1Neomycin biosynthesis is regulated positively by AfsA-g and NeoR in Streptomyces fradiae CGMCC 4.7387显示文摘Neomycins are a group of aminoglycoside antibiotics with both clinical and agricultural applications.To elucidate the regulatory mechanism of neomycin biosynthesis,we completed draft genome sequencing of a neomycin producer Streptomyces fradiae CGMCC 4.7387 from marine sediments,and the neomycin biosynthesis gene cluster was identified.Inactivation of the afsA-g gene encoding a γ-butyrolactone(GBL) synthase in S.fradiae CGMCC 4.7387 resulted in a significant decrease of neomycin production.Quantitative RT-PCR analysis revealed that the transcriptional level of neoR and the aphA-neoGH operon were reduced in the afsA-g::aac(3)Ⅳ mutant.Interestingly,a conserved binding site of AdpA,a key activator in the GBL regulatory cascade,was discovered upstream of neoR,a putative regulatory gene encoding a protein with an ATPase domain and a tetratricopeptide repeat domain.When neoR was inactivated,the neomycin production was reduced about 40%in comparison with the WT strain.Quantitative RT-PCR analysis revealed that the transcriptional levels of genes in the aphA-neoGH operon were reduced clearly in the neoR::aac(3)Ⅳ mutant.Finally,the titers of neomycin were improved considerably by overexpression of qfsA-gand neoR in S.fradiae CGMCC 4.7387.Xiangxi Meng Wenzhao Wang Zhoujie Xie Pengwei Li Yue Li Zhengyan Guo Yingjian Lu Jie Yang Kaile Guan Zhaoxin Lu Huarong Tan Yihua Chen 2017Science China(Life Sciences)2017,60,9:7
2Comprehensive Treatment and Prognostic Factors in Patients with Carcinoma of the Cervical Stump显示文摘OBJECTIVE To evaluate the applicability of combined therapyand the prognostic factors in patients with carcinoma of thecervical stump (CCS).METHODS The clinical records of 60 CCS patients whounderwent combined treatment in our hospital during a periodfrom January 2000 to December 2007, were collected andretrospectively analyzed. The prognostic factors were studiedusing univariate analysis. Analytical evaluation of the independentprognostic factors was performed using COX proportionalhazardsregression model.RESULTS The 1-, 3- and 5-year survival rates of the 60 patientswere 95%, 78% and 68%, respectively, with a median survivaltime of 32 months. Univariate survival analysis showed thatthese independent prognostic factors included positive pelviclymph nodes (P = 0.001), lymphovascular tumor embolus (P =0.001), and adjuvant chemotherapy (P = 0.011). In the 60 cases,postoperative local recurrence in the pelvic cavity occurred in1 and distant metastasis in 3. Related complications, such asradiocystitis, recto-vaginal fistula and vesico-vaginal fistula werefound in 6 of the total cases (10%). The serum levels of squamousepithelium antigen detected before and after treatment weresignificantly different (P = 0.000). The incidence of CCS is low;however, the disease is difficult to cure due to the high incidenceof complications and to the frequency of distant metastasis.Therefore, individualized treatment is needed. Complications fromsubtotal hysterectomy (STH) should be treated and controlledaggressively. Careful follow-up as well as close monitoring andobservation for significant symptoms in the postoperative coursewill enhance clinical outcome.CONCLUSION Cancer of the cervical stump has a lowmorbidity and severe complications, and most recurrences aredistant metastases. Because it is difficult to cure, there is a needto design a treatment regimen for each individual patient basedon the factors deemed as high risk. 3Description of a New Species of Amolops (Anura:Ranidae) from Tibet,China显示文摘A new species,Amolops pallasitatus sp.nov.is described based on specimens collected from Chentang Town,Dinggyê County,southern Tibet,China.The new species can be distinguished from other known congeners by mitochondrial divergence and morphological characteristics including:(1)medium body size,SVL 70.6–72.3 mm in adult females;(2)skin smooth over the entire body;(3)absence of dorsolateral fold;(4)tympanum small,edge indistinct,less than half of eye diameter;(5)vomerine teeth in two short oblique;(6)circummarginal and transverse grooves absent on disk of the first finger;(7)presence of inner metacarpal tubercle;(8)toes fully webbed,webbing formula Ⅰ0-0^-Ⅱ0-1/2Ⅲ0-1^+Ⅳ1^+-0Ⅴ;(9)absence of outer metatarsal tubercle and tarsal glands;(10)tibiotarsal articulation of the hind limb reaches posterior corners of the eye;(11)dorsum yellow-green,with irregular dark brown blotches without margins;(12)blotches concentrated on the dorsum,less on the flanks.In morphology,Amolops pallasitatus sp.nov.is similar to A.himalayanus and A.formosus,the difference between them is length of hind limbs,web of toe and dorsal colour pattern.The systematic placement of the new species within the genus is unresolved and it is not assigned to any recognized species group,for the lack of convictive evidences.Shuo QI Zhengyan ZHOU Zhitong LYU Yuyan LU Han WAN Mian HOU Keji GUO Pipeng LI 2019Asian Herpetological Research2019,10,4:1
