| 1 | Stretching and imaging studies of single DNA molecules显示文摘DNA molecules were stretched on silanized mica surface with the molecular combing technique, and detected with fluorescence microscopy and atomic force microscopy. Meantime, DNA molecules were stretched with a modified dynamic molecular combing technique and studied with atomic force microscopy. The results indicate that, compared with the dynamic molecular combing technique, the modified dynamic molecular combing technique has advantages of less-sample demand and less contamination to sample; as compared with the molecular combing technique, it has better aligning effect and reproducibility. | 2 | Imaging bacteriorhodopsin-like molecules of claret-membranes from Tibet halobacteria xz515 by atomic force microscope显示文摘Halobacteria H.sp.xz 515 was isolated from a salt lake in Tibet. Although proton release-and-uptake across claret membrane is in reverse order compared to bacteri-orhodopsin in purple membrane from Halobacterium Sali-narum, and its efficiency of proton pump is much lower, AFM image shows that the molecules are still arranged in a two-dimensional hexagonal lattice of trimers. Primary structure of C- to G-helix of the archaerhodopsin shows that it has only 56% homology with bacteriorhodopsin. But the interactive amino acid residues at the interface between B-and D-helixes are conserved. | 3 | Microplastic pollution and enrichment of distinct microbiota in sediment of mangrove in Zhujiang River estuary,China显示文摘The microbial communities colonized on microplastics(MPs)have attracted widespread attention.However,few studies focused on the MPs impacts on mangrove ecosystems,particularly on bacterial communities.We investigated the MPs pollution in mangrove of Zhujiang(Pearl)River estuary(ZRE).To study the potential risk posed by MPs to the mangrove ecosystems,the differences in bacterial communities,functions,and complexity between MPs and sediment samples were reported for the first time.Microplastics(2991±1586 items/kg dry weight(dw))in sediment were mainly fibers and polyethylene,mostly transparent,and in size less than 0.5 mm.Bacterial communities and functions significantly differed from MPs in mangrove sediment.Compared with sediment,MPs significantly enriched members of Proteobacteria,Bacteroidetes,and Actinobacteria,as well as the bacteria associated with plastic-degrading and human diseases on their surface,suggesting that microbial communities on MPs may promote MPs degradation and the spread of diseases,posing potential risk to mangrove ecosystems and human health.Although bacteria on MPs exhibited a lower diversity,the co-occurrence network analysis indicated that network of bacteria colonized on MPs was bigger and more complex than those of mangrove sediment,illustrating that MPs can act as a distinct habitat in this special ecosystem.This study provides a new perspective for increasing our understanding of microplastic pollution in mangrove ecosystems. | Muting YAN Xiaofeng CHEN Wei CHU Weixin LI Minqian LI Zeming CAI Han GONG | 2023 | Journal of Oceanology and Limnology2023,41,1: | 1 |

| 5 | Real time observation of the photocleavage of single DNA molecules显示文摘A method for real time observation of photo-cleavage of stretched λDNA at single molecular level by a fluorescent microscope coupled with CCD is developed. DNA molecules stained with YOYO-1 are stretched by the mo-lecular combing technique and fixed on a modified slide. Then the process of photocleavage and relaxation of DNA under radiation of blue light is observed. We speculate that the conformation change of stretched DNA and the effect of water are likely to facilitate the effect of YOYO photocleav-age DNA molecules. | 7 | Investigation of chromatin folding patterns by atomic force microscopy显示文摘The chromatin folding patterns in air and liquid were studied by atomicforce microscopy (AFM). A gentle water-air interface method was adopted to spreadchromatin from interphase nucleus of chicken erythrocyte. The chromatin was ah-sorbed on APS-Inica surfaCe and studied with AFM. Beads-on-a-string were observedand many higher-order structures such as superbeads with dimensions 40-60urn indiameter and 4-7 urn in height were found to string together to make chromatin fibers.When sample spreading and absorbing time were shortened, higher-order chromatinfibers with 60-120 urn in width were observed in air as well as under water environment.
