| 1 | Search for naive human pluripotent stem cells显示文摘Normal mouse pluripotent stem cells were originally derived from the inner cell mass(ICM) of blastocysts and shown to be the in vitro equivalent of those pre-implantation embryonic cells, and thus were called embryonic stem cells(ESCs). More than a decade later, pluripotent cells were isolated from the ICM of human blastocysts. Despite being called human ESCs, these cells differ significantly from mouse ESCs, including different morphology and mechanisms of control of pluripotency, suggesting distinct embryonic origins of ESCs from the two species. Subsequently, mouse pluripotent stem cells were established from the ICMderived epiblast of post-implantation embryos. These mouse epiblast stem cells(Epi SCs) are morphological and epigenetically more similar to human ESCs. This raised the question of whether cells from the human ICM are in a more advanced differentiation stage than their murine counterpart, or whether the available culture conditions were not adequate to maintain those human cells in their in vivo state, leading to a transition into Epi SC-like cells in vitro. More recently, novel culture conditions allowed the conversion of human ESCs into mouse ESC-like cells called nave(or ground state) human ESCs, and the derivation of nave human ESCs from blastocysts. Pers.is a pioneer species native to South America able to release allelochemicals that affect germination and development of other plant species.The aim of this work was to evaluate the allelopathic effect of S.virgata on the germination and development of co-occurring species from gallery forest and on agronomic species.Two forest native species,Enterolobium contortisiliquum(Vell.) Morong and Sapindus saponaria L.,and two agronomic species(these as control),Oryza sativa L.and Solanum lycopersicum L.were used in the lab and greenhouse assays with seeds and leaf extracts of S.virgata.Agronomic species were more affected than native species when co-germinated with seeds of S.virgata.The germination percentage and speed germination index of the agronomic species were reduced in both in vitro and greenhouse assays.In the same assays,the seeds of native species showed no significant differences in the parameters mentioned.However,the initial growth of the four species assayed was affected,with reduction in plant length and shoot diameter followed by significant reduction in plantlet shoot and root weights.In the assays with irrigation of S.virgata leaf extracts,no significant inhibitory effects on germination were observed for all species.Height and shoot diameter of the native species were not affected by the leaf extracts,nor were fresh and dry weights.However,these parameters increased in the agronomic species as they were irrigated with leaf extracts.Based on our data we conclude that seed leachates of S.virgata affect germination and seedling development of other species,suggesting that its invasive behavior is due mainly to seed allelochemicals.Although with less pronounced effects on native species,allelopathy of S.virgata might also influence Interspecific competition in its natural environment. | Vera Lygia El Id Bárbara Vale da Costa Daiane Salete Broch Mignoni Marina Belloni Veronesi Kelly Simoes Marcia Regina Braga Nelson Augusto dos Santos Junior | 2015 | Journal of Forestry Research2015,26,2: | 0 |
| 18 | Hyponatremia is associated with more severe biliary disease显示文摘BACKGROUND Sodium level is an important clinical predictor of complex biliary disease.Hyponatremia has been observed in conjunction with biliary disease,however the nature of this association remains unclear.AIM To investigate the association between serum sodium and severe biliary disease.METHODS Of 920 patients with gallstone disease treated at the SFVA Hospital from 1989-2019 were studied.We conducted multivariate analyses of correlation between sodium level and biliary disease severity,the presence/location of biliary bacteria,and other factors.Minimum sodium level pre-intervention was collected.Gallstones,bile,and blood(as relevant)were cultured.Illness severity was characterized:(1)None(no infectious manifestations);(2)Systemic inflammatory response syndrome;(3)Severe illness(gangrenous cholecystitis,cholangitis,necrotizing pancreatitis);and(4)Multiple organ dysfunction syndrome(bacteremia,hypotension,organ failure).Comorbidity was defined using the Charlson Comorbidity Index(CCI).RESULTS Decreased sodium level significantly correlated with worsening illness severity,ascending bacterial infection,gangrenous changes,elevated CCI score,increasing age,male sex,and glucose.On multivariate analysis,all factors,except age,gender and glucose,independently correlated with sodium level and factors were additive.CONCLUSION This unique study is the first to explore,with such granularity,the relationship between biliary disease and sodium.No prior studies have examined specific culture and clinical data.It illustrates an inverse,independent correlation between illness severity and sodium.Culture data demonstrate that sodium decreases as infection ascends from gallstone colonization to bactibilia to bacteremia.Patient comorbidity and gangrenous changes also independently correlate with sodium on multivariate analysis.Sodium level is an important clinical indicator of disease severity for patients with biliary disease. | Michael John Zobel Lygia Stewart | 2020 | World Journal of Gastrointestinal Surgery2020,12,2: | 0 |
