| 3 | Efficacies of β-L-D4A against Hepatitis B virus in 2.2.15 cells显示文摘AIM:To investigate the antiviral effect of beta-L-enantiomer of 2',3'-didehydro-2',3'-dideoxyadenosine (β-L-D4A) on 2.2.15 cells transfected with the hepatitis B virus (HBV) genome. METHODS:Lamivudine (3TC) as a positive control. Then, HBV DNA in treated 2.2.15 cells and the Hepatitis B surface antigen (HBsAg) in the culture supernatants were detected to determine the inhibitory effect of β-L-D4A. At the same time, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was used to detect the survival ratio of 2.2.15 cells. RESULTS:β-L-D4A has a dose-dependent inhibitory effect on HBV DNA replication;this effect was apparent when the concentration was above 1 mol/L. When β-L-D4A was at the highest concentration, 100 mol/L, the HBsAg inhibition ratio was above 50%. The Therapeutic index (TI) of β-L-D4A was above 2.1. | 5 | GSPAC water movement in extremely dry area显示文摘Under an extremely arid condition,a PVC greenhouse was built on the top of Mogao Grottoes in gobi area.The results of 235-day constant extraction of condensed water on the greenhouse film and soil water content showed that 2.1 g/(m2·d) groundwater moved up and exported into the soil,and a phreatic water evaporation existed in the extreme dry area where the groundwater is buried deeper than 200 m.After a prolonged export,the soil water content in the greenhouse was not lower but obviously higher than the original control ones.According to the monitored parameters including relative humidity and absolute humidity of soil,and temperature outside and inside the greenhouse,it was found that there is the available condition and mechanism for the upward movement of groundwater,and also it can be sure that the exported water was not from the soil and atmosphere outside the greenhouse.Phreatic water,an important source for soil water,interacts with atmosphere moisture via soil respiration.Soil salinity also has important effects on soil water movement and spatial-temporal heterogeneity.The extremely dry climate,terrestrial heat and change of upper soil temperature are the fundamental driving forces of water transportation and phreatic water evaporation in the Groundwater-Soil-Plant-Atmosphere Continuum(GSPAC) system. | HongShou Ll WanFu WANG GuoBin ZHANG ZhengMo ZHANG XiaoWei WANG | 2011 | Journal of Arid Land2011,3,2: | 4 |

| 9 | Design,synthesis,and anti-inflammatory evaluation of a series of novel amino acid-binding 1,5-diarylpyrazole derivatives显示文摘Aim: To design and synthesize a series of novel amino acid-binding 1,5-diarylpyrazole derivatives, which are intended to act as prodrugs with better aqueous solubility than celecoxib, and which will exert potent anti-inflammatory activi-ties after being converted to their parent compounds in vivo. Methods: To introduce an amino acid, celecoxib analogs containing amino or methylamino group were synthesized first through multi-step chemical reactions. All the synthesized compounds were screened in an intact cell-based assay in vitro and in carrageenan-induced mouse paw edema in vivo. Some active compounds were selected for further evaluation in a carrageenan-induced rat paw edema model. The preliminary pharmacokinetics experiments were conducted using high performance liquid chromatography/mass spectrometry (HPLC/MS). Results: Celecoxib, 6 of the 1,5-diarylpyrazole class of celecoxib analogs, and their amino acid derivatives (hydrochloride salts) were synthesized. In vitro screening, the hydrochloride salts showed decreased inhibitory effects on cyclooxygenase (COX)- 1 and COX-2 compared with their parent compounds, but some exhibited potent anti-inflammatory activity in vivo. Compound 4a was selected for further evaluation, and its anti-inflammatory effect was equivalent to that of celecoxib after oral administration in the carrageenan-induced rat paw edema model. At three doses (25 mg/kg, 50 mg/kg, and 100 mg/kg) the percentage inhibition on edema was 20.7%, 52.6%, and 62.6% (for compound 4a) and 27.8%, 38.4%, and 40.1% (for celecoxib), respectively. Preliminary pharmacokinetic evaluations support the hypothesis that compound 4a was actually converted to its parent compound, compound 4. Conclusion: The compound bound with amino acid acts like prodrug, which can exert anti-inflammatory effect similar to celecoxib after being converted to its parent compound.
