1Ferroptosis is essential for diabetic cardiomyopathy and is prevented by sulforaphane via AMPK/NRF2 pathways显示文摘Herein,we define the role of ferroptosis in the pathogenesis of diabetic cardiomyopathy(DCM)by examining the expression of key regulators of ferroptosis in mice with DCM and a new ex vivo DCM model.Advanced glycation end-products(AGEs),an important pathogenic factor of DCM,were found to induce ferroptosis in engineered cardiac tissues(ECTs),as reflected through increased levels of Ptgs2 and lipid peroxides and decreased ferritin and SLC7 A11 levels.Typical morphological changes of ferroptosis in cardiomyocytes were observed using transmission electron microscopy.Inhibition of ferroptosis with ferrostatin-1 and deferoxamine prevented AGE-induced ECT remodeling and dysfunction.Ferroptosis was also evidenced in the heart of type 2 diabetic mice with DCM.Inhibition of ferroptosis by liproxstatin-1 prevented the development of diastolic dysfunction at 3 months after the onset of diabetes.Nuclear factor erythroid 2-related factor 2(NRF2)activated by sulforaphane inhibited cardiac cell ferroptosis in both AGE-treated ECTs and hearts of DCM mice by upregulating ferritin and SLC7 A11 levels.The protective effect of sulforaphane on ferroptosis was AMP-activated protein kinase(AMPK)-dependent.These findings suggest that ferroptosis plays an essential role in the pathogenesis of DCM;sulforaphane prevents ferroptosis and associated pathogenesis via AMPK-mediated NRF2 activation.This suggests a feasible therapeutic approach with sulforaphane to clinically prevent ferroptosis and DCM.Xiang Wang Xinxin Chen Wenqian Zhou Hongbo Men Terigen Bao Yike Sun Quanwei Wang Yi Tan Bradley B.Keller Qian Tong Yang Zheng Lu Cai 2022Acta Pharmaceutica Sinica B2022,12,2:20
2QTL mapping for plant height and fruit branch number based on RIL population of upland cotton显示文摘Background:Plant height(PH)and fruit branch number(FBN)are important traits for improving yield and mechanical harvesting of cotton.In order to identify genes of PH and FBN in cotton germplasms to develop superior cultivars,quantitative trait loci(QTLs)for these traits were detected based on the phenotypic evaluation data in nine environments across four locations and 4 years and a previously reported genetic linkage map of an recombinant inbred line(RIL)population of upland cotton.Results:In total,53 QTLs of PH and FBN,were identified on 21 chromosomes of the cotton genome except chromosomes c02,c09-c11,and c22.For PH,27 QTLs explaining 3.81%–8.54%proportions of phenotypic variance were identified on 18 chromosomes except c02,c08-c12,c15,and c22.For FBN,26 QTLs explaining 3.23%–11.00%proportions of phenotypic variance were identified on 16 chromosomes except c02-c03,c06,c09-c11,c17,c22-c23,and c25.Eight QTLs were simultaneously identified in at least two environments.Three QTL clusters containing seven QTLs were identified on three chromosomes(c01,c18 and c21).Eleven QTLs were the same as previously reported ones,while the rest were newly identified.Conclusions:The QTLs and QTL clusters identified in the current study will be helpful to further understand the genetic mechanism of PH and FBN development of cotton and will enhance the development of excellent cultivars for mechanical managements in cotton production.LIU Ruixian XIAO Xianghui GONG Juwu LI Junwen ZHANG Zhen LIU Aiying LU Quanwei SHANG Haihong SHI Yuzhen GE Qun IQBAL Muhammad Sajid CHEN Quanjia YUAN Youlu GONG Wankui 2020Journal of Cotton Research2020,3,1:1
3Analysis of the effect of medical insurance on cancer inpatients: A 10-year retrospective study on a large hospital in Northeast China显示文摘Objective The aim of the study was to analyze hospital costs for cancer inpatients availing different methods of payment and the influencing factors, to provide inputs to improve the medical insurance payment policy. Methods We analyzed the information related to length of hospital stay, hospitalization cost, and self-pay cost, collected from one large-scale, Grade A, Class Three hospital in Shenyang, China, during 2004–2013.Results The number of cancer inpatients with different payment types(medical insurance group and non-medical insurance group) presented a rising trend. Further, the ratio of medical insurance inpatients increased rapidly(from 22.2% to 48.7%); however, this group was still a minority. The length of hospital stay became shorter(21 d vs. 17 d; P = 0.000) while the gap got narrower; the hospitalized expense showed an upward trend and the difference was remarkable($24048.6 ± $4376.28 vs. $20544.36 ± $4057.01; P = 0.000). 