4NMDAR-dependent somatic potentiation of synaptic inputs is correlated withβamyloid-mediated neuronal hyperactivity显示文摘Background:βAmyloid(Aβ)-mediated neuronal hyperactivity,a key feature of the early stage of Alzheimer’s disease(AD),is recently proposed to be initiated by the suppression of glutamate reuptake.Nevertheless,the underlying mechanism by which the impaired glutamate reuptake causes neuronal hyperactivity remains unclear.Chronic suppression of the glutamate reuptake causes accumulation of ambient glutamate that could difuse from synaptic sites at the dendrites to the soma to elevate the tonic activation of somatic N-methyl-D-aspartate receptors(NMDARs).However,less attention has been paid to the potential role of tonic activity change in extrasynaptic glutamate receptors(GluRs)located at the neuronal soma on generation of neuronal hyperactivity.Methods:Whole-cell patch-clamp recordings were performed on CA1 pyramidal neurons in acute hippocampal slices exposed to TFB-threo-β-benzyloxyaspartic acid(TBOA)or human Aβ_(1-42) peptide oligomer.A series of dendritic patch-clamp recordings were made at diferent distances from the soma to identify the location of the changes in synaptic inputs.Moreover,single-channel recording in the cell-attached mode was performed to investigate the activity changes of single NMDARs at the soma.Results:Blocking glutamate uptake with either TBOA or the human Aβ_(1-42) peptide oligomer elicited potentiation of synaptic inputs in CA1 hippocampal neurons.Strikingly,this potentiation specifcally occurred at the soma,depending on the activation of somatic GluN2B-containing NMDARs(GluN2B-NMDARs)and accompanied by a substantial and persistent increment in the open probability of somatic NMDARs.Blocking the activity of GluN2B-NMDARs at the soma completely reversed both the TBOA-induced or the Aβ_(1-42)-induced somatic potentiation and neuronal hyperactivity.Conclusions:The somatic potentiation of synaptic inputs may represent a novel amplifcation mechanism that elevates cell excitability and thus contributes to neuronal hyperactivity initiated by impaired glutamate reuptake in AD.Yifei Bao Xin Yang Yi Fu Zhengyan Li Ru Gong Wei Lu 2021Translational Neurodegeneration2021,10,3:0
5Prevalence of Wolbachia in 10 Tenebrionidae stored-product insects and spatiotemporal infection dynamics in Tribolium confusum(Jaquelin Du Val)(Coleoptera:Tenebrionidae)显示文摘Insect symbionts Wolbachia used for pest population control has focused on vector pest species and agricultural insects while rare reports in Chinese stored-product insect samples.In this paper,we surveyed the prevalence of Wolbachia using a PCR detection method in ten Tenebrionidae stored-product insects.Subsequently,the spatiotemporal Wolbachia infection dynamics in Tribolium confusum and Wolbachia elimination patterns using tetracycline treatment were investigated in detail by TaqMan®probe real-time quantitative PCR,and host reproductive fitness parameters were compared.T.confusum was the only Wolbachia infected species in all the surveyed species.Wolbachia infection density consistently increased with the development of T.confusumand plateaued at 3.7×107 wsp copies per individual insect at the young adult stage.Wolbachia densities in females were higher than that in males with a significant difference at the pupae stage and varied among different tissues and organs.Uninfected female beetles were completely incapable of producing mature progenies when crossed with Wolbachia infected males.Embryogenesis and egg hatch rate were specifically inhibited after Wolbachia elimination,while other traits,including the number of eggs,pupation rate and sex ratio,remained unaffected by tetracycline treatment.Our results showthat the TaqMan®probe qPCR is a reliable detection method for quantifying the density of Wolbachia as compared to qualitative detection of wsp gene by PCR and relatively quantified by real-time qPCR.The fitness results indicated that Wolbachia infection was not an obligate symbiont and benefited the host confused flour beetle.Yujie Lu Shiyuan Miao Zhengyan Wang Sibao Wang 2019Grain & Oil Science and Technology2019,2,4:0
6Dual-Band Flexible MIMO Antenna with Self-Isolation Enhancement Structure for Wearable Applications显示文摘A two-element dual-band flexible multi-in multi-out antenna which can be used for wearable applications is proposed in this paper.The antenna consists of two radiating elements fed by coplanar waveguide,and a shielding layer,which are all made of flexible conductive cloth MKKTN260.Each radiating element is composed of two coupled split ring-shaped bending strips.The proposed antenna shows two measured impedance bandwidth(S_(11)<−10 dB)of 2.39-2.48 GHz and 5.72-5.88 GHz,so that it can be used for 2.4 GHz and 5.8 GHz ISM(industrial scientific medical)applications.The two coupled split rings form a self-isolation enhancement structure and can realize polarization diversity at 2.4 GHz band and radiation shielding at 5.8 GHz band,respectively.High isolation(>30 dB)has been achieved for both the bands.Other characteristics for wearable applications like gain,efficiency,specific absorption rate,and bending performances were also studied.YANG Lingsheng XIE Yizhang JIA Hongting QU Meixuan LU Zhengyan LI Yajie 2023Chinese Journal of Electronics2023,32,4:0
7Morphology of Kaloula rugifera and K. borealis Tadpoles(Anura: Microhylidae: Kaloula verrucosa Group) in China显示文摘The external morphology and internal oral features of the tadpoles of Kaloula rugifera and K. borealis are described in this paper. Similarities between these two larvae include external morphology, oral disc, internal oral features(floor and roof), terminal mouth, lateral eyes, single midventral spiracle, absence of keratinized jaw sheaths and labial teeth, absence of lingual papillae and postnarial arena, while the body of K. rugifera is bigger than that of K. borealis at stages from 36 to 39 and the number of papillae in the buccal cavity is 12–19 and 9–16, respectively in the two species.
