guangheng 2024Rice science2024,31,1:0
13Microacoustic Imaging Technique for Bulk Microstructure Characterization of Woven CFRC Materials显示文摘In short wavelength case ultrasound propagation through woven carbon fiber-rein-forced composite (CFRC) materials essentially depends on their microstructure.Fiber bundle and weaving ply can be considered as basic structural elements of woven CFRC laminates.These structural units provide effective reflectors when ultra-short impulse high frequency ultrasound impinges on woven laminates.The reflection coefficient R0 achieves level of 3%-15% at ply-ply interface,0.2-0.4 in resin pocket region.This magnitude of reflection coefficient offers reasonable acoustic contrast to see packing,lay-up and resin pockets in woven laminate body by acoustic imaging methods.Microacoustic imaging technique is based on time-resolved echoes from reflections in woven CFRC laminates.A focal system with low aperture angle is used to generate 20-40 probe ultrasonic pulse wave.Its spatial resolution is up to 60mm at 50MHz in woven CFRC laminate.The results presented in this paper demonstrates the possibility and potentiality of characterization of microstrueture of woven CFRC laminates.Packing,lay-up,weaving nature,fiber bundle,resin pocket as well as their topology are exhibited clearly in the acoustic images by using microaeoustic technique.Liu Songping Guo Enming Liu Feifei Li Legang V.M.Levin Yu S Petronyuk 2007航空制造技术2007,50,z1:0
14Mechanochemical synthesis of oxygenated alkynyl carbon materials with excellent Hg(Ⅱ) adsorption performance from CaC2 and carbonates显示文摘Adsorptive removal of heavy metal ions from wastewater is very important,and the key is the development of efficient sorbents.In this work,oxygenated alkynyl carbon materials(OACMs)were synthesized via mechanochemical reaction of CaC_(2) and a carbonate(CaCO_(3),Na2CO_(3),or NaHCO_(3))at ambient temperature.The resultant OACMs are micro mesoporous carbon nanomaterials with high specific area(>648 m2 g^(-1)),highly crosslinked texture,and rich alkynyl and oxygenated groups.The OACMs exhibit excellent Hg(Ⅱ)adsorption due to the soft acid-soft base interaction between alkynyl and Hg(Ⅱ),and OACM-3 derived from CaC_(2) and NaHCO_(3) has the saturated Hg(Ⅱ)adsorbance of 483.9 mg g^(-1)along with good selectivity and recyclability.The adsorption is mainly chemisorption following the Langmuir mode.OACM-3 also shows high adsorbance for other heavy metal ions,e.g.256.6 mg g^(-1)for Pb(II),232.4 mg g^(-1)for Zn(II),and 198.7 mg g^(-1)for Cu(II).This work expands the mechnochemical reaction of CaC_(2)with carbonates and possibly other oxyanionic salts,provides a new synthesis approach for functional alkynyl carbon materials with excellent adsorption performance for heavy metal ions,as well as a feasible approach for CO2 resource utilization.Yingjie Li Songping Li Xinyi Xu Hong Meng Yingzhou Lu Chunxi Li 2023Green Energy & Environment2023,8,1:0
15Apoferritin nanoparticle based dual-antigen influenza conjugate vaccine with potential cross-protective efficacy against heterosubtypic influenza virus显示文摘Ferritin nanoparticles with self-assembling properties have been widely explored as vaccine carrier by displaying foreign antigens through genetic fusion strategy.In the present work,an apoferritin(AFt)nanoparticle was tested as influenza vaccine carrier by chemically conjugating a matrix protein 2 ectodomain(M2e)antigen peptide or/and the full-length hemagglutinin(HA)antigen on the outer surface of the AFt,with heterobifunctional sSMCC or SM(PEG)_(24) containing PEG chain as linkers.To each AFt nanoparticle,about 30-32 M2e or 1.8 HA antigen could be coupled.The AFt-(PEG)24-M2e,in which the M2e was coupled through SM(PEG)_(24) containing PEG chain,conferred higher protective efficacy in immunized mice than AFt-M2e did,but was less effective than AFt-(PEG)_(24)-HA.When both M2e and HA were coupled,the synthesized dual-antigen vaccine candidate AFt-(PEG)_(24)-M2e/HA elicited high level of M2e and HA antigen-specific antibodies and conferred 100%protection against lethal infection of homologous PR8 HI N1 virus strain and 70%protection against a heterologous A/FM/1/47(FM1,H1N1)strain,which was more effective than the M2e or HA single antigen vaccine candidates.The potential cross-protective effect of the dual-antigen vaccine was further demonstrated by significant specific hemagglutination inhibition(HAI)titers in serum of the immunized mice against three other heterologous viral strains including A/Singapore/GPl908/2015(IVR-180)H1N1,A/Anhui/1/2005 H5N1,and A/Hong Kong H3N2.Yanan Sheng Jiangxue Wei Zhengjun Li Zhiguo Su Guanghui Ma Songping Zhang 2022Particuology2022,20,5:0
