| 1 | A high-resolution clay mineralogical record in the northern South China Sea since the Last Glacial Maximum,and its time series provenance analysis显示文摘High-resolution clay mineralogical analysis of Core MD05-2904 in the northern South China Sea(SCS) covering the period since the Last Glacial Maximum shows that illite(29%-48%),smectite(14%-45%),chlorite(17%-28%),and minor kaolinite(6%-14%) comprise the clay mineral assemblage,and that time series variation does not present glacial-interglacial cyclicity.Provenance analysis indicates three end-member sources:almost all smectite derives from Luzon,all kaolinite is sourced from the Pearl River,and illite and chlorite originate from both the Pearl River and Taiwan.By comparing clay mineral compositions in surface sediments from the three major source areas and of the SCS,we reconstructed a time series of clay mineral contribution from the major provenances to the northern slope of the SCS using the linear separation method for illite crystallinity.There were three stages of provenance change.(1) During 24.1-17.5 ka BP,contributions from Taiwan and Luzon were similar(30%-40%),while that from the Pearl River was only 25%.(2) During 17.5-14.0 ka BP,the contribution from Luzon decreased rapidly to 20%-25%,while that from Taiwan increased to 35% from an average of 25% at 18 ka BP,and that from the Pearl River increased largely to 40%.(3) During the Holocene,differences in contributions from the three major provenances increased:the contribution from Luzon increased slightly and then remained at 27%-35%,that from Taiwan increased rapidly and then remained at 55%-60%,and that from the Pearl River decreased to 15%.The change in clay mineral contributions from different provenances is influenced mainly by clay mineral production,monsoon rainfall denudation,oceanic current transport,and sea-level change. | LIU ZhiFei LI XiaJing COLIN Christophe GE HuangMin | 2010 | Chinese Science Bulletin2010,55,35: | 20 |
| 2 | Iterative spherical simplex unscented particle filter for CNS/Redshift integrated navigation system显示文摘We propose an improved Unscented Particle Filter(UPF) algorithm for the Celestial Navigation System/Redshift(CNS/Redshift) integrated navigation system. The algorithm adopts the iterated spherical simplex unscented transformation rather than the traditional unscented transformation. The navigation performance of the proposed algorithm is assessed by several indexes. | 3 | CircRNA.0007127 triggers apoptosis through the miR-513a-5p/CASP8 axis in K-562 cells显示文摘Background:Circular RNAs(circR NAs)are covalently closed single-stranded RNAs with multiple biological functions.CircRNA.0007127 is derived from the carbon catabolite repression 4-negative on TATA-less(CCR4-NOT)complex subunit2(CNOT2),which was found to regulate tumor cell apoptosis through caspase pathway.Methods:Potential circR NA.0007127 target microRNAs(miRNAs)were analyzed by miRanda,TargetScan,and RNAhybrid software,and the miRNAs with binding sites for apoptosis-related genes were screened.The roles of circR NA.0007127 and its downstream target,microR NA(miR)-513a-5p,were validated by quantitative real-time polymerase chain reaction(qPCR),flow cytometry,mitochondrial membrane potential,immunofluorescence,western blot,and caspase-8(CASP8)protein activity in vitro in HO-induced K-562 cells.The circRNA.0007127-miR-513a-5p and CASP8-miR-513a-5p interactions were verified by luciferase reporter assays.Results:Silencing circRNA.0007127 decreased cell apoptosis by inhibiting CASP8 pathway activation in K-562 cells.Compared with the control group,the expression of CASP8 was reduced by 50%and the 43-kD fragment of CASP8 protein was significantly reduced(P≤0.05).The luciferase reporting assay showed that circRNA.0007127 combined with miR-513a-5p or CASP8,with extremely significant differences(P≤0.001).The overexpression of miR-513a-5p inhibited the gene expression level of CASP8in a human myeloid leukemia cell model(75%change)and the level of a 43-kD fragment of CASP8 protein(P small-interfering RNA(siRNA)and the miR-≤0.01).The rescue experiment showed that cotransfection with circRNA.0007127513a-5p inhibitor increased CASP8 gene expression and the apoptosis rate,suggesting that the miR-513a-5p inhibitor is a circRNA.0007127siRNA antagonist.Conclusions:CircRNA.0007127 regulates K-562 cell apoptosis through the miR-513a-5p/CASP8 axis,which can serve as a novel powerful molecular target for K-562 cells. | Xiajing LI Yiyu ZHANG Ning WANG Zhaohu YUAN Xiaojie CHEN Qicong CHEN Hui DENG Xinxin TONG Honglin CHEN Yuyou DUAN Yaming WEI | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,9: | 1 |

| 8 | Hypoglycemic activities of flowers of Xanthoceras sorbifolia and identification of anti-oxidant components by off-line UPLC-QTOF-MS/MS-free radical scavenging detection显示文摘Objective: To identify phytochemical constituents present in the extract of flowers of Xanthoceras sorbifolia and evaluate their anti-oxidant and anti-hyperglycemic capacities.Methods: The AlCl3colorimetric method and Prussian Blue assay were used to determine the contents of total flavonoids and total phenolic acids in extraction layers, and the bioactive layers was screened through anti-oxidative activity in vitro. The Waters ACQUITY UPLC system and a Waters ACQUITY UPLC BEH C18column(2.0 mm × 150 mm, 5 μm) were used to identify the ingredients. And anti-oxidative ingredients were screened by off-line UPLC-QTOF-MS/MS-free radical scavenging. The ameliorative role of it was further evaluated in a high-fat, streptozotocin-induced type 2 diabetic rat model and the study was carried out on NADPH oxidase(PDB ID: 2CDU) by molecular docking.Results: Combined with the results of activity screening in vitro, the anti-oxidative part was identified as the ethyl acetate layer. A total of 24 chemical constituents were identified by liquid chromatographymass spectrometry in the ethyl acetate layer and 13 main anti-oxidative active constituents were preliminarily screened out through off-line UPLC-QTOF-MS/MS-free radical scavenging. In vivo experiments showed that flowers of X. sorbifolia could significantly reduce the blood glucose level of diabetic mice and alleviate liver cell damage. Based on the results of docking analysis related to the identified phytocompounds and oxidase which involved in type 2 diabetes, quercetin 3-O-rutinoside, kaempferol-3-O-rhamnoside, isorhamnetin-3-O-glucoside, and isoquercitrin showed a better inhibitory profile.Conclusion: The ethyl acetate layer was rich in flavonoids and phenolic acids and had significant anti-oxidant activity, which could prevent hyperglycemia.
