| 1 | Cloning and analysis of expression profile of 13 WRKY genes in rice显示文摘The transcriptional factor WRKY proteins contain the highly conserved amino acid sequence WRKYGQK as well as the novel zinc-finger-like motifs Cys2His2 or Cys2HisCys. A search of the rice genome identi-fied 97 genes encoding WRKY proteins. Of these 97 WRKY homologs found in rice, 13 cDNAs encoding WRKY proteins were consequently isolated from a rice cDNA library con-structed from 4℃-treated shoots by probing for the con-served WRKY domain. Northern blotting analysis revealed that 10 of 13 OsWRKY genes were differentially regulated in plants that were treated by the four following abiotic stress factors: NaCl,PEG,cold (4℃) and heat (42℃). The resulting expression profiles exhibited great differences in both the manner and timing of their response to the four different abiotic treatments. | 2 | Cloning and Characterization of Adinbitor,a Novel Disintegrin from the Snake Venom of Agkistrodon halys brevicaudus stejneger显示文摘Adinbitor was cloned from Agkistrodon halys brevicaudus stejneger and characterized as a novel disintegrin. In this study, total RNA was extracted from venom gland and used in RT-PCR to generate a cDNA which is 219 bp long. The sequence encoded a polypeptide composed of 73 amino acids, including 12 cysteines, an RGD motif, and the signature motif of disintergrin. Recombinant Adinbitor (rAdinbitor) was expressed in E. coli and purified by using the His?Bind affinity chromatography. The IC50 for inhibiting human platelet aggregation and bFGF-induced proliferation of ECV304 cells was 6 μΜ and 0.89 μΜ respectively. Furthermore, Adinbitor significantly inhibited angiogenesis both in vivo and in vitro. | 4 | Efficient polymerization of acrylonitrile catalyzed by divalent lanthanide complex/sodium phenolate systems显示文摘Four divalent lanthanide complexes Sm(ArO)2 -(THF)4) Yb(ArO)2(THF)3, Eu(ArO)2(THF)3 (ArO=2,6-di-tert-butyl-4-methylphenolate) and (ButCp)2Sm(THF)2 were synthesized. Their catalytic activities on the polymerization of acrylonitrile were studied. The catalytic activities were influenced by the central metal ions involved. The catalytic activities of these divalent lanthanide complexes can be greatly increased by adding NaOC6H2-2,6-But2-4-Me, NaOC6H4-4-But, or NaOC10H6-2-Me. | 5 | Magnetic properties and magnetoresistance of HfFe6Ge6—type Dy1—xGdxMn6Ge6(x=0.1—0.6)compounds显示文摘The magnetic properties and magnetoresistance effects of Dy1-xGdxMn6Ge6(x=0.1-0.6) compounds have been studied by magnetic properties and resistivity measurements in applied magnetic fields up to 5T. The compounds with x=0.1,0.2,0.4 and 0.5 order antiferromagnetically at 425,428,430 and 432K, respectively, and there are second magnetic phase transitions below 100K. The compound with x=0.6 exhibits a transition from ferrimagnetic to antiferromagnetic, then to ferrimagnetic state again with decreasing temperature. Furthermore, it displays a field-induced metamagnetic transition, and its threshold field decreases with increasing temperature. | 20 | Expression of a novel pyridoxal kinase mRNA splice variant,PKH-T, in human testis显示文摘Aim: To identify the genes specifically expressed in human adult and fetal testes and spermatozoa.Methods: A human testis cDNA microarray was established. Then mRNAs of human adult and fetal testis and spermatozoa were purified and probes were prepared by a reverse transcription reaction with mRNA as the template.The microarray was hybridized with probes of adult and fetal testes and spermatozoa. The nucleic acid sequences of differentially expressed genes were determined and homologies were searched in the databases of GenBank. Results:A novel human testis-specific gene, PKH-T, was identified by hybridizing adult and fetal testis and spermatozoa probes with a human testis cDNA microarray. The cDNA of PKH-T was 1069 bp in length. The cDNA sequence of this clone was deposited in the Genbank (AY303972) and PKH-T was also determined as Interim GenSymbol (Unigene,HS.38041). PKH-T contained most PKH conserved motif. The 239 amino acid sequences deduced from the 719 bp open reading frame (ORF) had a homology with the gene PKH (U89606). PKH-T was specifically and strongly expressed in the testis. Comparison of the differential expressions of PKH and PKH-T in testes of different develop-mental stages indicated that PKH-T was expressed in the adult testis and spermatozoa, while PKH, in the adult, fetal and aged testes. PKH-T had no expression in the testis of Sertoli cell only and partially spermatogenic arrest patients.Conclusion: PKH-T is a gene highly expressed in adult human testis and spermatozoa.
