2Therapeutic dendritic cell vaccines engineered with antigen-biomineralized Bi_(2)S_(3) nanoparticles for personalized tumor radioimmunotherapy显示文摘Therapeutic vaccines,an exciting development in cancer immunotherapy,share the goal of priming of personalized antigen-specific T-cell response by precise antigen presentation of dendritic cells(DCs),but major obstacles include insufficient antigen loading and off-target to DCs remain to their success.Here,we developed an imageable therapeutic vaccine with whole-antigen loading and target delivery constructed by ovalbumin(OVA)-biomineralized Bi_(2)S_(3) nanoparticles-pulsed DCs.Relying on the strong X-ray absorption and fluorescence labeling performance of Bi_(2)S_(3)@OVA nanoparticles,the in vivo spatiotemporal fate of the vaccine(Bi_(2)S_(3)@OVA@DC)can be noninvasively monitored by computed tomography and near-infrared fluorescence imaging in real time.The Bi_(2)S_(3)@OVA@DC can rapidly and durably accumulate in draining lymph nodes and thus trigger stronger T-cell responses compared to OVA-pulsed DCs.Meanwhile,Bi_(2)S_(3)@OVA@DC can further achieve in vivo antitumor effects against OVA-expressing B16F10 melanoma when combined with fractionated radiotherapy,resulting from the upregulation of cytotoxic CD8^(+)T cells and restraint of regulatory T cells in the tumor microenvironment,and the systemical secretion of OVA-specific IgG1/IgG2α antibody.Overall,we successfully fabricated an engineered DC vaccine featured in high whole-antigen loading capacity that can be precisely delivered to the lymphatic system for visualization,serving as a powerful therapeutic platform for cancer radioimmunotherapy.Huan Yu Haoxiang Guo He Zu Heng Ding Subin Lin Yangyun Wang Leshuai W Zhang Yong Wang 2022Aggregate2022,3,5:1
3Visualizable Delivery of Nanodisc Antigen-Conjugated Adjuvant for Cancer Immunotherapy显示文摘Engineering synthetic vaccines is promising for improving the efficacy of cancer immunotherapy.One of the major challenges in the development of vaccines is achieving controllable codelivery of antigen and adjuvant to lymph nodes for maximizing the antitumor immune responses.To address this issue,we herein developed an innovative visualizable nanodisc vaccine based on ovalbumin(OVA)and short-stranded oligodeoxynucleotides containing unmethylated cytosine-phosphate-guanine(CpG)motifs.The nanovaccine was fabricated by covalently attaching CpG onto the surface of a nanodisc antigen formed upon self-assembly of amphiphilic molecular conjugates of OVA and cypate(Cy),a near-infrared(NIR)fluorescent dye,for noninvasively visualizing the delivery of the resulting nanovaccines.Systematic in vitro experiments demonstrated that the engineered nanovaccines can specifically locate to dendritic cells(DCs)via toll-like receptor 9 and membrane thiols,and then efficiently activate DCs.The animal experiments combining NIR fluorescence imaging with the lymphatic T-cell phenotype and key cytokine secretion analyses revealed that targeted lymphatic homing of the mature DCs and consequent priming of CD8^(+)T cells were enabled to initiate strong tumor-specific T-cell responses with robust immune memory effects.Thus,this study offers a visualizable platform for optimizing the efficacy of nanovaccines toward cancer immunotherapies.Yangyun Wang Subin Lin Hanqiu Jiang Yuan Gu Yanxian Wu Jie Ma Yubin Ke Leshuai W.Zhang Yong Wang Mingyuan Gao 2022CCS Chemistry2022,4,4:0
4Simulation and Modeling of Common Mode EMI Noise in Planar Transformers显示文摘The transformer is the key circuit component of the common-mode noise current when an isolated converter is working.The highfrequency characteristics of the transformer have an important influence on the common-mode noise of the converter.Traditionally,the measurement method is used for transformer modeling,and a single lumped device is used to establish the transformer model,which cannot be predicted in the transformer design stage.Based on the transformer common-mode noise transmission mechanism,this paper derives the transformer common-mode equivalent capacitance under ideal conditions.According to the principle of experimental measurement of the network analyzer,the electromagnetic field finite element simulation software three-dimensional(3D)modeling and simulation method is used to obtain the two-port parameters of the transformer,extract the high-frequency parameters of the transformer,and establish its electromagnetic compatibility equivalent circuit model.Finally,an experimental prototype is used to verify the correctness of the model by comparing the experimental measurement results with the simulation prediction results.LI Wei JI Jingkang LIU Yuanlong SUN Jiawei LIN Subin 2023ZTE Communications2023,21,3:0