4Current advances in pathogen-plant interaction between Verticillium dahliaeand cotton provide new insight in the disease management显示文摘Verticillium wilt is the second serious vascular wilt caused by the phytopathogenic fungus Verticillium dahliae Kleb.It has distributed worldwide,causing serious yield losses and fiber quality reduction in cotton production.The pathogen has developed different mechanisms like the production of cell wall degrading enzymes,activation of virulence genes and protein effectors to succeed in its in fection.Cott on plant has also evolved multiple mechanisms in response to the fungus infection,including a strong production of lignin and callose deposition to strengthen the cell wall,burst of reactive oxygen species,accumulation of defene hormones,expression of defense-related genes,and target-directed strategies like cross-kingdom RNAi for specific virulent gene silencing.This review summarizes the recent progress made over the past two decades in understanding the interactions between cotton plant and the pathogen Verticillium dahliae during the infection process.The review also discusses the achievements in the control practices of cotton verticillium wilt in recent years,including cultivation practices,biological control,and molecular breeding strategies.These studies reveal that effective management strategies are needed to control the disease,while cultural practices and biological control approaches show promising results in the future.Furthermore,the biological control approaches developed in recent years,including antagonistic fungi,endophytic bacteria,and host induced gene sile ncing strategies provide efficie nt choices for in teg rated disease management.PALANGA Koffi Kibalou LIU Ruixian GE Qun GONG Juwu LI Junwen LU Quanwei LI Pengtao YUAN Youlu GONG Wankui 2021Journal of Cotton Research2021,4,3:0
5Rapid Identification of a Candidate Gene Related to Fiber Strength Using a Superior Chromosome Segment Substitution Line from Gossypium hirsutum × Gossypium barbadense via Bulked Segregant RNA-Sequencing显示文摘Cotton is the most widely cultivated commercial crop producing natural fiber around the world.As a critical trait for fiber quality,fiber strength principally determined during the secondary wall thickening period.Based on the developed BC5F3:5 CSSLs(chromosome segment substitution lines)from Gossypium hirsutum CCRI36×G.barbadense Hai 1,the superior MBI9915 was chosen to construct the secondary segregated population BC7F2 with its recurrent parent CCRI36,which was subsequently subjected to Bulk segregant RNA-sequencing(BSR-seq)for rapid identification of candidate genes related to fiber strength.A total of 4 fiber-transcriptome libraries were separately constructed and sequenced,including two parents(CCRI36 and MBI9915)and two extreme pools at 20 DPA(days post anathesis).Through multiple comparisons,536 DEGs(differentially expressed genes)were overlapped at 20 DPA.Allelic-polymorphism comparison in mRNA sequences revealed 831 highly probable SNPs between two extreme pools related to fiber strength.Linkage analysis was performed between two extreme pools with SNP-index method.Eighteen correlated regions with 1981 annotation genes were obtained between two pools at 20 DPA,of which 12 common DEGs were similarly identified both between two parents and two pools.One gene(Gh_A07G0837)in the candidate region related to fiber strength was differentially expressed in both parents and extreme pools and involved in fiber strength development through reactive oxygen species(ROS)activity.Co-expression analysis of Gh_A07G0837 showed that Gh_A07G0837 may cooperate with other genes to regulate fiber strength.The reliability of BSR-seq results was validated by the quantitative real-time PCR(qRT-PCR)experiments on 5 common DGEs 20 DPA.Co-expressed analysis results indicated that there were some genes expressed especially low in MBI9915,resulting in good fiber strength.Focusing on bulked segregant analysis on the extreme pools derived from superior CSSL population,this study indicates that BSR-seq can be efficiently applied on rapid identification of candidate genes related to fiber strength,which make contributions to our understanding of fiber quality formation in cotton.Qi Zhang Pengtao Li Aiying Liu Shaoqi Li Quanwei Lu Qun Ge Junwen Li Wankui Gong Xiaoying Deng Haihong Shang Yuzhen Shi Youlu Yuan 2021Phyton-International Journal of Experimental Botany2021,90,3:0
