| 1 | Identification of maize seed varieties based on near infrared reflectance spectroscopy and chemometrics显示文摘False seeds can often be seen in the maize seed market,leading to a serious decline in maize yield.Those existing variety identification methods are expensive,time consuming,and destructive to seeds.The aim of this study is to develop a cheap,fast and non-destructive method which can robustly identify large amounts of maize seed varieties based on near-infrared reflectance spectroscopy(NIRS)and chemometrics.Because it is difficult to establish models for every variety in the market,this study mainly investigated the performance of models based on a large number of samples(more than 40 major varieties in the market).The reflectance spectra of maize seeds were collected by two modes(bulk kernels mode and single kernel mode).Both collection modes can be applied to identification,but only the single kernel mode can be applied to purity sorting.The spectra were pretreated with smoothing,the first derivative and vector normalization;and then principal component analysis(PCA),linear discriminant analysis(LDA)and biomimetic pattern recognition(BPR)were applied to establish identification models.The environmental factors such as producing areas and years have a significant influence on the performance of the models.Therefore,the method to improve the robustness of the models was investigated in this study.New indexes(correct acceptance degree(CAD),correct rejection degree(CRD)and correct degree(CD))were defined to analyze the performance of the models more accurately.Finally,the models obtained a mean correct discrimination rate of over 90%,and exhibited robust properties for samples harvested from different areas and years.The results showed that NIR technology combined with chemometrics methods such as PCA,LDA,and BPR could be a suitable and alternative technique to identify the authenticity of maize seed varieties. | Yongjin Cui Lanjun Xu Dong An Zhe Liu Jiancheng Gu Shaoming Li Xiaodong Zhang Dehai Zhu | 2018 | International Journal of Agricultural and Biological Engineering2018,11,2: | 3 |
| 2 | The role of human ADA2a in the regulation of p53 acetylation and stability显示文摘The tumor suppressor protein p53 is a well-known transcription factor that functions as a critical component of the genotoxic stress response via regulating the expression of effector proteins that control cellular fate following DNA damage. The human p300/CBP-associated factor(PCAF)-containing histone acetyltransferase(HAT) complex is important for the stability and activity of p53 via its acetylation. The human homolog of yeast alteration/deficiency in activation 2a(ADA2a) is a stable component of the human PCAF-containing HAT complex. In this study,we demonstrated that p53 and hADA2a physically interact with each other in human HEK 293T cells. Using overexpression and small interfering RNA-mediated knockdown,we demonstrated that hADA2a stabilizes p53 via promoting its acetylation at lysine 320 - a PCAF-dependent acetylation site. Furthermore,hADA2a can potentiate the transcriptional activity of p53 at the BAX and p21 promoters to induce cell apoptosis and cell cycle arrest. | 3 | Experimental replacement of thoracic esophageal segment with a biomaterial artificial esophagus in dogs显示文摘Objective:To implant 80 mm-long artificial esophagi constructed of biomateriai in dogs,observe the perioperative survival rates and the incidence of postoperative complications,and study the mechanisms of postoperative healing.Methods:Specimens of the implanted esophagus,the'neo-esophagi',were taken for histopathologic study 1,3,6,12,24 months after operation.Results:The incidence of anastomotic leakage after the artificial esophagus implantation was 3.33%.The perioperative survival rate was 96.67%.The incidence of postoperative stenosis in the'neo-esophagi'was 81.48%;the stenoses were treated by expanding with esophagoscopy and implanting a stent.Epithelization of the mucosa in the'neo-esophagi'was completed in 3 to 6 months after surgery Structures such as submucosal muscle layers,mucous glands,nerve fibers,capillaries,etc.were regenerated after 12 months,and then reconstruction of the fibrous connective tissue layer was completed.Conclusion:Implanting a biomaterial artificial esophagus accomplishes safe reconstruction of defects in the esophagus.Advanced cellular structure of'neo-esophagus'can be regenerated after 1 year.Postoperative stenosis,which is related to hyperplasia and retraction of scar tissue,is still the most common complications which limiting the clinical application of the artificial esophagus. | Zhang Lanjun Rong Tiehua Su Xiaodong Wu Qiuliang Xu Guoliang Lin Peng Long Hao FU Jianhua | 2008 | Journal of Medical Colleges of PLA(China)2008,23,1: | 1 |

| 6 | A Study of Molecular Resection Margins for Esophageal Squamous Cell Carcinoma Using Large Pathologic Sections显示文摘OBJECTIVE It has been shown that application of molecular biological techniques to surgical margins of some cancers could predict risk of local recurrence. However, the optimal length of surgical resection with tumor-free surgical margins for esophageal squamous cell carcinoma (ES-CC) is unknown. This study was conducted to evaluate the optimal length of surgical resection for ESCC with molecularly tumor-free surgical margins marked by p53 and Ki67. METHODS Surgical specimens from 70 patients with ESCC were collected for study. The lengths of the upper margin, tumor, and lower margin of every specimen were measured during the operation. Each specimen was divided into three large pathologic sections, stained with H&E and immunohistochemically for p53 and Ki67, and examined microscopically. The lengths of the upper and lower resection ends were measured for p53 and Ki67 positive expression. The actual surgical lengths were calculated by the principle of rational shrinkage. RESULTS All surgical margins were histologically tumor-free, while the positive rates of p53 and Ki67 were 66% and 54%. The positive rates of p53 and Ki67 in the upper resection end were 17% and 20%. The mean lengths of the upper resection end showing p53 and Ki67 positive expression were 1.08±1.12 cm and 1.64±1.01 cm, and the maximum lengths were 3.73 cm and 3.26 cm. The positive rates of p53 and Ki67 in the lower resection end were 20% and 23%. The mean lengths of the lower resection end of p53 and Ki67 with positive expression were 1.11±1.15 cm and 1.34±0.94 cm, and the maximum lengths were 3.73 cm and 3.61 cm.