6Dynamic Expression Analysis and Introgressive Gene Identification of Fiber Length Using Chromosome Segment Substitution Lines from G.hirsutum×G.barbadense显示文摘Fiber length is a critical trait that principally determines cotton spinning quality,while Upland cotton as the most widely cultivated Gossypium species around the world subjects to the relatively ordinary fiber performance.Chromosome segment substitution lines(CSSLs)have been introduced in cotton breeding to take full advantages of superior fiber quality and high yield from Sea Island and Upland cotton,respectively,which serve as ideal materials for elucidating the genetic mechanism of complex quantitative traits.Here,three CSSLs derived from CCRI45(G.hirsutum)×Hai1(G.barbadense),two superior(MBI7561 and MBI7747)and one(MBI7285)with ordinary fiber-quality,were subjected to transcriptome sequencing during fiber elongation together with their recurrent parent CCRI45,and 471.425 million clean reads were obtained with 91.47%average Q30 and 45.23%mean GC content.In total,5,673 differentially expressed genes(DEGs)were identified from multi-sample comparisons,which were mainly involved in the oxidation-reduction process,protein phosphorylation,regulation of transcription,DNA template,and carbohydrate metabolic process.Eight temporal expression patterns were monitored on the DEGs of different lines,of which the significantly enriched profile revealed higher similarities between two superior CSSLs or the ordinary CSSL and CCRI45 with respect to fiber performance.Based on the intersection between the predicted introgressive genes from RNAseq data and the published gene information from the G.barbadense genome,1,535 introgressive genes were identified in three CSSLs.Further analysis of the three common introgressive sections in superior CSSLs revealed eight candidate genes that were identified to be involved in fiber development,namely,O-fucosyltransferase family protein(GB_A02G0240),glutamine synthetase 2(GB_A02G0272),Ankyrin repeat family protein(GB_A02G0264),beta-6 tubulin(GB_D03G1742),WRKY DNA-binding protein 2(GB_D03G1655),quinolinate synthase(GB_D07G0623),nuclear factor Y,subunit B13(GB_D07G0631),and leucine-rich repeat transmembrane protein kinase(GB_D07G0797).Our results provide novel insights into the mechanism underlying fiber formation and lay a solid foundation for further high-efficiency determination of candidate genes by combining RNA-seq data and pivotal chromosome regions.Pengtao Li Quanwei Lu Xianghui Xiao Rui Yang Xixi Duan 2021Phyton-International Journal of Experimental Botany2021,90,1:0
7Genome-wide identification,characterization,and expression analysis of aluminum-activated malate transporter genes(ALMTs)in Gossypium hirsutum L.显示文摘Aluminum-activated malate transporters(ALMT)are widely involved in plant growth and metabolic processes,including adaptation to acid soils,guard cell regulation,anion homeostasis,and seed development.Although ALMT genes have been identified in Arabidopsis,wheat,barley,and Lotus japonicus,little is known about its presence in Gossypium hirsutum L.In this study,ALMT gene recognition in diploid and tetraploid cotton were done using bioinformatics analysis that examined correlation between homology and evolution.Differentially regulated ALMT genetic profile in G.hirsutum was examined,using RNA sequencing and qRT-PCR,during six fiber developmental time-points,namely 5 d,7 d,10 d,15 d,20 d,and 25 d.We detected 36 ALMT genes in G.hirsutum,which were subsequently annotated and divided into seven sub-categories.Among these ALMT genes,34 had uneven distribution across 14/26 chromosomes.Conserved domains and gene structure analysis indicated that ALMT genes were highly conserved and composed of exons and introns.The GhALMT gene expression profile at different DPA(days post anthesis)in different varieties of G.hirsutum is indicative of a crucial role of ALMT genes in fiber development in G.hirsutum.This study provides basis for advancements in the cloning and functional enhancements of ALMT genes in enhancing fiber development and augmenting high quality crop production.QUANWEI LU YUZHEN SHI RUILI CHEN XIANGHUI XIAO PENGTAO LI JUWU GONG RENHAI PENG YOULU YUAN 2022BIOCELL2022,46,5:0
